The Role Of Dax-1 In Regulating Pluripotency In Mouse Embryonic Stem Cells, Anthony Torres Jan 2013

The Role Of Dax-1 In Regulating Pluripotency In Mouse Embryonic Stem Cells, Anthony Torres

Master's Theses

The orphan receptor Dax-1 is highly expressed in pluripotent embryonic stem (ES) cells and shows a correlative reduction in expression as these cells differentiate. While it is known that Dax-1 is expressed in pluripotent mouse ES cells, the precise function of Dax-1 in these cells is not as well understood. Recent studies employing RNA interference (RNAi) to specifically reduce the expression of the Dax-1 gene in mouse ES cells found that upon the knock down of Dax-1, ES cells differentiated. These findings indicate that Dax-1 functions in a novel role in the maintenance of a relatively undifferentiated state in ES …

He Works And She Does Too : Race And Gender Differences In Relationship Between Earner Status, Marital Satisfaction, And Division Of Family Tasks, Samantha M. Rivera Jan 2013

He Works And She Does Too : Race And Gender Differences In Relationship Between Earner Status, Marital Satisfaction, And Division Of Family Tasks, Samantha M. Rivera

Theses, Dissertations, and Projects

This study sought to fill gaps in literature by exploring similarities and differences in marital satisfaction and division of family tasks between Caucasian and Mexican-American dualearner and single-earner couples. The subsample included in the analysis was a subset from the Supporting Father Involvement study based in California. Participants were 522 couples, with two-thirds Mexican-American and approximately one fourth Caucasian. The study examined the following four questions: (1) How are race/ethnicity and couple's earner status associated with marital satisfaction? (2) Does the SFI intervention affect couple satisfaction differently for distinct racial/ethnic and earner groups? (3) How are race/ethnicity and couple's earner …

Biomarkers Of Maternal And Fetal Exposure To Organochlorine Pesticides Measured In Pregnant Hispanic Women From Brownsville, Texas, Ken Sexton, Jennifer J Salinas, Thomas J Mcdonald, Rose M Z Gowen, Rebecca P Miller, Joseph B Mccormick, Susan P Fisher-Hoch Jan 2013

Biomarkers Of Maternal And Fetal Exposure To Organochlorine Pesticides Measured In Pregnant Hispanic Women From Brownsville, Texas, Ken Sexton, Jennifer J Salinas, Thomas J Mcdonald, Rose M Z Gowen, Rebecca P Miller, Joseph B Mccormick, Susan P Fisher-Hoch

Faculty, Staff and Student Publications

Biomarkers of organochlorine pesticides were measured in both venous and umbilical cord blood from 35 pregnant Hispanic women living in Brownsville, Texas, USA. Gas chromatography with an electron capture detector was used to analyze specimens for 30 individual pesticides or their metabolites. Results indicate that blood concentrations were relatively low for most individual compounds, but that high-end (upper 10th percentile) values for total DDT were comparatively high. Although health effects associated with measured blood concentrations are uncertain, there is concern that fetal exposure to low levels of these OC compounds, either individually or in combination, might contribute to subsequent health …

Defining A Simplified Pharmacophore For Simocyclinone D8 Inhibition Of Dna Gyrase, Lauren Gaskell Jan 2013

Defining A Simplified Pharmacophore For Simocyclinone D8 Inhibition Of Dna Gyrase, Lauren Gaskell

Theses and Dissertations

The type II topoisomerase subfamily of enzymes has been clinically targeted by the widely used, broad-spectrum quinolone class of antibacterials. Due to emerging drug-resistant strains of bacteria, the quinolones’ effectiveness is threatened. The natural product simocyclinone D8 (SD8) has shown the ability to inhibit the type II topoisomerase, DNA gyrase, even when mutated to be resistant to the quinolones. In order to determine the pharmacophore required for SD8 binding to DNA gyrase, 16 compounds were synthesized. These compounds were then tested by surface plasmon resonance for their ability to inhibit the DNA – DNA gyrase binding interaction. It was found …

Computational Analysis Of Rho Gtpase Cycling, Cibele Vieira Falkenberg, Leslie M. Loew Jan 2013

Computational Analysis Of Rho Gtpase Cycling, Cibele Vieira Falkenberg, Leslie M. Loew

UCHC Articles - Research

The Rho family of GTPases control actin organization during diverse cellular responses (migration, cytokinesis and endocytosis). Although the primary members of this family (RhoA, Rac and Cdc42) have different downstream effects on actin remodeling, the basic mechanism involves targeting to the plasma membrane and activation by GTP binding. Our hypothesis is that the details of GTPase cycling between membrane and cytosol are key to the differential upstream regulation of these biochemical switches. Accordingly, we developed a modeling framework to analyze experimental data for these systems. This analysis can reveal details of GDI-mediated cycling and help distinguish between GDI-dependent and -independent …

From August 22 to September 16, 2012, atmospheric mercury (Hg) was measured from a common manifold in the field during the Reno Atmospheric Mercury Intercomparison eXperiment. Data were collected using Tekran systems, laser induced fluorescence, and evolving new methods. The latter included the University of Washington-Detector for Oxidized Mercury, the University of Houston Mercury instrument, and a filter-based system under development by the University of Nevada-Reno. Good transmission of total Hg was found for the manifold. However, despite application of standard protocols and rigorous quality control, systematic differences in operationally defined forms of Hg were measured by the sampling systems. …

Dna-Binding Motif And Target Genes Of The Imprinted Transcription Factor Peg3, Michelle M. Thiaville, Jennifer M. Huang, Hana Kim, Muhammad B. Ekram, Tae Young Roh, Joomyeong Kim

Faculty Publications

The Peg3 gene is expressed only from the paternally inherited allele located on proximal mouse chromosome 7. The PEG3 protein encoded by this imprinted gene is predicted to bind DNA based on its multiple zinc finger motifs and nuclear localization. In the current study, we demonstrated PEG3's DNA-binding ability by characterizing its binding motif and target genes. We successfully identified target regions bound by PEG3 from mouse brain extracts using chromatin immunoprecipitation analysis. PEG3 was demonstrated to bind these candidate regions through the consensus DNA-binding motif AGTnnCnnnTGGCT. In vitro promoter assays established that PEG3 controls the expression of a given …

Publications and Research

The ultimate goal of this multi-article series is to develop a methodology to generate continuous fields of tree height and biomass. The first paper demonstrated the need for Allometric Scaling and Resource Limitation (ASRL) model optimization and its ability to generate spatially continuous fields of tree heights over the continental USA at coarse (1 km) spatial resolution. The objective of this second paper is to provide an assessment of that approach at site scale, specifically at 12 FLUXNET sites where more accurate data are available. Estimates of tree heights from the Geoscience Laser Altimeter System (GLAS) waveform data are used …
