Despite numerous studies, little is known about the influential factors impacting treponemal disease susceptibility. Yet, forms of treponemal disease, specifically yaws, are a major source of morbidity in the developing world. Bioarchaeological materials may assist with deficit by testing full ranges of disease expression. This study explores the relationship between treponemal disease ecology, specifically yaws, and susceptibility through six southeastern United States archaeological samples. Results show early life health experiences have a relationship to later life disease expression. However, influences of climate may only be seen at a cross-regional level. Pin1 is a Prolyl Isomerase that catalyzes cis-trans isomerization of peptides with pSer/Thr-Pro motifs in many cell signaling proteins. This conformational switch is implicated in diseases. Pin1 activity is considered a target for therapeutic applications. Pin1 targets motifs by its N-terminal WW-binding domain. A C-terminal PPIase domain is responsible for catalysis. To understand how Pin1 coordinates its enzymatic activities, it is necessary to probe how the domains behave in the presence of substrates. Urban Flow-Through Facilities' Media Compositions For Stormwater Quality And Quantity Improvements, Emily Gwynne Overbey

Urban stormwater management is evolving toward sustainable approaches which rely on dispersed small-scale bioretention BMPs. One such BMP is the flow-through planter, commonly applied in areas where infiltration into in situ soil is restricted or not possible. A project was developed to evaluate 18, vertically scaled flow-through mesocosms. Three replicates of six treatments, including four soil mixtures containing varied percentages of sand, compost and topsoil, were tested for orthophosphate and nitrate removal, volume reduction capabilities, and peak flow attenuation through the application of a synthetic solution over a simulated 2-inch, Type II storm event. Runoff volume was significantly (p < 0.05) reduced compared to controls. Nutrient levels observed along the hydrograph at different time-steps and flow rates revealed patterns not apparent in cumulative results. The observation of preferential flow patterns along with variability in nutrient removal across treatments highlights the need for design modifications of flow-through facilities.

The USDA Natural Resources Conservation Service (NRCS) implemented the Migratory Bird Habitat Initiative (MBHI) in summer 2010 to provide wetlands for waterbirds inland from the Deepwater Horizon Oil Spill. To evaluate MBHI and associated wetland management practices, I estimated seed and waterbird densities in MBHI and non-managed wetlands in the Mississippi Alluvial Valley (MAV) of Mississippi and Louisiana. Although not statistically different, wetlands enrolled in MBHI contained 1.26 and 1.53 times more seed biomass and seeds consumed by waterfowl than non-managed wetlands, respectively. The dissertation describes research work on development of bimetallic heterogeneous catalysts for oxidation of organic compounds. Binuclear metal complexes are an interesting class of compounds due to their catalytic activity. The approach involves anchoring a triketone ligand to Cab-O-Sil via a linker. Specifically, silica gel was allowed to react with p-chloromethylphenyltrimethoxysilane followed by coupling with deprotonated triketone compounds. The viability of this approach was verified by performing the benzylation reaction of triketones with benzyl halides under basic conditions. The benzylation of 2,4,6-heptanetrione and 1,5-diphenyl-1,3,5-pentanetrione is achieved with benzyl bromide using n-tetrabutylammonium fluoride as base. Stink bugs (Hemiptera: Pentatomidae) are key pests of soybean, Glycine max (L.), in Mississippi. Historically, yield loss derived from direct feeding by stink bugs has been considered the greatest threat to producers. However, quality reductions resulting from seed infections caused by microorganisms including Phomopsis longicolla are also a concern. Experiments were conducted in 2010 and 2011 to determine if stink bugs are associated with the incidence of P. longicolla in Mississippi soybean production. Data from experiments suggest that stink bugs are capable of transporting P. longicolla between two points. Data further suggest stink bugs and P. longicolla have the potential …

Using the concept of bone functional adaptation, this study analyzes femoral and humeral cross-sectional properties of human skeletal materials from Morton Shell Mound on the Louisiana coast. This work helps fill a gap in such analyses in the southern U.S. and contributes to an understanding of the functional adaptation of the human skeleton. Properties were compared to those of other prehistoric Southeastern fisher-hunter-gatherers from Gold Mine, Plash Island, and several Georgia coast sites to assess mobility and activity patterns among inland and coastal groups. The porcine antral follicle, which consists of an oocyte and surrounding follicular components, including theca, granulosa, and cumulus cells and follicular fluid, is an essential microenvironment for oocyte development and maturation. Investigating cellular and molecular events in the context of the whole follicle will aid in our understanding of interactions between the oocyte and the follicular components. The onarm water storage (OFWS) systems at Metcalf and Pitts farm in Porter Bayou Watershed were monitored for changes in nutrient levels and water withdrawal for irrigation from March 2012 to April 2013. Nitrogen and phosphorus levels were generally higher during the early part of the growing season. The OFWS systems can reduce nitrate and phosphorus in runoff up to 81% and 85%, respectively. However, the systems did not consistently reduce sediment and nutrients especially after significant rainfall and runoff events. In this dissertation, I develop a non-representational approach to metaethics that avoids the ontological and epistemic problems that traditional realist views face: accounting for the nature of moral facts and our knowledge of these facts in a way that comports with naturalism. According to expressivism—the most popular non-representational account in contemporary metaethics—moral claims don’t aim to represent moral facts or properties, but instead function to express our non-cognitive attitudes like approval or disapproval. This is a promising approach, but the view has substantial difficulties accounting for important features of moral discourse, like objectivity and complicated moral reasoning. I develop an …

