The Huntington's disease gene (HTT) CAG repeat mutation undergoes somatic expansion that correlates with pathogenesis. Modifiers of somatic expansion may therefore provide routes for therapies targeting the underlying mutation, an approach that is likely applicable to other trinucleotide repeat diseases. Huntington's disease Hdh(Q111) mice exhibit higher levels of somatic HTT CAG expansion on a C57BL/6 genetic background (B6.Hdh(Q111) ) than on a 129 background (129.Hdh(Q111) ). Linkage mapping in (B6x129).Hdh(Q111) F2 intercross animals identified a single quantitative trait locus underlying the strain-specific difference in expansion in the striatum, implicating mismatch repair (MMR) gene Mlh1 as the most likely candidate modifier. …

Quantitative Relationship Of Soil Texture With The Observed Population Density Reduction Of Heterodera Glycines After Annual Corn Rotation In Nebraska, Oscar Perez-Hernandez, Loren J. Giesler
Department of Plant Pathology: Faculty Publications
Soil texture has been commonly associated with the population density of Heterodera glycines (soybean cyst nematode: SCN), but such an association has been mainly described in terms of textural classes. In this study, multivariate analysis and a generalized linear modeling approach were used to elucidate the quantitative relationship of soil texture with the observed SCN population density reduction after annual corn rotation in Nebraska. Forty-five commercial production fields were sampled in 2009, 2010, and 2011 and SCN population density (eggs/100 cm3 of soil) for each field was determined before (Pi) and after (Pf) annual corn rotation from ten 3 3 …

Preliminary Assessment Of Diabetic Youth’S Acceptance Of Cinnamon In Treating Diabetes: A Telephone Interview, Valerie Amend, Dawn Vincent, Valerie Amend, Kimberli Pike
Journal of Human Sciences and Extension
Medications in treatment and control of diabetes can be costly for pediatric patients and families. Therefore, individuals may seek complementary and alternative therapies, such as cinnamon, in addition to traditional treatments. The objective of this study was to determine acceptability of using cinnamon in treatment of diabetes in a pediatric population with diabetes. Seventy-six pediatric diabetes patients at a diabetes clinic participated in a one-time telephone interview using an 18-item, validated questionnaire. Descriptive statistics were utilized to determine cinnamon acceptability and preferences. The majority of the subjects were between 16-18 years (43%, n = 33), Caucasian (62%, n = 47), …

Conversations About The Weight Of America's Children: Barriers Which Prevent Healthcare Providers From Discussing Childhood Obesity, Catherine Blow, Alisa Allicock, Carolynn Desandre, Chandra Cooper-Samuels
Journal of Human Sciences and Extension
A systematic review of the literature was conducted to identify the barriers that prevent practitioners from identifying and counseling parents and caregivers of overweight or obese children. Once identified, barriers were organized into thematic categories (parental, provider, and professional barriers) and recommendations were generated to facilitate discussion about childhood obesity between professionals and parents. Childhood obesity is a significant public health problem. Healthcare providers must be able to effectively communicate with caregivers and put childhood obesity at the front of healthcare discussions. This article provides a synthesis of the relevant literature and makes recommendations for healthcare providers to overcome the …

Killerflip: A Novel Lytic Peptide Specifically Inducing Cancer Cell Death, B Pennarun, G. Gaidos, O Bucur, A Tinari
Dartmouth Scholarship
One of the objectives in the development of effective cancer therapy is induction of tumor-selective cell death. Toward this end, we have identified a small peptide that, when introduced into cells via a TAT cell-delivery system, shows a remarkably potent cytoxicity in a variety of cancer cell lines and inhibits tumor growth in vivo, whereas sparing normal cells and tissues. This fusion peptide was named killer FLIP as its sequence was derived from the C-terminal domain of c-FLIP, an anti-apoptotic protein. Using structure activity analysis, we determined the minimal bioactive core of killerFLIP, namely killerFLIP-E. Structural analysis of cells using …

Neratinib Overcomes Trastuzumab Resistance In Her2 Amplified Breast Cancer, A Canonici, M Gijsen, M Mullooly, R Bennett, Et Al
Peninsula Medical School
Trastuzumab has been shown to improve the survival outcomes of HER2 positive breast cancer patients. However, a significant proportion of HER2-positive patients are either inherently resistant or develop resistance to trastuzumab. We assessed the effects of neratinib, an irreversible panHER inhibitor, in a panel of 36 breast cancer cell lines. We further assessed its effects with or without trastuzumab in several sensitive and resistant breast cancer cells as well as a BT474 xenograft model. We confirmed that neratinib was significantly more active in HER2-amplified than HER2 non-amplified cell lines. Neratinib decreased the activation of the 4 HER receptors and inhibited …

Fine-Scale Distributions Of Plankton And Larval Fishes: Implications For Predator-Prey Interactions Near Coastal Oceanographic Features, Adam Tyler Greer
Fine-Scale Distributions Of Plankton And Larval Fishes: Implications For Predator-Prey Interactions Near Coastal Oceanographic Features, Adam Tyler Greer
Open Access Dissertations
Describing the distributions of organisms on scales relevant to individuals (1-100 m) is critical to understanding predator-prey interactions within the plankton. This has driven the development of plankton imaging technology with synoptic physical parameters (temperature, salinity, depth), which facilitates high-resolution taxonomic and spatial descriptions. We utilized a novel In Situ Ichthyoplankton Imaging System (ISIIS) that addressed some shortcomings of other imaging systems by allowing for the simultaneous sampling of both abundant (e.g., copepods, appendicularians, marine snow) and rare (e.g., fish larvae, medusae, ctenophores) members of the plankton community. The main objectives of this study were 1) to describe the physical …
Apoptotic Regulation Of B Cell Development And Autoimmunity, Jacqueline A. Wright
Apoptotic Regulation Of B Cell Development And Autoimmunity, Jacqueline A. Wright
Open Access Dissertations
Misguided immune reactions against self can cause autoimmunity. Although multiple factors are likely involved, a common feature is altered lymphocyte homeostasis due to dyregulated apoptosis. Here, we show that the integration of signals from the BCR and BAFF-R promotes survival of T2 cells that have passed the self-reactivity checkpoint. BCR engagement produces NF-κB2 protein, a substrate used by the BAFF-R for the activation of the alternative NF-κB pathway. In contrast, pre-checkpoint T1 cells fail to integrate BCR and BAFF-R signals and remain sensitive to BCR-mediated-apoptosis. Btk and c-Rel are critical in this signal integration and B-cell survival. Importantly, BAFF-R promotes …

Including Capabilities Of Local Actors In Regional Economic Development: Empirical Results Of Local Seaweed Industries In Sulawesi, Mark T. J. Vredegoor, Bartjan W. Pennink
The South East Asian Journal of Management
Stimson, et al. (2009) developed one of the most relevant and well known model for Regional Economic Development. This model covers the most important factors related to economic development question. However, this model excludes the social components of development. Local community should be included in terms of the development of a region. This paper introduced to the Stimson model “Skills” and “Knowledge” at the individual level for local actors indicating the capabilities at the individual level and introduced “Human Coordination” for the capabilities at the collective level. In our empirical research we looked at the Indonesian seaweed market with a …
