The cellular stress response (CSR) is one of the most highly conserved mechanisms among all organisms. Cellular stress can be defined as damage or the threat of damage to proteins, macromolecules and/or DNA. The response to damage can involve cell cycle regulation, protein chaperoning, DNA repair or, if macromolecular damage is too severe, apoptotic mechanisms can be initiated. This thesis details experiments that were designed to examine the cellular response to non-lethal environmental stressors at the protein level, using two fish species as study models. Two proteins that can cause cell cycle arrest and apoptosis mechanisms were examined. Expression of …

The Negative Regulation Of The Antiviral Signaling Pathway, Soratree Charoenthongtrakul
Open Access Dissertations
A key feature of the innate antiviral immune response is a rapid nonspecific response to virus infection largely mediated by the induction and extracellular secretion of type I interferons (IFNs) that restrict virus replication. Cytoplasmic sensors such as RIG-I recognize viral RNA and trigger antiviral signaling pathways that upregulate IFN transcription. However, it remains largely unknown how antiviral signaling is negatively regulated to maintain homeostasis after the elimination of virus. We have identified the RING domain-containing protein RING finger 11 (RNF11) as a novel negative regulator of innate antiviral signaling. Overexpression of RNF11 downregulated IFN-β expression and enhanced viral replication …

The Function Of Yeast Pas Kinase, Dr. Julianne Grose
The Function Of Yeast Pas Kinase, Dr. Julianne Grose
Journal of Undergraduate Research
PAS kinase is a newly discovered member of the nutrient sensing kinases that regulates glucose homeostasis in mice and yeast. The aim of our 2009 MEG “The Function of Yeast PAS kinase” was to characterize the role of a known PAS kinase substrate, Ugp1, as well as to identify novel PAS kinase substrates. We have made significant progress on each of these aims, which has facilitated the preparation of publications, presentations and abstracts. In addition, the findings described below have opened up further research as evidenced by our 2010 and 2011 MEG proposals as well as a 2009 NIH grant …

High-Activity Mutants Of Butyrylcholinesterase For Cocaine Hydrolysis And Method Of Generating The Same, Chang-Guo Zhan, Hoon Cho, Hsin-Hsiung Tai
Pharmaceutical Sciences Faculty Patents
A novel computational method and generation of mutant butyrylcholinesterase for cocaine hydrolysis is provided. The method includes molecular modeling a possible BChE mutant and conducting molecular dynamics simulations and hybrid quantum mechanical/molecular mechanical calculations thereby providing a screening method of possible BChE mutants by predicting which mutant will lead to a more stable transition state for a rate determining step. Site-directed mutagenesis, protein expression, and protein activity is conducted for mutants determined computationally as being good candidates for possible BChE mutants, i.e., ones predicted to have higher catalytic efficiency as compared with wild-type BChE. In addition, mutants A199S/A328W/Y332G, A199S/F227A/A328W/Y332G, A199S/S287G/A328W/Y332G, …
The Role Of Nuclear Bmp2 (Nbmp2) In Skeletal Muscle, Dr. Laura Bridgewater
The Role Of Nuclear Bmp2 (Nbmp2) In Skeletal Muscle, Dr. Laura Bridgewater
Journal of Undergraduate Research
The goal of this project was to characterize the molecular pathway by which nBmp2 disrupts Ca2+ handling in skeletal muscle of nBmp2 mutant mice. Previously, we had worked with Dr. Chad Hancock’s lab to measure muscle function in nBmp2 mutant mice, and those results showed a slowing in the rate at which Ca2+ is pumped from the cytoplasm back into the sarcoplamic reticulum at the conclusion of a skeletal muscle contraction. Our rationale for this MEG study was that characterizing the muscle dysfunction phenotype in the nBmp2 mutant mice would ultimately lead to a molecular level understanding of the functions …

Quantum Tricks In The Shadows Of Relativity And The Coulomb Force: Teaching Devices And Students How To Think Ultra-Fast, Ilias Perakis
Physics Seminars
The technological demand to bump the Gigahertz switching speed limit of today's magnetic memory and logic devices into the Terahertz regime underlies the entire field of spin-electronics and integrated multi-functional nano-devices. In this talk, I use theory and experiment to show how all-optical switching based on the quantum-mechanical manipulation of spins with a train of laser pulses could meet this challenge. The creation of magnetic correlations within femtoseconds, i.e. faster than one period of lattice oscillations, reveals a new temporal regime of magnetism. This field opened new directions for manipulating materials out-of-equilibrium using Terahertz, Mid-infrared, and X-ray pulses.

