The Angoumois grain moth, Sitotroga cerealella (Olivier) (Lepidoptera: Gelechiidae), is a pest of stored corn, Zea mays L., and other grains throughout the world. Sitotroga cerealella are routinely exposed to temperatures below 20°C in regions of the U.S. where corn is grown, yet there are no data describing adult life history parameters below 20°C. We determined longevity, fecundity, and survivorship of eggs at a range of temperatures that represent environmental conditions to which S. cerealella are exposed in corn stored in the U.S. Longest male longevity was 31 d at 10°C, and shortest male longevity was 4 d at 35 …

Unmanned aerial vehicles have been developed and applied to support agricultural production management. Compared with piloted aircraft, an Unmanned Aerial Vehicle (UAV) can focus on small crop fields at lower flight altitudes than regular aircraft to perform site-specific farm management with higher precision. They can also “fill in the gap” in locations where fixed winged or rotary winged aircraft are not readily available. In agriculture, UAVs have primarily been developed and used for remote sensing and application of crop production and protection materials. The cross-reactivity of mycobacterial antigens in immune-based diagnostic assays has been a major concern and a criticism of the current tests that are used for the detection of paratuberculosis. In the present study, Mycobacterium avium subsp. paratu- berculosis recombinant proteins were evaluated for antigenic specificity compared to a whole-cell sonicate preparation (MPS). Measures of cell-mediated immunity to M. avium subsp. paratuberculosis antigens were compared in calves inoculated with live M. avium subsp. paratuberculosis, M. avium subsp. avium (M. avium), Mycobacterium kansasii, or Mycobacterium bovis. The infection biology of Mycobacterium avium subsp. paratuberculosis has recently crystallized, with added details surrounding intestinal invasion. The involvement of pathogen-derived effector proteins such as the major membrane protein, oxidoreductase, and fibronectin attachment proteins have been uncovered. Mutations constructed in this pathogen have also shed light on genes needed for invasion. The host cell types that are susceptible to invasion have been defined, along with their transcriptional response. Anaerobic lagoons are a standard practice for the treatment of swine wastewater. This practice relies heavily on microbiological processes to reduce concentrated organic material and nutrients. Despite this reliance on microbiological processes, research has only recently begun to identify and enumerate the myriad and complex interactions that occur in this microbial ecosystem. To further this line of study, we utilized a next-generation sequencing (NGS) technology to gain a deeper insight into the microbial communities along the water column of four anaerobic swine wastewater lagoons. In cereal seeds, mutations in one or more starch synthases lead to decreased amylopectin and increased amylose content. Here, the impact of starch synthase IIa (SSIIa or SGP-1) mutations upon durum starch was investigated. A screen of durum accessions identified two lines lacking SGP-A1, the A genome copy of SGP-1. The two lines were determined to carry a 29 bp deletion in the first exon of SSIIa. The SGP-A1 nulls were crossed with the durum variety ‘Mountrail’ and F5 derived SGP-A1 null progeny lines were treated with EMS. From each EMS population, one SGP-B1 null mutation …

