Oxidation Of Thrombomodulin Methionine 388 In Cigarette Smokers, Samrat B. Thapa May 2013

Oxidation Of Thrombomodulin Methionine 388 In Cigarette Smokers, Samrat B. Thapa

Graduate Theses and Dissertations

This work tested the hypothesis that oxidation of methionine 388 in thrombomodulin is higher in cigarette smokers, and thus a likely contributor towards the hypercoagulable state in smokers. Thrombomodulin, a protein cofactor found on endothelial cell surfaces, regulates the activity of thrombin. Thrombin bound to thrombomodulin no longer converts fibrinogen to fibrin, but instead activates Protein C which, in turn, stops the coagulation cascade by inactivation of clotting factors. The oxidation of methionine 388 of thrombomodulin has been shown in vitro to dramatically decrease the anticoagulant cofactor activity of thrombomodulin. The blood of cigarette smokers is more prone to clot …

Times Are A Changin': Gender Role Orientation And Alcohol Use, Jessica Lauren Fugitt May 2013

Times Are A Changin': Gender Role Orientation And Alcohol Use, Jessica Lauren Fugitt

Graduate Theses and Dissertations

Alcohol misuse is an important public health concern as it is related to an elevated probability of experiencing a number of negative life events. Historically, alcohol consumption patterns seemed to differ according to sex, but more recent research suggests that the difference has narrowed as gender role orientations have converged. Further, it seems that gender role orientation more reliably predicts differences in patterns of use than biological sex. The goal of this two-part study was to investigate the relation of gender role orientation and alcohol use behaviors with the expectation that masculine gender role orientation would be associated with these …

Dynamics And Model Of The Pore-Forming Protein Lysenin, Eric Krueger May 2013

Dynamics And Model Of The Pore-Forming Protein Lysenin, Eric Krueger

Graduate Theses and Dissertations

Membrane transporters are a class of membrane proteins that function to provide a pathway across a cell membrane for the movement of ions and biomolecules. Investigations into the regulatory mechanism of these systems are hindered by their extensive preparation requirements compounded by their fragility and instability. However, lysenin, a pore-forming protein extracted from the earthworm Eisenia foetida, provided a unique opportunity to study a protein which is stable in both a soluble and membrane phase. Lysenin channels possess several important properties characteristic of ion channels without the inherent difficulties that plague investigations with biologically vital membrane transporters like voltage-gated ion …

Design And Fabrication Of Nanofluidic Systems With Integrated Sensing Electrodes For Rapid Biomolecule Characterization, Taylor Bradley Busch May 2013

Design And Fabrication Of Nanofluidic Systems With Integrated Sensing Electrodes For Rapid Biomolecule Characterization, Taylor Bradley Busch

Graduate Theses and Dissertations

A transparent nanofluidic system with embedded sensing electrodes was designed and fabricated by integrating Atomic Force Microscopy (AFM) nanolithography, Focused Ion Beam (FIB) milling and metal deposition, and standard microfabrication processing. The fabrication process started with the evaporation of chrome/gold (Cr/Au) onto a Pyrex 7740 wafer followed by photolithography and wet etching of the microchannels. The wafer was patterned a second time to form Au microelectrodes with 15-45 micrometer separation gaps in the nanochannel region. Sensing electrodes (up to one micron wide) were then deposited using FIB to bridge the gaps. The nanochannels were realized through both AFM nanolithography and …

Interactions Between Ions And Lysenin Nanochannels And Their Potential Applications As Biosensors, Radwan Awwad Al Faouri May 2013

Interactions Between Ions And Lysenin Nanochannels And Their Potential Applications As Biosensors, Radwan Awwad Al Faouri

Graduate Theses and Dissertations

Lysenin is classified as a pore-forming toxin protein that is isolated from the earthworm Eisenia fetida and consists of 297 amino acids [1]. Lysenin inserts large conducting pores (3.0-4.7 nm in diameter) into artificial membranes (BLM) which include sphingomyelin. These pores (channels) are open and oriented upon insertion into the bilayer lipid membrane. Lysenin channels gate at positive voltages (voltage-induced gating), but not at negative voltages. Lysenin pores also exhibit activity modulation in response to changes in ionic strength and pH, indicating that electrostatic interaction is responsible for Lysenin conductance activities. In this line of inquiries, and by modulating Lysenin …

The proposed regulatory modifications will require processors to revalidate HACCP plan controls to achieve either a 6.5 or a 7.0 log10 inactivation of the …

Design And Fundamental Understanding Of Minimum Quantity Lubrication (Mql) Assisted Grinding Using Advanced Nanolubricants, Parash Kalita May 2013

Design And Fundamental Understanding Of Minimum Quantity Lubrication (Mql) Assisted Grinding Using Advanced Nanolubricants, Parash Kalita

