The Retina-Prosthesis Interface In Two Rat Models Of Retinitis Pigmentosa And Functional Changes Associated With Photoreceptor Loss., James William Fransen Aug 2013

The Retina-Prosthesis Interface In Two Rat Models Of Retinitis Pigmentosa And Functional Changes Associated With Photoreceptor Loss., James William Fransen

Electronic Theses and Dissertations

With increasingly successful clinical trials, the use of prosthetic devices to replace the function of photoreceptors has now become a viable option to treat degenerative diseases of the retina, including age-related macular degeneration (AMD) and retinitis pigmentosa (RP). Using prosthetics, patients have demonstrated the ability to read large text, recognize small objects, and navigate. While promising, this represents coarse visual function, with the highest visual acuity equivalent to 20/560, still below the legal definition of blindness, 20/200. To investigate the benefit of more sophisticated designs, I implanted two types of photovoltaic prosthetic devices in two models of RP, Tg S334ter-3 …

6-Phosphofructo-2-Kinase Inhibition Induces Autophagy As A Survival Mechanism., Alden Klarer Aug 2013

6-Phosphofructo-2-Kinase Inhibition Induces Autophagy As A Survival Mechanism., Alden Klarer

Electronic Theses and Dissertations

Altered metabolism has long been recognized as a defining characteristic of tumor cells. The increased glycolytic phenotype, an observation credited to Otto Warburg in 1956, is almost universal for cancer cells, making metabolic enzymes attractive targets for cancer therapy. However, anti-metabolic drugs, thus far, have not lived up to expectations as stand-alone agents to treat this disease. Unlike glycolytic enzymes that directly catabolize glucose to pyruvate, the family of 6-phosphofructo-2-kinase/fructose-2,6-bisphosphatases (PFKFBs) control the conversion of fructose-6-phosphate to and from fructose-2,6-bisphosphate, a key regulator of the glycolytic enzyme phosphofructokinase-1 (PFK-1). One isoform, PFKFB3, has been shown to be highly expressed by …

Enhanced Penetration Of Lipid Functionalized Nanoparticles In 3d Cell Cultures., Christopher Gene England 1988- Aug 2013

Enhanced Penetration Of Lipid Functionalized Nanoparticles In 3d Cell Cultures., Christopher Gene England 1988-

Electronic Theses and Dissertations

Nanoparticle-based systems can alleviate systemic toxicity via surface functionalization to promote tissue-specific targeting as well as passive targeting abilities [1], such as the enhanced permeation and retention effect (EPR) [2]. The transport of nanoparticles is limited in hypoxic tumor regions due to the typically impaired tumor vasculature. To enhance diffusion of nanotherapeutics within tumor tissue; functionalized citrate gold nanoparticles and silica gold nanoshells with surface modifications of phosphatidylcholine (PC) and high density lipoprotein (HDL) were synthesized. 3D cell cultures were used as a representative model of portions of hypoxic tissue in liver, lung, and pancreatic solid tumors. Our results indicate …

Behavioral Rhythm, Sleep, And Mood Disturbances In Newly Admitted Nursing Home Residents., Irene M. Kostiwa

Electronic Theses and Dissertations

Older adults entering a nursing home face many physical and psychosocial challenges, all at a point in life when their bodies may be least equipped to handle such adversity. Data for the present study were collected for 186 incoming residents from 6 nursing homes over the course of 1 year. Fifty-six participants were interviewed at the earliest possible time point following admission and rest-activity data were collected via actigraphy for 1 week, after which participants were interviewed a second time. Newly admitted residents in this sample were functionally dependent, highly medicated, and diagnosed with many conditions associated with sleep and/or …

Cytochrome B5 Reductase Encoded By Cbr1 Is Essential For A Functional Male Gametophyte In Arabidopsis, Laura L. Wayne, James G. Wallis, Rajesh Kumar, Jennifer E. Markham, John Browse Aug 2013

Cytochrome B5 Reductase Encoded By Cbr1 Is Essential For A Functional Male Gametophyte In Arabidopsis, Laura L. Wayne, James G. Wallis, Rajesh Kumar, Jennifer E. Markham, John Browse

Department of Biochemistry: Faculty Publications

In all eukaryotes, NADH:cytochrome b5 reductase provides electrons, via cytochrome b5, for a range of biochemical reactions in cellular metabolism, including for fatty acid desaturation in the endoplasmic reticulum. Studies in mammals, yeast, and in vitro plant systems have shown that cytochrome b5 can, at least in some circumstances, also accept electrons from NADPH:cytochrome P450 reductase, potentially allowing for redundancy in reductase function. Here, we report characterization of three T-DNA insertional mutants of the gene encoding cytochrome b5 reductase in Arabidopsis thaliana, CBR1. The progeny of plants heterozygous for the cbr1-2 allele segregated 6% homozygous mutants, while cbr1-3 and cbr1-4 …

