Tobacco use is the leading cause of preventable death in the United States. The Family Smoking Prevention and Tobacco Control Act was signed into law in 2009 and gave the U.S. Food and Drug Administration (FDA) the authority to implement pictorial health warning labels (HWLs) on cigarette packages. Multiple studies investigating self-reported affective, cognitive and behavioral impacts of HWLs suggest that the most effective warnings include imagery that depicts physical damage to the body due to smoking. However, self-report methods of assessment used in these studies may be biased. Far less is known about how HWLs directly modulate brain activity. …

A persistent question in epigenetics is how heterochromatin is targeted for assembly at specific domains, and how that chromatin state is faithfully transmitted. Stable heterochromatin is necessary to silence transposable elements (TEs) and maintain genome integrity. Both the RNAi system and heterochromatin components HP1 (Swi6) and H3K9me2/3 are required for initial establishment of heterochromatin structures in S. pombe. Here we utilize both loss of function alleles and the newly developed Drosophila melanogaster transgenic shRNA lines to deplete proteins of interest at specific development stages to dissect their roles in heterochromatin assembly in early zygotes and in maintenance of the silencing …

Head and neck (HN) cancer is the fifth most prevalent malignancy worldwide with 90% of them categorized as squamous cell carcinomas (SCC). Carcinomas of the oropharynx, pharynx and the oral cavity, comprise a subset of HNSCC and are referred to as oropharyngeal squamous cell carcinomas (OPSCC). Up to 60% of OPSCC and 25% of HNSCC are positive for high-risk human papillomavirus (HR-HPV), primarily HPV16. HPV positive and HPV negative OPSCC’s are molecularly and biologically distinct with differences in risk factors, age of presentation and clinical behavior. High-throughput proteomics analysis involves the rapid identification and characterization of large sets of proteins in complex biological samples. Tandem mass spectrometry (MS/MS) has become the leading approach for the experimental identification of proteins. Accurate analysis of the data produced is a computationally challenging process that relies on a complex understanding of molecular dynamics, signal processing, and pattern classification. In this work we address these modeling and classification problems, and introduce an additional data-driven evolutionary information source into the analysis pipeline.

Hepatitis C virus: HCV) is positive strand, blood-borne, hepatotropic RNA virus that causes chronic infection in ~170 million people worldwide and is the leading cause of liver transplantation in the United States. HCV entry and attachment is mediated by the envelope protein E2 through interaction with several cellular receptors including CD81, scavenger receptor B1: SRB-1), claudin-1, and occludin, although the exact mechanism by which these receptors facilitate infection remains unclear, largely due to the absence of a structural model of E2. It greatly influences the performance of fuel cells, especially the concentration losses. Two approaches were developed to improve flow field designs in this dissertation. Competitive social interactions are ubiquitous in nature, but their genetic basis is difficult to determine. Much can be learned from single gene knockouts in a eukaryote microbe. The mutants can be competed with the parent to discern the social impact of that specific gene. Dictyostelium discoideum is a social amoeba that exhibits cooperative behavior in the construction of a multicellular fruiting body. An exploratory study was done to unveil a more contemporary and intimate look at women and aging in U.S. society. This qualitative research explored what women do, if anything, to confront pressures associated with aging. Purposeful sampling was used to homogenize the participant sample for increased richness of data. Criterion was as follows: women, 50’s age range, married with children, Caucasian, from the Midwest, and who currently use one or more anti‐aging products. "This thesis investigates the influence of random disorder and dissipation on zero-temperature quantum phase transitions. Both phenomena can fundamentally change the character of the phases of a quantum many-particle system and of the transitions between them. If dissipation and disorder occur simultaneously in a system undergoing a quantum phase transition, particularly strong effects can be expected. In the first paper reproduced in this thesis, we study a single quantum rotor coupled to a sub-Ohmic dissipative bath. We find that this system undergoes a quantum phase transition from a delocalized phase to a localized phase as the dissipation strength is increased. …

"Explosives and energetics are common soil and groundwater pollutions. This research was to develop novel phytoforensics approaches on energetics. Four different plants species, including woody perennial trees and monocot grasses, were planted both in soil and sand reactors with continuous exposure to a mixture of explosives in the greenhouse. Time dependent assessments were carried out to determine kinetic mechanisms of uptake, transport and accumulation. Plant concentrations were analyzed by both traditional solvent extraction and novel sap analysis methods. "Acetaminophen (APAP) is the most widely used pharmaceutical analgesic-antipyretic agent in the world, but its toxicity is a common cause of drug-induced hepatotoxicity. With APAP toxicity, cellular glutathione (GSH) is depleted. This results in the availability of N-acetyl-p-benzoquinone imine (NAPQI), is a toxic metabolite of APAP that binds to cellular macromolecules, which leads to cell necrosis. N-acetyl cysteine (NAC), a GSH precursor, is the only approved antidote for an acetaminophen overdose. It is a negatively charged molecule that diminishes its penetration into the cells, thereby requiring fairly high doses that increase the severity of side effects. In addition, oral and …

