Without northern doughface Democrats, and northern states like Indiana, the South could not have held dominance in American politics during the sectional crisis. Anchoring the extreme end of the doughface North was Indiana’s slaveholding senator Jesse Bright (his holdings were in Kentucky). Yet, he was no flailing radical pushed to the margins of northern politics. Bright was the chief party boss who by the mid to late 1850s controlled the state of Indiana. He was one of the most influential leaders getting James Buchanan into the presidency. He did this, in part, because Indiana was a conservative state that disliked …

Only recently have scholars given particular attention to the development of the racial discourse present in early Christian apologetics. This study is aimed at understanding the Latin and Greek literary antecedents to the development of a Christian discourse on race and identity and examining in detail the apex of this discourse in the work of third century apologist Origen of Alexandria. Origen’s work represented the apex of an evolving discourse that, while continuing to use traditional vocabulary, became increasingly universalizing with the growth of the Roman Empire. Behavioral traits can be remarkably flexible depending on the conditions in which they are expressed, yet, in spite of this flexibility, persistent differences between individuals appear to limit the potential expression of behaviors. For example, despite evidence that parents provide variable amounts of parental care in response to changing environmental conditions, they also differ in the overall level of care they provide. I used a behavioral reaction norm approach to study individual variation in parental care behavior in free-living house sparrows (Passer domesticus). Unlike many amphibians, the paedomorphic axolotl (Ambystoma mexicanum) rarely undergoes external morphological changes indicative of metamorphosis. However, internally, some axolotl tissues undergo cryptic metamorphic changes. A previous study examined interspecific patterns of larval brain gene expression and found that these species exhibited unique temporal expression patterns that were hypothesized to be morph specific. This thesis tested this hypothesis by examining differences in brain gene expression between juvenile (JUV), paedomorphic (PAED), and metamorphic (MET) axolotls. I identified 828 genes that were expressed differently between JUV, PAED, and MET. Expression estimates from JUV were compared to estimates from PAED and …

Nuclear precursor of mature messenger RNA (pre-mRNA) splicing is one of the most highly regulated processes in eukaryotic organisms. In addition to its role in the removal of constitutive or alternative introns present in the pre-mRNA, splicing is also highly integrated into other layers of gene expression. This study investigates the potential role of the nuclear branchpoint binding protein (BBP) outside of the pre-mRNA splicing cycle. More specifically, we were interested in the biological relevance of its association with two cytoplasmic proteins Smy2 and Syh1. Following the 2010 Deepwater Horizon (DWH) spill, sediment cores were analyzed from marshes at various levels of oiling to determine how deeply oil penetrated sediment in these marsh environments, and if at these sites it had quantifiably affected benthic ecosystems. Minimum mixing depths were determined from penetration of the lithogenic radionuclide 234Th, which ranged from 0.25 to 4.5 cm. Sediment accumulation rates were determined using 210Pb, with verification from 137Cs in selected cores. Lead-210 profiles revealed long-term (decadal) mixing. Bay Jimmy, Louisiana was significantly affected by the DWH oil spill, as indicated by total polycyclic aromatic hydrocarbon …

This research examines the political geographies of Palestinian children, and the ways in which their everyday spaces and practices are shaped by broader social and political processes. This research begins with an investigation into the role of the child in the moral geopolitics of humanitarianism and the Palestinian/Israeli conflict. From here, the research explores how the competing discourses of Palestinian nationalism and international humanitarianism, and the legacy of forced migration, have shaped the subjectivity of Palestinian children and the spaces of childhood in a West Bank refugee camp, from homes, to schools, streets, and youth centers. Practitioners of ecological restoration are increasingly adopting a genetic perspective when recreating historical landscapes. Genes are often endowed with the capacity to reveal specific and distinct relationships between organisms and environments. In this dissertation, I examine how genetic technologies and concepts are shaping ecological restoration practices. This research is based on two and a half years of fieldwork in Chicago. I employed participant observation and semi-structured interviews to compare how restorationists in two plant science institutions employ genetic concepts in their projects. One institution uses high-tech genetic methods to guide practice while the other uses lower-tech genetic approaches. Each group …

