The Ca(2+) sensor S100A1 is essential for proper endothelial cell (EC) nitric oxide (NO) synthase (eNOS) activation. S100A1 levels are greatly reduced in primary human microvascular ECs subjected to hypoxia, rendering them dysfunctional. However mechanisms that regulate S100A1 levels in ECs are unknown. Here we show that ECs transfected with a S100A1-3' untranslated region (UTR) luciferase reporter construct display significantly reduced gene expression when subjected to low oxygen levels or chemical hypoxia. Bioinformatic analysis suggested that microRNA -138 (MiR-138) could target the 3'UTR of S100A1. It is well established that male rats have an advantage in acquiring place-learning strategies, allowing them to learn spatial tasks more readily than female rats. However many of these differences have been examined solely during acquisition or in 24h memory retention. Here, we investigated whether sex differences exist in remote long-term memory, lasting 30d after training, and whether there are differences in the expression pattern of molecular markers associated with long-term memory maintenance. Specifically, we analyzed the expression of protein kinase M zeta (PKMζ) and the α-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid (AMPA) receptor subunit GluA2. Zuzu's Petals relates the travails of Jules and Julie, a couple whose marriage is in the process of breaking apart. Jules has a "fanboyish" obsession with the 1946 Frank Capra film "It's a Wonderful Life." Furthermore, he is fixated on his own past, and his eccentric behavior has caused his disenchanted wife to seek romantic and emotional solace in David, an old flame from high school. Their child, Zuzu, who was named at Jules' insistence after a key character in the Capra film, has herself sensed that something is amiss in her parents' dealings with each other. Atmospheric CO2 concentration is hypothesized to influence vegetation distribution via tree-grass competition, with higher CO2 concentrations favouring trees. The stable carbon isotope (delta C-13) signature of vegetation is influenced by the relative importance of C-4 plants (including most tropical grasses) and C-3 plants (including nearly all trees), and the degree of stomatal closure - a response to aridity in C-3 plants. Compound-specific delta C-13 analyses of leaf-wax biomarkers in sediment cores of an offshore South Atlantic transect are used here as a record of vegetation changes in subequatorial Africa. In vertebrates, nerve muscle communication is mediated by the release of the neurotransmitter acetylcholine packed inside synaptic vesicles by a specific vesicular acetylcholine transporter (VAChT). Here we used a mouse model (VAChT KDHOM) with 70% reduction in the expression of VAChT to investigate the morphological and functional consequences of a decreased acetylcholine uptake and release in neuromuscular synapses. Upon hypertonic stimulation, VAChT KDHOM mice presented a reduction in the amplitude and frequency of miniature endplate potentials, FM 1-43 staining intensity, total number of synaptic vesicles and altered distribution of vesicles within the synaptic terminal. Background
De novo protein modeling approaches utilize 3-dimensional (3D) images derived from electron cryomicroscopy (CryoEM) experiments. The skeleton connecting two secondary structures such as α-helices represent the loop in the 3D image. The accuracy of the skeleton and of the detected secondary structures are critical in De novo modeling. It is important to measure the length along the skeleton accurately since the length can be used as a constraint in modeling the protein.
Results
We have developed a novel computational geometric approach to derive a simplified curve in order to estimate the loop length along the skeleton. The method …

Though still in relative infancy, the field of flapping flight has potential to have a far-reaching impact on human life. Nature presents a myriad of examples of successful uses of this locomotion. Human efforts in flapping flight have seen substantial improvement in recent times. Wing kinematics are a key aspect of this study. This study summarizes previous wing trajectory generators and presents a new trajectory generation method built upon previous methods. This includes a novel means of commanding unequal half-stroke durations subject to robotic trajectory continuity requirements. Additionally, previous optimization methods are improved upon. The Glass Polyalkenoate Cement (GPC) series (LCon., LGa-1 and LGa-2) containing gallium (Ga) and a 50 wt% addition of E11 polyacrylic acid (PAA), previously developed, was evaluated for ion release, specifically Si, Ca, Zn and Ga, and E. coli inhibition. The maximum inhibition was observed in the t = 0 samples and was 0.35 mm for LCon. and 0.65 mm for LGa-1 and LGa-2. © 2013 IEEE.

SidA (siderophore A) is a flavin-dependent N-hydroxylating monooxygenase that is essential for virulence in Aspergillus fumigatus. SidA catalyzes the NADPH- and oxygen-dependent formation of N5-hydroxyornithine. In this reaction, NADPH reduces the flavin, and the resulting NADP+ is the last product to be released. The presence of NADP+ is essential for activity, as it is required for stabilization of the C4a-hydroperoxyflavin, which is the hydroxylating species. As part of our efforts to determine the molecular details of the role of NADP(H) in catalysis, we targeted Ser-257 for site-directed mutagenesis and performed extensive characterization of the S257A enzyme. Alcoholism is a persistent substance abuse disorder that is associated with negative health, social, and economic outcomes. Treatment strategies for alcohol use disorders are limited, and only three drugs have been approved by the FDA for treatment. Although behavioral therapy and drug combination strategies improve abstinence outcomes, the majority of those in treatment will not achieve long-term abstinence. Therefore, better treatment strategies are needed. While much progress has been made toward understanding the neurobiology of alcoholism, this knowledge has not been effectively translated into treatment strategies. Animal models of alcohol drinking have been crucial to this research effort, but until …

This work aims to determine the effect of substituting TiO2 for SiO2 in a 0.62SiO2-Na2O-0.24CaO based glass-ceramic scaffold. High temperature X-ray Diffraction (HT-XRD) was used to determine the sintering temperature (700oC). Both optical microscopy and x-ray micotomography was used to determine the average pore size (540-680ìm) of each scaffold. Cytocompatibility of each scaffold was conducted using murine mesenchymal stem cells. © 2013 IEEE.

In the first part (lasting approx 45 minutes) Mr. Rhodes discusses growing up in the Bloomington-Normal area and so is able to offer stories about his experiences attending IWU events before and during his student days. Genes encoding two proteins corresponding to elongation factor G (EF-G) were cloned from Pseudomonas aeruginosa. The proteins encoded by these genes are both members of the EFG I subfamily. The gene encoding one of the forms of EF-G is located in the str operon and the resulting protein is referred to as EF-G1A while the gene encoding the other form of EF-G is located in another part of the genome and the resulting protein is referred to as EF-G1B. Engineered osmosis (EO) is a state-of-the-art technology which harnesses the natural phenomenon of osmosis to address global issues related to water and energy. In this process, an osmotic pressure drives water across a semi-permeable membrane from a dilute feed solution to a concentrated draw solution. EO has the potential to sustainably produce fresh water at low energy cost, generate electricity and recover high-value dissolved solids. However, EO has not progressed beyond conceptualization and lab scale studies due to obstacles in membrane design, draw solution recovery, system integration, scale-up, and definitive process economics. A field calibration study of low density polyethylene (LDPE) for measuring atmospheric concentrations of persistent organic pollutants (POPs) was performed in East Providence (RI), USA. LDPE samplers were collected after 3, 7, 10, 14, 17 and 21 days of exposure along with samples from a co-deployed high volume sampler. Uptake kinetics of POPs by LDPEs were confirmed both by using an uptake study over time and the inclusion of performance reference compounds (PRCs). Results indicated that only POPs with log sampler-air partitioning coefficient (KPE-A) ≤ 7.6 were approaching equilibrium by the end of the deployment period, whereas all the …