Graduate Theses and Dissertations

Abrasive grinding is widely used across manufacturing industry for finishing parts and components requiring smooth superficial textures and precise dimensional tolerances and accuracy. Unlike any other machining operations, the complex thermo-mechanical processes during grinding produce excessive friction-induced energy consumption, heat, and intense contact seizures. Lubrication and cooling from grinding fluids is crucial in minimizing the deleterious effects of friction and heat to maximize the output part quality and process efficiency. The conventional flood grinding approach of an uneconomical application of large quantities of chemically active fluids has been found ineffective to provide sufficient lubrication and produces waste streams and pollutants …

Improvements In Microdialysis Sampling Extraction Efficiency Using Recycled Flow, Justin Cole Deaton May 2013

Improvements In Microdialysis Sampling Extraction Efficiency Using Recycled Flow, Justin Cole Deaton

Graduate Theses and Dissertations

Incomplete recovery during microdialysis sampling is hindering important research in neurology, proteomics, and immunology. Although the current generalized solution, decreasing volumetric flow rates (Q), has been and will remain to be a useful strategy it has reached it's a physical limitation due to evaporation at the collection site. Consequently, many important signaling molecules, such as signaling proteins, remain difficult to study. It is more fundamental to consider relative recovery as a function of the interaction time between the perfusate and the environment surrounding the probe.

In this work an increase in relative recovery was predicted by a mathematical model. Using …

What Is Woman? : Gentlemen Prefer Blondes, Ladies Almanack, And Woman's Search For Her Identity In The 1920s, Timothy Coyne May 2013

What Is Woman? : Gentlemen Prefer Blondes, Ladies Almanack, And Woman's Search For Her Identity In The 1920s, Timothy Coyne

Theses, Dissertations and Culminating Projects

In the introductory chapter of her book, The Gender o f Modernity, Rita Felski writes, “If our sense of the past is inevitably shaped by the explanatory logic of narrative, then the stories that we create in turn reveal the inescapable presence and power of gender symbolism” (1). Anita Loos’s Gentlemen Prefer Blondes and Djuna Barnes’s Ladies Almanack are two such stories; however, they both do more than simply reveal gender’s presence and power. These works of literature question the gender ideologies of the early twentieth century, challenging their power and inescapability by producing other, perhaps unknown, unthought of or …

Fe(Vi) As A New Chemical Agent For Treatment Of Landfill Leachate, Christopher Gravesen

Theses, Dissertations and Culminating Projects

More than 50% municipal solid waste (MSW) is being disposed of within landfills in the United States. Despite many benefits, landfills inevitably produce highly contaminated wastewater - landfill leachate. Landfill leachate represents one of the most challengeable wastewaters in the world. Refractory organic matters and ammonia nitrogen are two major persistent traditional contaminants in landfill leachate. The contaminants ought to be properly removed prior to discharge into natural water resources.

Ferrate (FeO42-) is a potential water treatment chemical agent in which iron is in its + 6 oxidation state. In engineering practice, Fe(VI) can concurrently function as …

Modeling Amyloid-Ss Self-Assembly : Stability Of On-Pathway Aggregate Formation, Andrew Kevin Mauro May 2013

Modeling Amyloid-Ss Self-Assembly : Stability Of On-Pathway Aggregate Formation, Andrew Kevin Mauro

Theses, Dissertations and Culminating Projects

Protein misfolding and concomitant aggregation towards amyloid formation is the underlying biochemical commonality among a wide range of human pathologies. Amyloid formation involves the conversion of proteins from their native monomeric states (intrinsically disordered or globular) to well-organized, fibrillar aggregates in a nucleation-dependent manner. Understanding the mechanism of aggregation is important not only to gain better insight into amyloid pathology but also to simulate and predict molecular pathways. One of the main impediments in doing so is the highly stochastic nature of interactions that complicates the development of meaningful insights. In this study, we have utilized a well-characterized intermediate along …

Evaluation Of Different Methods For Detection And Enumeration Of Indicator Organisms In Water Bodies Of Northern New Jersey, Alexandra Peri May 2013

Evaluation Of Different Methods For Detection And Enumeration Of Indicator Organisms In Water Bodies Of Northern New Jersey, Alexandra Peri

Theses, Dissertations and Culminating Projects

Bodies of water, particularly those used for drinking or recreation, are monitored for many different types of pollution. One type that causes especial concern is fecal contamination from humans and other mammals. Bodies of water affected by fecal contamination may harbor bacteria and viruses that can threaten human health. Identifying the source of such microbes is the first step towards eliminating the contamination, and making the body of water usable again. Fecal pollution is typically identified through the detection of indicator organisms, which are present in feces in large numbers. Examples of such organisms include total coliforms, and subsets of …

The Transformative Power Of Voice In George Bernard Shaw's Pygmalion, Nicole L. Scimone May 2013

The Transformative Power Of Voice In George Bernard Shaw's Pygmalion, Nicole L. Scimone

Theses, Dissertations and Culminating Projects

George Bernard Shaw’s Pygmalion is first and foremost a play about voice, particularly about the voice of flower-girl-tumed-lady Liza Doolittle. Though the voice is not Liza’s true self, it is the way the Liza’s identity can be expressed, and thus an important marker of identity transformations in the play. This work explores three different ways in which Shaw discusses voice in the play: as singing instruction, scientific methods for recording voice, and vocalizing automata and dolls.