Preventative Behavioral Parent Training In A Primary Care Context: Initial Evaluation Of A Universal Prevention Program For Disruptive Behavior Disorders, Jessica L. Malmberg Aug 2013

Preventative Behavioral Parent Training In A Primary Care Context: Initial Evaluation Of A Universal Prevention Program For Disruptive Behavior Disorders, Jessica L. Malmberg

All Graduate Theses and Dissertations, Spring 1920 to Summer 2023

One of the biggest challenges parents face is effectively managing their child’s engagement in various disruptive behaviors including noncompliance, tantrums, and aggression. Typically when children begin exhibiting disruptive behaviors, parents will express their concerns to their pediatricians; however, there are significant barriers to parents gaining adequate guidance due to clinic time constraints, insurance reimbursement issues, and the limited training pediatricians receive in addressing these concerns. As such, children are generally referred to outside mental health clinics where additional barriers arise including waitlist delays and mental health stigmatization. The treatment for conduct problems (CP) that has proven most effective is referred …

Running Among Thorns : Perspectives On Ethiopian/United States Educational Experiences., Bruce Whearty 1952- Aug 2013

Running Among Thorns : Perspectives On Ethiopian/United States Educational Experiences., Bruce Whearty 1952-

Electronic Theses and Dissertations

This dissertation is a qualitative exploration of the perspectives of Ethiopian immigrants to the United States who have educational experience in both cultures. By interviewing five respondents and asking them to reflect on the role that their education played in their acculturation to the United States, a series of case histories was developed. The case histories were analyzed using the Differentiated Multi-Dimensional Model of Acculturation to determine if they upheld the two predictions made by this model: 1) acculturation for any particular individual proceeds independently at different rates across three different dimensions: language, behavior, and cultural identity; and 2) acculturation …

Preclinical Safety Assessment Of Griffithsin-Based Vaginal Microbicides., Amanda Lasnik Aug 2013

Preclinical Safety Assessment Of Griffithsin-Based Vaginal Microbicides., Amanda Lasnik

Electronic Theses and Dissertations

Griffithsin (GRFT) is a protein derived from the red alga Griffithsia sp. off the coast of New Zealand. It binds and inactivates number of enveloped viruses including HIV-1. For a product to be developed as a microbicide, it must be safe and effective after repeated use. For initial in vivo safety testing we exposed mice repeatedly to various vaginal products and found that increased concentration of mouse serum albumin (MSA) in the vaginal lumen is predictive of epithelial damage. Repeated intravaginal administration of GRFT did not result in increased amounts of MSA, but the known epithelial disrupters nonoxynol-9 and benzylkonium …

Mitigating The Problem Of Vulture Holdout: International Certification Boards For Sovereign Debt Restructurings, John A. E. Pottow Aug 2013

Mitigating The Problem Of Vulture Holdout: International Certification Boards For Sovereign Debt Restructurings, John A. E. Pottow

Law & Economics Working Papers

The Great Recession has brought greater sovereign debt defaults, which in turn has brought a surfeit of academic explorations and policy discussions of sovereign debt restructuring. The purpose of this article is to offer yet one more idea for the hopper of what to do with the seemingly intractable problem of restructuring sovereign bond debt. The field does not lack for statutory and contractual proposals, from SDRM to CACs, but it is not yet sufficiently saturated that another proposal cannot join the mix. The proposal is for the establishment of international certification boards that can give a stamp of approval …

Expression Of C-Fos In Hilar Mossy Cells Of The Dentate Gyrus In Vivo., Aine M Duffy, Michael J Schaner, Jeannie Chin, Helen E Scharfman Aug 2013

Expression Of C-Fos In Hilar Mossy Cells Of The Dentate Gyrus In Vivo., Aine M Duffy, Michael J Schaner, Jeannie Chin, Helen E Scharfman

Department of Neuroscience Faculty Papers

Granule cells (GCs) of the dentate gyrus (DG) are considered to be quiescent--they rarely fire action potentials. In contrast, the other glutamatergic cell type in the DG, hilar mossy cells (MCs) often have a high level of spontaneous activity based on recordings in hippocampal slices. MCs project to GCs, so activity in MCs could play an important role in activating GCs. Therefore, we investigated whether MCs were active under basal conditions in vivo, using the immediate early gene c-fos as a tool. We hypothesized that MCs would exhibit c-fos expression even if rats were examined randomly, under normal housing conditions. …

The study assumed a contextual approach and focused on different class contexts and students’ literacy practices. The class, which took place in a resource-rich, multimodal environment, was perceived by students as a positive learning experience. The distribution of resources and their role in teaching and learning varied and depended on the …

Vascular Reactivity Of Immature Arterialized Capillaries, Sara Hellstrom Aug 2013