Polymer electrolyte membrane (PEM) fuel cells in particular are researched because of their high power density, large range of operating conditions, green products, and ease of scalability. These issues include variations in reactant distribution, materials issues for the bipolar plate, and flooding caused by poor water management. "Driven micro-cavities embedded in the wall beneath turbulent supersonic boundary layers are analyzed using two-dimensional computational fluid dynamics. This concept is a passive flow control technique in which very small cavities formed by arrays of thin vertical walls are oriented transverse to the flow direction and underlie the boundary layer. The purpose is to reduce or eliminate skin friction drag. Various micro-cavity configurations were analyzed at locations (0.1 m and 1 m) downstream of the leading edges of flat plates, for free-stream Mach numbers of 1.2, 2.0, and 3.0. This case study explored the impact of a scheduling intervention on the Georgia High School Graduation Test standardized achievement test scores of students identified as in need of remediation in the content areas of English and mathematics, paired with the perception of this scheduling model from key informants in the study. Students in the scheduling intervention were enrolled in a year-long, alternating content-day block scheduling model. The streaming model is a popular model for writing high-throughput parallel applications. A streaming application is represented by a graph of computation stages that communicate with each other via FIFO channels. In this report, we consider the problem of mapping streaming pipelines — streaming applications where the graph is a linear chain — in order to maximize throughput. In a parallel setting, subsets of stages, called components can be mapped onto different computing resources. Urban American Indians: A Comparison Of Father Involvement Predictors Across Race, Jonathan Padilla, Peter Ward, Gordon E. Limb

Father involvement in families of urban American Indians has been a neglected area of research. In this study, the authors examined the associations among parental relationship quality, father involvement, and coresidence. The authors conducted a multiple group analysis through structural equation modeling comparing 107 American Indian fathers with fathers of the general population in the Fragile Families and Child Wellbeing Study. Although the results were similar between groups, findings indicated two key differences for urban American Indians. First, emotional supportiveness had a stronger relationship with physically active engagement for American Indian fathers. Second, parental relationship quality had greater total standardized …

The development of a novel micro-system integrating SU-8 polymer micro-grippers with a tensile force sensor for handling and characterizing the mechanical properties of delicate biological materials, such as fibrils, is presented. The micro-grippers are actuated by the electro-thermal effect and have gripping forces comparable to the common 'hot-and-cold-arm' grippers. A robust finite element model was developed to investigate system performance and validated experimentally. A new micro-mechanical calibration method using a piezoelectric manipulator with a micro-force measurement system was successfully applied to test the structure. Coral reefs across the world are under threat from a range of stressors, and while there has been considerable focus on the impacts of these stressors on corals, far less is known about their effect on other reef organisms. The 1997-8 El-Niño Southern Oscillation (ENSO) had notable and severe impacts on coral reefs worldwide, but not all reef organisms were negatively impacted by this large-scale event. Objective: To evaluate the midterm results of Contegra conduit.
Methods: The retrospective study comprised patient record at Aga Khan University Hospital, Karachi, of conduits implanted between May 2007 and June 2012. Data collection was made from the clinical notes and from serial echocardiograms by a single cardiologist. The last followup echocardiography was done at the time of data collection in June 2012. SPSS 19 was used for statistical analysis.
Results:
A total of 18 conduits had been implanted (16-22mm) during the study period. Median age at the time of surgery was 9 years (range: 2.5-16 years). Early mortality was seen …

Objective: The objective of this study was to evaluate the frequency and outcome of graft versus host disease after stem cell transplantation for various haematological disorders in Pakistan.
Materials and Methods: Pretransplant workup of the patient and donor was performed. Mobilization was done with G-CSF 300 mu g twice daily for five day. Standard GvHD prophylaxis was done with methotrexate 15mg/m(2) on day +1 followed by 10mg/m(2) on days +3 and +6 and cyclosporine. Grading was done according to the Glucksberg classification.

Focal adhesions are specialized cell contact sites of distinct molecular composition and structure that bridge the actin cytoskeleton to the extracellular matrix and provide for efficient bidirectional transmission of biochemical and mechanical signals between the intra- and extracellular compartment.
