This case study dissertation examines the implementation of digital technology in a mid-sized public school district in southern Minnesota. The methodology involved unstructured interviews and close observation of several teachers, administrators and related staff in the junior-senior high building. These observations were presented in informative vignettes that help to tell the story of the culture of innovation in this school, as well as highlight some of the implementation strategies that aid this school and district in meeting its goal to meet all learners in a 21st century society. Inclusion is a step in the right direction for providing equal opportunities for those with disabilities. Inclusion is the integration of special education students in general education classrooms to the maximum extent which is appropriate for the individual. Court cases throughout history have paved the way for equality for all students. Great movements such as Individuals with Disabilities Education Act (IDEA) in 2001 began to make a change in the education of students with disabilities. Alzheimer’s disease (AD) is characterized by neurofibrillary tangles and extracellular amyloid-β plaques (Aβ). Despite ongoing research, some ambiguity remains surrounding the role of Aβ in the pathogenesis of this neurodegenerative disease. While several studies have focused on the mutations associated with AD, our understanding of the epigenetic contributions to the disease remains less clear. To that end, we determined the changes in DNA methylation in differentiated human neurons with and without Aβ treatment. We isolated the DNA from neurons treated with Aβ or vehicle, and digested the two samples with either a methylation-sensitive (HpaII) or a methylation-insensitive (MspI) restriction endonuclease. …

Our nation’s colleges and universities have always sought to prepare their graduates for life and work in their own era. The pressures we face today, both from outside the academy and within the higher education community, are complex, interlocking, and hard to manage. Some of these challenges require us to rethink what it means to be educated in today’s world and to explore ways to provide a coherent and meaningful educational experience in the face of the turbulence, uncertainty, and fragmentation that characterizes much of higher education today.

Climate change is changing not only our physical world, but also our intellectual, social, and moral worlds. We are realizing that our situation is profoundly unsafe, interdependent, and uncertain. What, then, does climate change demand of economists, as human beings and as professionals? A discipline of economics based on Enlightenment notions of mechanism and disembodied rationality is not suited to present problems. This essay suggests three major requirements: first, that we take action; second, that we work together; and third, that we focus on avoiding the worst, rather than obtaining the optimal. Regulatory T cells (Tregs) are required for the induction and maintenance of immune homeostasis. Singh et al. demonstrated that depletion of Tregs in mice results in a loss of self-tolerance that manifests in the development of autoimmune diseases such as Inflammatory Bowel Disease (IBD). In humans, autoimmune diseases such as IBD, Multiple Sclerosis (MS), and Type 1 Diabetes are thought to occur due to a deficiency in the number or function of Tregs. In his Century Cycle of plays, August Wilson tells ten distinct stories of families in or linked to the Hill District, an African American community in Pittsburgh; one play taking place in each decade of the twentieth century. Through these plays, Wilson's audience sees the Hill District and America evolve, while prejudice, oppression, and poverty remain constant. Many scholars argue that sexism provides a fourth common factor, asserting that Wilson portrays the female characters in the male-fantasized, stereotypical roles of the Mammy or the Jezebel figure, rather as realistic, empowered, and complex women. Biting flies are economically important blood-feeding pests of medical and veterinary significance. Chemosensory-based biting fly behaviours, such as host/nutrient source localization and ovipositional site selection, are intriguing targets for the development of supplemental control strategies. In an effort to expand our understanding of biting fly chemosensory pathways, transcripts encoding the highly conserved insect odorant co-receptor (Orco) were isolated from two representative biting fly species, the stable fly (Scal\Orco) and the horn fly (Hirr\Orco). Orco forms a complex with an odour-specific odorant receptor to form an odour-gated ion channel. The biting fly transcripts were predicted to encode …

Summary. Erebia serotina was described in 1953 as a scarce, low-elevation endemic Pyrenean species flying late in the season. At least 34 individuals are known from various locations. However, the absence of females suggests a hybrid origin, and E. epiphron and either E. pronoe or E. manto have been proposed as possible parents. Electrophoretic analysis of five allozyme loci and sequencing of three mitochondrial DNA segments and one nuclear gene now demonstrate that E. serotina results from the cross between E. epiphron females and E. pronoe males. Several fungal pathogens have been identified on ornamental and native stands of switchgrass (Panicum virgatum L.). Diseases of switchgrass, particularly rust, have been largely neglected and are likely to become the major limiting factor to biomass yield and quality, especially when monocultured over a large acreage. Based on teliospore morphology and internal transcribed spacer-based diagnostic primers, the rust pathogen collected from switchgrass research fields in Oklahoma was identified as Puccinia emaculata. Sustainable bioenergy production depends upon the efficiency with which crops use available water to produce biomass and store carbon belowground. Therefore, water use efficiency (WUE; productivity vs. annual evapotranspiration, ET) is a key metric of bioenergy crop performance. We evaluate WUE of three potential perennial grass bioenergy crops, Miscanthus × giganteus (miscanthus), Panicum virgatum (switchgrass), and an assemblage of prairie species (28 species), and Zea mays–Glycine max rotation, during the establishment phase in Illinois. Neosporosis is an important cause of bovine abortion worldwide. Many aspects of transmission of Neospora caninum in nature are unknown. The white-tailed deer (Odocoileus virginianus) is considered one of the most important wildlife reservoirs of N. caninum in the USA. During the hunting seasons of 2008, 2009, and 2010, brains of 155 white-tailed deer fetuses were bioassayed in mice for protozoal isolation. Viable N. caninum (NcWTDMn1, NcWTDMn2) was isolated from the brains of two fetuses by bioassays in mice, and subsequent propagation in cell culture. Dams of these two infected fetuses had antibodies to N. caninum by Neospora …