Vascular Reactivity Of Immature Arterialized Capillaries, Sara Hellstrom

Biomedical Engineering

Peripheral arterial occlusive disease (PAOD) is a globally prevalent cardiovascular disease in which atherosclerotic plaques narrow arterial lumen diameters and restrict blood flow to down stream tissues. The impact of these occlusions can be mitigated by collateral vessels that connect parallel arterial branches and act as natural bypasses to maintain perfusion. Some patients with PAOD may not have robust collateral networks to accommodate ischemic tissues in the event of an occlusion and, therefore, may be more susceptible to hypoxia and tissue necrosis. In animal models that lack collateral arterioles, capillaries can arterialize and form functional collaterals; however, in the early …

Genome-Wide Prediction Of Age At Puberty And Reproductive Longevity In Sows, Julie Kathleen Tart, Rodger K. Johnson, Justin W. Bundy, N. N. Ferdinand, A. M. Mcknite, Jennifer R. Wood, Phillip S. Miller, M. F. Rothschild, Matthew L. Spangler, Dorian J. Garrick, Stephen D. Kachman, Daniel C. Ciobanu

Department of Animal Science: Faculty Publications

Traditional selection for sow reproductive longevity is ineffective due to low heritability and late expression of the trait. Incorporation of DNA markers into selection programs is potentially a more practical approach for improving sow lifetime productivity. Using a resource population of crossbred gilts, we explored pleiotropic sources of variation that influence age at puberty and reproductive longevity. Of the traits recorded before breeding, only age at puberty significantly affected the probability that females would produce a first parity litter. The genetic variance explained by 1-Mb windows of the sow genome, compared across traits, uncovered regions that influence both age at …

An Evaluation Of The Nontarget Effects Of Transgenic Bacillus Thuringiensis Maize On Arbuscular Mycorrhizal Fungi In The Soil Ecosystem, Tanya Elizabeth Amy Cheeke Aug 2013

An Evaluation Of The Nontarget Effects Of Transgenic Bacillus Thuringiensis Maize On Arbuscular Mycorrhizal Fungi In The Soil Ecosystem, Tanya Elizabeth Amy Cheeke

Dissertations and Theses

My dissertation research examined the effect of the cultivation of insect-resistant Bacillus thuringiensis (Bt) maize on the soil environment with a goal of understanding how to obtain a balance between technological advancement and maintenance of a healthy soil ecosystem. Although Bt plants may help to reduce pesticide use, conferring benefits to farm workers and the environment, there are still unresolved questions about how the cultivation of Bt plants affects soil organisms. For this dissertation project, I used 14 different genotypes of Bt maize and non-Bt maize (Zea mays) to investigate the effects of transgenic Bt plants on the colonization ability, …

The Effects Of Variation In Collagen D-Spacing On Compact Bone Viscoelasticity: A Finite Element Analysis, Miguel A. Mendoza Aug 2013

The Effects Of Variation In Collagen D-Spacing On Compact Bone Viscoelasticity: A Finite Element Analysis, Miguel A. Mendoza

Master's Theses

The D-spacing that is characteristic of collagen and its structural arrangement was previously thought to be a constant value. Much research is revealing that it is actually a distribution of values in biological tissues. Recent ovine experimentation has also shown that the D-spacing distribution is significantly altered following estrogen depletion. While ewes contain some major biological differences between their human counterparts, they are an economical and robust large animal model for postmenopausal osteoporosis. So, the exploration of the possible implications that D-spacing has on the mechanical properties of the whole bone utilizing animal models and computational methods is warranted. Six …

The Role Of Gnrh-Ii And Its Receptor In Testicular Function, Amy T. Desaulniers Aug 2013

The Role Of Gnrh-Ii And Its Receptor In Testicular Function, Amy T. Desaulniers

Department of Animal Science: Dissertations, Theses, and Student Research

The second mammalian isoform of GnRH (GnRH-II) has been linked to regulation of cell proliferation, feed intake, and the interaction between energy balance and reproductive behavior. In contrast to the native form of GnRH (GnRH-I), GnRH-II is an inefficient modulator of gonadotropin secretion. Unlike many species, a functional receptor (GnRHR-II) specific to this ligand has been discovered in the pig that may be directly involved in testosterone production. Therefore, our objective was to identify the role of GnRH-II and its receptor in testicular function. First, there was 6-fold more GnRHR-II protein in the testis than anterior pituitary gland of boars. …

Empathy And Threatened Egotism In Men’S Use Of Violence In Intimate Relationships, Jessica H. Turner Aug 2013

Empathy And Threatened Egotism In Men’S Use Of Violence In Intimate Relationships, Jessica H. Turner

Electronic Theses and Dissertations

The current study was undertaken to explore the relationship between self-esteem, narcissism, and empathy with intimate partner violence perpetration among men in 2 samples: college students and inmates. The sample was analyzed both as an aggregate and separately. A negative relationship was hypothesized between intimate violence perpetration and both self-esteem and empathy. A positive relationship was expected between intimate violence perpetration and narcissism. A 2-way interaction was examined between self-esteem and narcissism as a test of threatened egotism, defined as high self-esteem coupled with high narcissism, which was not expected in the current study. Empathy was hypothesized to moderate the …