Knowledge of the behavior and movement patterns of European starlings (Sturnus vulgaris L.) is important to wildlife managers that seek to resolve conflicts at livestock facilities. We captured and radio tagged 10 starlings at each of 5 dairies in northeastern Ohio. From September 19 to October 31, 2007, we obtained sufficient data from 40 birds to study their behavior and movements. The birds visited the dairies where they were initially captured (home sites) on 85% of the days, spending 58% of each day at the dairies. House flies, (Musca domestica L.) and stable flies (Stomoxys calcitrans L.) are common pests on equine facilities. Biological control of these flies with pupal parasitoids has become increasingly popular with horse owners but has not been evaluated on equine facilities. Little is known of the substrate preferences of filth fly parasitoids on equine facilities, but the success of release programs may be affected by microhabitat preferences. Spalangia cameroni Perkins was evaluated for location preferences for parasitization of house fly and stable fly puparia in six substrates commonly found on equine farms in Florida. Drylands typically have distinctive vegetative patterns that reflect the underlying physical landscape. We use a geopedological classification to organize the landscape into five categories from broad scale to fine scale: (1) physiographic divisions which describe regional topography, such as mountains and basin floors; (2) regolith type which identifies residuum versus transported sediments; (3) parent materials which describe mineralogy; (4) landforms which provide meso-scale descriptions of topography; and (5) soil texture, a fine-scale variable important for infiltration, erodibility, and available water holding capacity. Current wet chemical methods for biomass composition analysis using two-step sulfuric acid hydrolysis are time-consuming, labor-intensive, and unable to provide structural information about biomass. Infrared techniques provide fast, low-cost analysis, are non-destructive, and have shown promising results. Chemometric analysis has allowed researchers to perform qualitative and quantitative study of biomass with both near-infrared and mid-infrared spectroscopy. This review summarizes the progress and applications of infrared techniques in biomass study, and compares the infrared and the wet chemical methods for composition analysis. Seedlings of Theobroma cacao CCN 51 genotype were grown under greenhouse conditions and exposed to increasing concentrations of Cu (0.005, 1, 2, 4, 8, 16, and 32 mg Cu L−1) in nutrient solution. When doses were equal or higher than 8mg Cu L−1, after 24 h of treatment application, leaf gas exchange was highly affected and changes in chloroplasts thylakoids of leaf mesophyll cells and plasmolysis of cells from the root cortical region were observed. Background: Identification of single nucleotide polymorphisms (SNPs) for specific genes involved in reproduction might improve reliability of genomic estimates for these low-heritability traits. Semen from 550 Holstein bulls of high (≥ 1.7; n = 288) or low (≤ −2; n = 262) daughter pregnancy rate (DPR) was genotyped for 434 candidate SNPs using the Sequenom MassARRAY® system. Three types of SNPs were evaluated: SNPs previously reported to be associated with reproductive traits or physically close to genetic markers for reproduction, SNPs in genes that are well known to be involved in reproductive processes, and SNPs in genes that …

1. Understanding predator–prey interactions of the arthropod community in any given ecosystem is essential in pinpointing the biological control services provided by natural enemies.
2. Hence, four prey-specific polymerase chain reaction (PCR) assays were developed to analyse the gut contents of the cotton predator community. The four targeted prey included a herbivore/pest, omnivore/pest, omnivore/beneficial, and carnivore/beneficial.
3. First, prey retention tests were conducted to determine how long a prey item of each target species could be detected in a predator after ingestion. The assays yielded highly variable inter-assay and intra-assay prey detection efficiencies.

Bees are among the most important pollinators of flowering plants in most ecosystems. Recent concerns about population decline worldwide have highlighted the urgent societal need for species-level systematic work that facilitates assessments of the status of pollinators and pollination services. This is a comprehensive, broadly comparative study on the diversity, biology, biogeography, and evolution of Anthidium Fabricius, 1804, one of the most diverse megachilid genera, containing more than 160 species worldwide. Fine root dynamics control a dominant flux of carbon from plants and into soils and mediate potential uptake and cycling of nutrients and water in terrestrial ecosystems. Understanding of these patterns is needed to accurately describe critical processes like productivity and carbon storage from ecosystem to global scales. However, limited observations of root dynamics make it difficult to define and predict patterns of root dynamics across broad spatial scales. Pathogen infection can induce plant volatile organic compounds (VOCs). We infected ‘McNeal’ wheat and ‘Harrington’ barley with a Fusarium spp. blend (F. graminearum, F. avenaceum and F. culmorum). Both cereals had the greatest VOC induction 14 days after pathogen innoculation, only slightly lower induction occurred at 7 days, but displayed no induction at 1 days. The induced VOC bouquet for both cereals included six green leaf volatiles (GLVs; e.g. (Z)-3-hexenol and (Z)-3-hexenyl acetate), four terpenes (linalool, linalool oxide, (Z)-β-ocimene and (E)-β-caryophyllene) and benzyl acetate. Recent behavioral and electrocortical studies have found that shy and socially anxious adults are hypersensitive to the processing of negative and ambiguous facial emotions. We attempted to extend these findings by examining the neural correlates of affective face processing in shy adults using an event-related fMRI design. We presented pairs of faces that varied in affective valence and intensity. The faces were morphed to alter the degree of intensity of the emotional expressive faces. Twenty-four (12 shy and 12 non-shy) young adult participants then made same/different judgments to these faces while in an MR scanner. We found that shy adults …