Genetic transformation via the biolistic method has been used to introduce genes encoding natural and novel high-molecular-weight glutenin subunits (HMW-GS) into wheat. The appearance of new seed proteins of sizes not predicted by the transgene coding sequences was noted in some experiments. In this report, the identities of thirteen of these novel proteins were determined by tandem mass spectrometry (MS/MS). Seven different proteins larger than and two proteins smaller than the native protein were shown to contain peptides from 1Dx5. The potential for improved management of Cercospora leaf spot (CLS), caused by Cercospora beticola, using the herbicide glyphosate in glyphosate-resistant sugar beet varieties was investigated. Controlled field experiments were conducted in 2008 and 2009 to determine if glyphosate and glyphosatee‒fungicide combinations improved the management of CLS in four commercial varieties of glyphosate-resistant sugar beet. Variety and fungicide main effects were significant for CLS development. However, regardless of the herbicide program, glyphosate or a conventional herbicide program, CLS development was not affected. Leaf chlorophyll content is an important variable for agricultural remote sensing because of its close relationship to leaf nitrogen content. The US dairy industry has realized tremendous improvements in efficiencies and milk production since the 1940s. During this time, farm and total cow numbers have decreased and average herd size has increased. This intensification, combined with the shift to a largely urban public, has resulted in increased scrutiny of the dairy industry by social and environmental movements and increased concern regarding the dairy industry’s sustainability. In response to these concerns, a group of scientists specializing in animal welfare, nutrient management, greenhouse gas emissions, animal science, agronomy, agricultural engineering, microbiology, and economics undertook a critical review of the US dairy industry. …

Trichinella is an important parasitic nematode of animals worldwide. Heat shock proteins are ubiquitous in nature and allow organisms to quickly respond to environmental stress. A portion of the Tsdaf-21 gene, a Caenorhabditis elegans daf-21 homologue encoding heat-shock protein 90 (Hsp90) was cloned from Trichinella spiralis. The partial nucleotide sequence resided near the 5′-end of the gene and encoded a polypeptide of 254 amino acid residues harboring a HATPase-c superfamily domain and Hsp90 protein domain. Phylogenetic analysis revealed that Tsdaf-21 is highly conserved and formed a monophyletic clade with other nematodes. The partial Tsdaf-21 transcript was subcloned and expressed for …

Background: Bovine tuberculosis (bTB) in wildlife species poses a threat to domestic livestock in many situations. Control programs for bTB in livestock depend on testing and slaughtering the positive animals; however, the currently available diagnostic tests often have poor specificity. In our previous study, we developed a specific and sensitive enzyme linked immunosorbent assay (ELISA) for another mycobacterial disease – Johne’s disease, using surface antigens of Mycobacterium avium ssp. paratuberculosis (MAP) extracted by briefly agitating the bacilli in 80% ethanol solution. Commissioned by President Dwight Eisenhower in 1958 and opened with a dedication ceremony in December 1961, the USDA, Agricultural Research Service (ARS), National Animal Disease Center (NADC) celebrated its 50-year anniversary in November 2011. Over these 50 years, the NADC established itself among the world’s premier animal health research centers. This review provides a critical overview of conservation practices that are aimed at improving water quality by retaining phosphorus (P) downstream of runoff genesis. The review is structured around specific downstream practices that are prevalent in various parts of the United States. Specific practices that we discuss include the use of controlled drainage, chemical treatment of waters and soils, receiving ditch management, and wetlands. The review also focuses on the specific hydrology and biogeochemistry associated with each of those practices. Biomass composition of switchgrass (Panicum virgatum L.) can affect its utilization by ruminants and its conversion to liquid fuels in a biorefinery. The objective of this study was to evaluate the effects of six generations of divergent breeding for forage in vitro dry matter digestibility (IVDMD) on switchgrass biomass composition, forage quality traits, and ethanol yield. Initially there was one cycle of selection for both low (C-1) and high IVDMD (C1 = cv. Trailblazer), followed by four additional breeding cycles for high IVDMD. Variable rate irrigation (VRI) systems are commercially available and can easily be retrofitted onto moving sprinkler systems. However, there are few reports on the application performance of such equipment. In this study, application uniformity of two center pivots equipped with a commercial VRI system were tested using fixed-plate sprinklers and a range of irrigation rates (100%, 80%, 70%, 50%, and 30%). Quantifying the effects of seasonal temperature and precipitation on cow–calf production on rangelands is challenging, as few long-term (420 yrs) studies have been reported. However, an understanding of how seasonal weather inconsistency affects beef production is needed for beef producers to better manage their herds on native rangelands to minimize enterprise risk with respect to climatic variability. This study was designed to determine whether methyl-β-cyclodextrin (MCD) can substitute for albumin in incubation medium for neonatal swine adipose tissue explants, or whether MCD affects metabolism and cytokine expression. Subcutaneous adipose tissue explants (100±10 mg) were prepared from 21-day-old pigs. Explants were incubated in medium 199 supplemented with 25mM HEPES, 1.0 nM insulin at 37°C. The medium also contained bovine serum albumin (BSA) or MCD at 0%, 0.05%, 0.1%, 0.2% or 0.3%. Tissue explants were treated with these media for 1 h and then switched to the same basal incubation medium containing 0.05% BSA. Explants were removed from basal …