First, the play is deeply influenced by Shaw’s background in singing instruction from his childhood. Shaw learned voice study from his mother’s beau, a …

Spectroscopic Investigation Of The Interaction Of Fadh In Dna Photolyase With Uv-Damaged Dna, Kyle Luke Williams

Theses, Dissertations and Culminating Projects

Skin cancer is the most prevalent malignancy found in humans, with the diagnosis rate continuing to steadily increase. The primary cause of this disease is overexposure to harmful ultraviolet (UV) radiation from sunlight, which can induce damage to the nitrogenous bases in DNA via dimerization. The most prevalent UV-induced photoproducts in DNA are cyclobutane pyrimidine dimers (CPDs), most commonly between adjacent thymidines.

Organisms have implemented methods by which to repair these CPDs, the primary of which are nucleotide excision repair (NER) and photoreactivation by photolyases. Photolyases are blue-light activated flavoproteins that are more efficient at recognizing and repairing CPDs than …

Personal Protective Equipment And Laboratory Safety Training: The Roles Of Attitude, Subjective Norm, And Perceived Control, Ami A. Ruffing

Dissertations

Chemical and biological agents can cause serious adverse occupational health effects, and can adversely impact environmental health. Adverse incidents occur in laboratories using chemical, biological and radiologic agents, and laboratories pose a number of dangers to workers. Adverse incidents occur more frequently in teaching and research institutions when compared to industrial laboratories. Good laboratory safety practices, including the use of personal protective equipment, can reduce the number and severity of laboratory accidents, thus reducing the risk of chemical, biological and radiologic exposure for workers and for the public. Improving laboratory safety training should also result in fewer lab accidents. This …

Climate Change, Shifts In Tropical Storm Regimes And Triadica Sebifera Invasion In Coastal Mississippi, United States, Shishir Paudel May 2013

Climate Change, Shifts In Tropical Storm Regimes And Triadica Sebifera Invasion In Coastal Mississippi, United States, Shishir Paudel

Dissertations

Global climate change is predicted to affect timing and severity of disturbance events (e.g., fire, drought, hurricanes, wind storms, and inundation), but the extent of these disturbance events and their impacts on natural ecosystems may vary regionally. Rising sea level, increased frequency and intensity of tropical storms, and altered inundation regimes are likely to create changing environmental conditions in low-lying coastal ecosystems. These large scale disturbances may increase resource availability and regeneration spaces, reduce competition, and possibly increase community vulnerability to invasion. Shifting disturbance regimes and invasion together are predicted to drive long-term shifts in coastal plant community structure and …

Exploring Signals Of Historical Demography In Boreal Mammals Through Integration Of Statistical Conservation Phylogenetics, Taxonomy, And Comparative Phylogeography, Jason L. Malaney

Biology ETDs

Understanding how diversity is partitioned across the landscape can provide perspectives related to the environmental processes that have influenced the evolutionary history of organisms. This main idea, often termed phylogeography, serves as the backdrop to my research where I explore three broad concepts including historical biogeography, cryptic diversity and ecology, and conservation phylogenetics. I address various questions in each of these concepts by using a set of mammals that are associated with montane and mesic environments of North America. More specifically, I focus on the jumping mice (Zapodidae) to test hypotheses that scale to the broader community. This approach allows …

Regulation Of Hydraulic Function And Consequences For Drought-Related Mortality In Mature Woody Plants, Jennifer A. Plaut May 2013

Regulation Of Hydraulic Function And Consequences For Drought-Related Mortality In Mature Woody Plants, Jennifer A. Plaut

Biology ETDs

Precipitation and temperature patterns are expected to shift over at least the next century due to anthropogenic climate change. Effects of these shifts may be felt most acutely in arid and semi-arid regions where water is already the most limiting resource. Vegetation mortality due to drought is a likely outcome of predicted future climate, with consequences for ecosystem structure and function and feedbacks on regional and global climate. Yet, mechanisms and interactions of key plant drought responses and the end-point of mortality remain poorly understood. In this dissertation I examine the roles of xylem cavitation resistance and stomatal control of …