Satellite imagery has been widely used to study internal solitary wave (ISW) generation mechanisms, evolution, and properties. The Nonlinear Internal Wave Initiative/Shallow Water 2006 (SW06) experiment in the Mid-Atlantic Bight (MAB) provided a unique dataset to study the vertical structure from their surface manifestation in the images, which can improve our understanding of ISW evolution, energy dissipation, and mixing processes. This thesis has two parts. Synapses are specialized cell-cell contacts where signals are reliably transduced from a neuron to its target cell in a regulated manner. Structurally, synapses are comprised of the pre- and post-synaptic terminals, and the synaptic cleft. Within each of these compartments is a complex network of molecular machinery, the components of which require constant turnover and “maintenance” to ensure a consistent relay of information. The residential community of the Islands of Cocoplum is located in Coral Gables, Florida along Biscayne Bay. It has served as a case study for mangrove mitigation techniques employed by Miami-Dade County's Regulatory and Economic Resources (RER) Coastal Resources division as a way of combating mangrove destruction. A database was created using Microsoft Excel and ArcGIS to identify and map all Class I coastal construction permits and mangrove violations since the onset of the Cocoplum Development in the 1980's through 2011. Surgical treatment for severe functional MR often involves mitral annuloplasty to improve leaflet apposition and ultimately downsize the dilated mitral valve. However, the high rate of operative mortalities of up to 6 ~12% have limited the more expanded use of this procedure. Recently, minimally invasive percutaneous transvenous mitral annuloplasty (PTMA) approaches using entirely catheter-based methods have been developed to reduce procedural morbidity and mortality. Insights Into Distinct Modulation Of α7 And α7β2 Nicotinic Acetylcholine Receptors By The Volatile Anesthetic, David D. Mowrey, Qiang Liu, Vasyl Bondarenko, Qiang Chen, Edom Seyoum, Yan Xu, Jie Wu, Pei Tang

Nicotinic acetylcholine receptors (nAChRs) are targets of general anesthetics, but functional sensitivity to anesthetic inhibition varies dramatically among different subtypes of nAChRs. Potential causes underlying different functional responses to anesthetics remain elusive. Here we show that in contrast to the α7 nAChR, the α7β2 nAChR is highly susceptible to inhibition by the volatile anesthetic isoflurane in electrophysiology measurements. Isoflurane-binding sites in β2 and α7 were found at the extracellular and intracellular end of their respective transmembrane domains using NMR. Functional relevance of the identified β2 site was validated via point mutations and subsequent functional measurements. Consistent with their functional responses …

Replication Protein A (RPA) is a heterotrimeric protein complex that binds single-stranded DNA. In plants, multiple genes encode the three RPA subunits (RPA1, RPA2 and RPA3), including five RPA1-like genes in Arabidopsis. Phylogenetic analysis suggests two distinct groups composed of RPA1A, RPA1C, RPA1E (ACE group) and RPA1B, RPA1D (BD group). ACE-group members are transcriptionally induced by ionizing radiation, while BD-group members show higher basal transcription and are not induced by ionizing radiation. Analysis of rpa1 T-DNA insertion mutants demonstrates that although each mutant line is likely null, all mutant lines are viable and display normal vegetative growth. The rpa1c and …

The GPA measure, we believe, is a better representation of student achievement on statewide standardized exams. In this report, we are presenting a list of the top 25 schools in each area. In some cases, these "top 25 lists" will contain more than 25 schools as some schools' GPA scores will be identical. This is not a new phenomenon, as we also exceeded 25 schools in previous reports when using the percent proficient and advanced metric as an indicator for student achievement; however, there are fewer ties using the more precise GPA measure.

Since its discovery and purification in 1971, DNA polymerase ß (Pol ߆) is one of the most well-studied DNA polymerases. Pol ß is a key enzyme in the base excision repair (BER) pathway that functions in gap filling DNA synthesis subsequent to the excision of damaged DNA bases. A major focus of our studies is on the cellular roles of Pol ß. We have shown that germline and tumor-associated variants of Pol ß catalyze aberrant BER that leads to genomic instability and cellular transformation. Our studies suggest that Pol ß is critical for the maintenance of genomic stability and that …

Elongation factor P (EF-P) is a conserved ribosome-binding protein that structurally mimics tRNA to enable the synthesis of peptides containing motifs that otherwise would induce translational stalling, including polyproline. In many bacteria, EF-P function requires post-translational modification with (R)-β-lysine by the lysyl-tRNA synthetase paralog PoxA. To investigate how recognition of EF-P by PoxA evolved from tRNA recognition by aminoacyl-tRNA synthetases, we compared the roles of EF-P/PoxA polar contacts with analogous interactions in a closely related tRNA/synthetase complex. The study also investigated if there was significant evidence to support the idea of using dramatic works as an example to illustrate adult learning and development theories and principles as an educational tool. Ten plays were analyzed along with an accompanying rubric that the evaluator completed while reading the plays. Target 2 of the 2020 Global Strategy for Plant Conservation (GSPC) calls for a comprehensive list of the world's threatened plant species. The lack of such a list is one of the greatest impediments to protecting the full complement of the world's plant species, and work to achieve this has been slow. An efficient system for identifying those species that are at risk of extinction could help to achieve this goal in a time frame sensitive to today's conservation needs.