Science, Technology, And Catholic Identity In The Education Of Professionals, Keith Douglass Warner, David S. Caudill
Journal of Catholic Education
The reception of Ex corde ecclesiae has been uneven across the disciplines, with scant interest in distinctly Catholic pedagogies outside of the humanities. This essay argues that Catholic universities can distinguish themselves by how they present science and technology in their curriculum by drawing from the interdisciplinary field of “science, technology & society,” or STS. We argue that discussions about Catholic identity, science, and human values can and should extend into the curriculum while simultaneously safeguarding academic freedom, and that this can readily be done in professional schools, such as law and engineering. We outline the contributions that STS as …

Mitochondrial Genetics Of Alzheimer's Disease And Aging, Perry Gene Ridge
Mitochondrial Genetics Of Alzheimer's Disease And Aging, Perry Gene Ridge
Theses and Dissertations
Mitochondria are essential cellular organelles and the location of the electron transport chain, the site of the majority of energy production in the cell. Mitochondria contain their own circular genome approximately 16,000 base pairs in length. The mitochondrial genome (mtDNA) encodes 11 protein-coding genes essential for the electron transport chain, 22 tRNA genes, and two rRNA genes. Mitochondrial malfunction occurs in many diseases, and changes in the mitochondrial genome lead to numerous disorders. Multiple mitochondrial haplotypes and sequence features are associated with Alzheimer's disease. In this dissertation we utilized TreeScanning, an evolutionary-based haplotype approach to identify haplotypes and sequence variation …

Marked Improvement Of Cytotoxic Effects Induced By Docetaxel On Highly Metastatic And Androgen-Independent Prostate Cancer Cells By Downregulating Macrophage Inhibitory Cytokine-1., M Mimeault, Sonny L. Johansson, Surinder K. Batra
Journal Articles: Biochemistry & Molecular Biology
BACKGROUND: Overexpression of macrophage inhibitory cytokine-1 (MIC-1) frequently occurs during the progression of prostate cancer (PC) to androgen-independent (AI) and metastatic disease states and is associated with a poor outcome of patients.
METHODS: The gain- and loss-of-function analyses of MIC-1 were performed to establish its implications for aggressive and chemoresistant phenotypes of metastatic and AI PC cells and the benefit of its downregulation for reversing docetaxel resistance.
RESULTS: The results have indicated that an enhanced level of secreted MIC-1 protein in PC3 cells is associated with their acquisition of epithelial-mesenchymal transition features and higher invasive capacity and docetaxel resistance. Importantly, …

The First Mission Of The Twelve Apostles: 1835, Maclane Elon Heward
Theses and Dissertations
The Quorum of the Twelve Apostles for the Church of Jesus Christ of Latter-day Saints is an administrative and ecclesiastical quorum. The Church, first organized in 1830, did not organize the Quorum of Twelve Apostles until 1835. When it was organized, Joseph Smith outlined the quorum's responsibilities through revelation. The Twelve were assigned two unique and specific responsibilities: to take the gospel to the nations of the earth and to form a traveling high council for the regulating of the Church outside of its stakes. The first opportunity for the Twelve to fulfill their responsibilities was in May 1835 when …
Protein-Surface Interactions With Coarse-Grain Simulation Methods, Shuai Wei
Protein-Surface Interactions With Coarse-Grain Simulation Methods, Shuai Wei
Theses and Dissertations
The interaction of proteins with surfaces is a major process involved in protein microarrays. Understanding protein-surface interactions is key to improving the performance of protein microarrays, but current understanding of the behavior of proteins on surfaces is lacking. Prevailing theories on the subject, which suggest that proteins should be stabilized when tethered to surfaces, do not explain the experimentally observed fact that proteins are often denatured on surfaces. This document outlines several studies done to develop a model which is capable of predicting the stabilization and destabilization of proteins tethered to surfaces. As the start point of the research, part …

Rock Creek Alliance V. U.S. Forest Service, Matt Pugh
Public Land & Resources Law Review
A coalition of environmental advocacy groups challenged the U.S. Forest Service, U.S. Fish and Wildlife Service, and the U.S. Department of Agriculture‟s approval of a mining project near Noxon, Montana. The proposed mine would operate partially on national forest land and involve tunneling beneath the Cabinet Mountains Wilderness to extract copper and silver. The plaintiffs challenged the mine approval by advancing claims under the Endangered Species Act, National Environmental Policy Act, Clean Water Act, Organic Administration Act, and the National Forest Management Act. The court entered summary judgment for the defendants on all but two counts. Minor changes must be …
