Publications and Research
The banana passion fruit (Passiflora tripartita Breiter, Passifloraceae) known as “tumbo” is very appreciated in tropical and subtropical countries of South America. Methanolic extracts from peel and the fruit juice of P. tripartita growing in Chile were analyzed for antioxidant capacity as well as for flavonoid and phenolic content. A chromatographic method was developed for the rapid identification of the main phenolics in the samples by HPLC-DAD and HPLC-MS. The fast fingerprint analysis allowed the detection of eighteen flavonoid C-glycosides and four flavonoid O-glycoside derivatives which were characterized by UV spectra and ESI-MS-MS analysis. Several of the C-glycosides detected are …

Automated Point-Of-Care Image Processing Methodology For The Diagnosis Of Malaria, Michael B. Jorgensen
Automated Point-Of-Care Image Processing Methodology For The Diagnosis Of Malaria, Michael B. Jorgensen
Master's Theses
Malaria has profoundly influenced human history for over four thousand years and despite numerous attempts at eradication, the prevention, diagnosis, and treatment of malaria have been largely ineffective. More than five hundred million people are affected by malaria every year resulting in over one million deaths. Drug resistance development by the parasite has diminished the effectiveness of numerous treatment options due, in part, to overtreatment of negative patients based on insufficient clinical algorithms and diagnostic methods. The goal of this research was to develop an image analysis algorithm to diagnose malaria with a high degree of sensitivity and specificity in …

Purpose: Notophthalmus viridescens, the red-spotted newt, possesses tremendous regenerative capabilities. Among the tissues and organs newts can regenerate, the lens is regenerated via transdifferentiation of the pigment epithelial cells of the dorsal iris, following complete removal (lentectomy). Under normal conditions, the same cells from the ventral iris are not capable of regenerating. This study aims to further understand the initial signals of lens regeneration.
Investigating Gp96 Biology And Its Macrophage-Intrinsic Role In Colitis-Associated Colon Tumorigenesis, Crystal Morales
Investigating Gp96 Biology And Its Macrophage-Intrinsic Role In Colitis-Associated Colon Tumorigenesis, Crystal Morales
Doctoral Dissertations
Gp96, a mammalian endoplasmic reticulum chaperone, is required for expression of multiple Toll-Like Receptors (TLRs) and integrins. This heat shock protein is upregulated in cancer and tumor associated macrophages, and it is known to cross-present tumor antigens. A better understanding of gp96 biology will aid the design of cancer drugs, as gp96 is a possible target. To study this molecule, we investigated an orthologous system in Drosophila. A BLAST search identified Drosophila gp93, an uncharacterized molecule, with 74% amino acid homology. Gp93 was found to rescue gp96 client expression in a gp96-deficient murine pre-B cell line. It was further …

Scanning In Situ Spectroscopy Pplatform For Imaging Surgical Breast Tissue Specimens, Venkataramanan Krishnaswamy, Ashley M. Laughney, Wendy A. Wells, Keith D. Paulsen, Brian W. Pogue
Dartmouth Scholarship
A non-contact localized spectroscopic imaging platform has been developed and optimized to scan 1 x 1 cm² square regions of surgically resected breast tissue specimens with ~150-micron resolution. A color corrected, image-space telecentric scanning design maintained a consistent sampling geometry and uniform spot size across the entire imaging field. Theoretical modeling in ZEMAX allowed estimation of the spot size, which is equal at both the center and extreme positions of the field with ~5% variation across the designed waveband, indicating excellent color correction. The spot sizes at the center and an extreme field position were also measured experimentally using the …

Adult central nervous system (CNS) neurons fail to regenerate following injury, and there is no repair or replacement of cells lost after injury or in neurodegenerative diseases. There is much interest in transplanting stem cell-derived neurons into the injured nervous system and enhancing the differentiation of donor cells into mature, integrated and functional neurons. Little is known, however, about what signals control the differentiation and integration of neurons, either during development or in the adult. Generating appropriate types of donor neurons from stem cells has been challenging because the signals that regulate neural subtype-specific fates are largely unknown. Therefore, it …

Neutrophils are characterized by their distinct nuclear shape, which is thought to facilitate the transit of these cells through pore spaces less than one-fifth of their diameter. We used human promyelocytic leukemia (HL-60) cells as a model system to investigate the effect of nuclear shape in whole cell deformability. We probed neutrophil-differentiated HL-60 cells lacking expression of lamin B receptor, which fail to develop lobulated nuclei during granulopoiesis and present an in vitro model for Pelger-Huët anomaly; despite the circular morphology of their nuclei, the cells passed through micron-scale constrictions on similar timescales as scrambled controls. We then investigated the …