Using learner centered instruction (LCI), a group of beef farmers were guided to design a farm in a layout that would most effectively offer continual learning opportunities for the beef practitioners themselves. Learner centered instruction was used as the primary teaching method for the project's completion. When using LCI, coupled with the correct facilitation of programs, student empowerment, and subject interest, positive experiences are more likely to result.

The purpose of this study was to identify risk and protective factors of preparatory and non-preparatory students within an agriculture education program at Anderson County High School in Lawrenceburg, Kentucky. The results of this study did not determine that there was a difference in risk and protective factors between preparatory and non-preparatory students within agriculture education courses. Pretreatment is an essential step in biofuel production from lignocellulose. Disruption of the lignin structure gives enzymes and fermentation organisms access to long chains of cellulose and hemicellulose. For this project’s purposes, the pretreatment must work within the framework of an on-farm butanol bioprocessing facility. Alkaline hydrogen peroxide (AHP) is a delignification method that potentially provides several advantages. At the alkaline pH, powerful hydroxyl radicals are formed; which attack lignin. Many aspects associated with conversion of lignocellulose to biofuels and other valuable products have been investigated to develop the most effective processes for biorefineries. Impervious surfaces such as roads, sidewalks, and roofs increase the volume of runoff generated in a watershed. Traditional stormwater management techniques emphasize conveyance of runoff away from impervious surfaces in order to reduce flooding. Rain gardens are becoming popular as a different means to manage stormwater in such a way that runoff is captured and infiltrated onsite rather than conveyed offsite. Coal-fired electricity production is the major emitter of CO2 and other greenhouse gases including NOx and SOx. Microalgae-based CO2 mitigation systems have been proposed to reduce the net CO2 emission from coal-fired power plants. The mammalian immune system is equipped to both eliminate pathogenic microorganisms and tumors, while remaining in homeostasis with commensal species at mucosal surfaces and tolerant towards self. Suppressor regulatory T cells (Tregs) are a major sentinel of this immunological tolerance. Induced Tregs (iTregs) arise in the periphery following the integration of cues from the metabolites, cytokines, etc. which make up its milieu. Dysregulation of iTreg development, function or homing underlies the etiology of many autoimmune diseases and immunopathologies. Due to the blood-brain barrier, delivery of many drugs to the brain has required intracranial surgery which is prone to complication. Here we show that Dopamine Neuron Stimulating Peptide 11 (DNSP-11), following non-invasive intranasal administration, protects dopaminergic neurons from a lesion model of Parkinson’s disease in the rat. A significant and dose-dependent increase in an index of dopamine turnover (the ratio of DOPAC to dopamine) was observed in the striatum of normal young adult Fischer 344 rats by whole-tissue neurochemistry compared to vehicle administered controls.