High power thulium fiber lasers are useful for a number of applications in both continuous-wave and pulsed operating regimes. The use of thulium as a dopant has recently gained interest due to its large bandwidth, possibility of high efficiency, possibility of high power and long wavelength ~1.8 – 2.1 μm. The longer emission wavelength of Tm-doped fiber lasers compared to Yb- and/or Er-doped fiber lasers creates the possibility for higher peak power operation due to the larger nonlinear thresholds and reduced nonlinear phase accumulation. HCMV employs numerous strategies to combat, subvert, or co-opt host immunity. One evolutionary strategy for this involves "capture" of a host gene and then its successive duplication and divergence, forming a gene family, many of which have immunomodulatory activities. The HCMV US12 family consists of ten tandemly arranged sequence-related genes in the unique short region of the HCMV genome (US12-US21). Each gene encodes a protein possessing seven predicted transmembrane domains, and patches of sequence similarity with cellular GPCRs and the bax inhibitor-1 family of anti-apoptotic proteins. The life history of a species is highly impacted by their reproductive strategy. In my dissertation I address the changing reproductive strategies in callitrichine New World monkeys and their genetic underpinnings using a phylogenetic approach. The necessity for a resolved phylogeny is universal to any comparative genomic study. Here we have constructed a reliable phylogenetic framework from which reproductive strategy could be studied in callitrichines. The search for an answer to the question of the achievement gap between Black and other minority students and their White counterparts has been pervasive. This study, however, reframes the question not by using the White student as a frame of reference against which to make the comparison but by using the Black student himself. Therefore, relying on some scholarly work that found that studies of achievement within ethnic groups yielded far more significant results than between groups, the study focused on a comparison of the academic achievement between urban Black students with learning disabilities and their typical peers. The relationship of the extra-cellular matrix (ECM) and secreted proteins to intra-cellular functions has an important impact on disease progression. The tissue inhibitors of metalloproteinases (TIMPs), of which there are four, are known for their broad metalloprotease inhibitory abilities. TIMP-1 has been widely studied as an inhibitor of matrix metalloproteinases (MMPs) and therefore as a molecule that prevents ECM degradation and proteolytic cleavage of extra-cellular molecules, processes thought to be critical for tumor invasion and metastasis. In budding yeast, as in higher eukaryotes, transcription of protein coding genes is executed by a highly specialized, conserved polymerase called RNA polymerase II (RNAPII). The transcription cycle of RNAPII has four major steps: initiation, elongation, termination, and reinitiation. The successful accomplishment of each of these steps requires a number of accessory factors. Many of these factors operate at multiple steps in the transcription cycle. Pre-exposure of the cornea to TLR5 ligand flagellin induces profound mucosal innate protection against infections by reprogramming gene expression. This study explored the flagellin-induced modifications of transcription factor expression and function, specifically of IRF1 and ATF3 in corneal epithelial cells to elucidate the transcriptional mechanisms underlying the protective function of flagellin on the cornea.
Initially we used Superarray to screen for transcription factors and identified Interferon Regulatory Factor (IRF) 1 and Activating Transcription Factor (ATF) 3 as the most drastically affected genes by flagellin pretreatment in P. aeruginosa challenged human corneal epithelial cells (CEC).