Parkinson’s disease, the second most common neurodegenerative disease, is characterized by the loss of dopaminergic neurons within the substantia nigra. Currently, the treatments available for PD are symptomatic treatments that do not stop the progression of the disease. Trophic molecules, such as glial cell-line derived neurotrophic factor (GDNF), have been evaluated as potential therapeutic molecules that could stop the loss of neurons and potentially restore some of the neurons that have already been lost. Traumatic brain injury (TBI) is a major cause of death and disability throughout the world with few pharmacological treatments available for individuals who suffer from neurological morbidities associated with TBI. Cellular and molecular pathological processes initiated at the time of injury develop into neurological impairments, with chronic sleep disorders (insomnia, hypersomnolence) being among the somatic, cognitive and emotional neurological impairments. Immediately post-injury, TBI patients report excessive daytime sleepiness, however, discordant opinions suggest that individuals should not be allowed to sleep or should be frequently awoken following brain injury. Metastasis is one of the main causes of death in cancer patients. Metastatic melanoma is a death sentence, as chemotherapeutic agents have a 5% success rate or do not extend survival beyond 10 months. The lack of effective chemotherapeutic agents for treating metastatic melanoma indicates a dire need to identify new drug targets and develop new therapies. Changes in both sleep architecture and cognition are common with age. Typically these changes have a negative connotation: sleep fragmentation, insomnia, and deep sleep loss as well as forgetfulness, lack of focus, and even dementia and Alzheimer’s disease. Research has shown that psychosocial stressors, such as isolation from family and friends or loss of a loved one can also have significant negative effects on sleep architecture and cognitive capabilities. This leaves the elderly in a particularly vulnerable situation: suffering from cognitive decline and sleep dysregulation already, and more likely to respond negatively to psychosocial stressors. Estrogens, such as the biologically active 17-b estradiol (E2) have many actions in the male and female brain. Not only does E2 regulate reproductive behavior in adults, it organizes and activates the brains of younger animals in a sex-specific manner. In addition, many human studies have shown E2 to provide protection against a variety of neurological disorders, including stoke. These studies have been controversial and depend largely on the type and timing of hormone replacement. Traumatic brain injury (TBI) is a prevalent CNS neurodegenerative condition that results in lasting neurological dysfunction, including potentially debilitating cognitive impairments. Despite the advancements in understanding the complex damage that can culminate in cellular dysfunction and loss, no therapeutic treatment has been effective in clinical trials, highlighting that new approaches are desperately needed. A therapy that limits cell death while simultaneously promoting reparative mechanisms, including post-traumatic neurogenesis, in the injured brain may have maximum effectiveness in improving recovery of function after TBI. Epileptogenesis is the complex process of the brain developing epileptic acitivity. Due to the role of glutamate and the hippocampus in synaptic plasticity a dysregulation in glutamate neurotransmission and hippocampal dysfunction are implicated in the process of epileptogenesis. However, the exact causal factors that promote epileptogenesis are unknown.
We study presynaptic proteins that regulate glutamate neurotransmission and their role in epileptogenesis. The presynaptic protein, tomosyn, is believed to be a negative regulator of glutamate neurotransmission; however, no one has studied the effects of this protein on glutamate transmission in vivo. In many areas of the agriculture, biological, physical and social sciences, spatial lattice data are becoming increasingly common. In addition, a large amount of lattice data shows not only visible spatial pattern but also temporal pattern (see, Zhu et al. 2005). An interesting problem is to develop a model to systematically model the relationship between the response variable and possible explanatory variable, while accounting for space and time effect simultaneously.

Cocaine is a widely abused drug without an FDA-approved medication. It has been recognized as an ideal anti-cocaine medication to accelerate cocaine metabolism producing biologically inactive metabolites via a route similar to the primary cocaine-metabolizing pathway, i.e. butyrylcholinesterase (BChE)-catalyzed hydrolysis. However, the native BChE has a low catalytic activity against cocaine. We recently designed and discovered a set of BChE mutants with a high catalytic activity specifically for cocaine. Currently, most of the RNAs used in lab research are prepared by in vitro transcription or chemical synthesis, which can be costly. In vivo expression in bacterial cells is another approach to RNA preparation that allows large scale production at a lower cost. However, there are some obstacles in bacterial expression, including RNA degradation in host cell, as well as RNA extraction and purification. tRNA and 5S RNA have been reported as scaffolds to circumvent the degradation problem. Dementia is increasingly recognized as a major and growing threat to public health worldwide, and there is a critical need for prevention and treatment strategies. However, it is necessary that appropriate methodologies are used in the identification of risk factors. The purpose of this dissertation research was to develop further the body of literature featuring Markov chains as an analytic tool for data derived from longitudinal studies of aging and dementia.
