Swine play an important role in the disease ecology of influenza. Having cellular receptors in common with birds and humans, swine provide opportunities for mixed infections and potential for genetic re-assortment between avian, human, and porcine influenza. Feral swine populations are rapidly expanding in both numbers and range and are increasingly coming into contact with waterfowl, humans, and agricultural operations. In this study, over 875 feral swine were sampled from six states across the United States for serologic evidence of exposure to influenza. In Oklahoma, Florida, and Missouri, USA, no seropositive feral swine were detected. Seropositive swine were detected in …

Genome-wide studies have strongly associated a non-synonymous polymorphism (rs16969968) that changes the 398th amino acid in the nAChR α5 subunit from aspartic acid to asparagine (D398N), with greater risk for increased nicotine consumption. We have used a pentameric concatemer approach to express defined and consistent populations of α3β4α5 nAChR in Xenopus oocytes. α5(Asn-398; risk) variant incorporation reduces ACh-evoked function compared with inclusion of the common α5(Asp-398) variant without altering agonist or antagonist potencies. Unlinked α3, β4, and α5 subunits assemble to form a uniform nAChR population with pharmacological properties matching those of concatemeric α3β4* nAChRs. α5 subunit incorporation reduces α3β4* …

RGS-containing RhoGEFs (RGS-RhoGEFs) represent a direct link between the G(12) class of heterotrimeric G proteins and the monomeric GTPases. In addition to the canonical Dbl homology (DH) and pleckstrin homology domains that carry out the guanine nucleotide exchange factor (GEF) activity toward RhoA, these RhoGEFs also possess RGS homology (RH) domains that interact with activated α subunits of G(12) and G(13). Although the GEF activity of p115-RhoGEF (p115), an RGS-RhoGEF, can be stimulated by Gα(13), the exact mechanism of the stimulation has remained unclear. We present high-resolution, near-infrared NIRSPEC observations of CO absorption toward six class II T Tauri stars: AA Tau, DG Tau, IQ Tau, RY Tau, CW Tau, and Haro 6–5b. 12CO overtone absorption lines originating from the circumstellar disk of each object were used to calculate line-of-sight gas column densities toward each source. We measured the gas/dust ratio as a function of disk inclination, utilizing measured visual extinctions and inclinations for each star. The majority of our sources show further evidence for a correlation between the gas/dust columndensity ratio and disk inclination similar to that found by Rettig et al.

Background
Soluble guanylyl cyclase (sGC) plays a central role in nitric oxide (NO)-mediated signal transduction in the cardiovascular, nervous and gastrointestinal systems. Alternative RNA splicing has emerged as a potential mechanism to modulate sGC expression and activity. C-α1 sGC is an alternative splice form that is resistant to oxidation-induced protein degradation and demonstrates preferential subcellular distribution to the oxidized environment of endoplasmic reticulum (ER).
Methodology/Principal Findings
Here we report that splicing of C-α1 sGC can be modulated by H2O2 treatment in BE2 neuroblastoma and MDA-MD-468 adenocarcinoma human cells. In addition, we show that the H2O2 treatment of MDA-MD-468 cells selectively …

Cancer stem cells (CSCs) may be responsible for tumor recurrence. Metabolic labelling of newly synthesized proteins with non-canonical amino acids allows us to discriminate CSCs in mixed populations due to the quiescent nature of these cells.

Graphene and graphene oxide (GO) are two-dimensional nanomaterials with promising biomedical application potential, including drug and gene delivery, bio-imaging, and photothermal therapy. However, the nature of interactions between these materials and cells is poorly understood. Here, cell surface adhesion, subcellular localization, and size-dependent uptake are investigated in C2C12 cells using bovine serum albumin (BSA)-functionalized GO. Small BSA-GO nanosheets enter cells through clathrin-mediated endocytosis (CME), while large nanosheets enter cells through both CME and phagocytosis. Cytotoxicity is minimal. AFM-based size/thickness characterization of BSA-GO is performed in a systematic fashion, and a practical protocol for such analysis is presented. Size-dependent uptake results …

In this study, we examined consistency of infant race/ethnicity across two data sources (N=2,63) using measures of sensitivity and positive predictive value. First we created and compared conventional measures of infant race/ethnicity from 2007 Oklahoma birth certificates and SEED for Oklahoma Kids baseline survey data, classifying infants as White, African American, American Indian, Asian, or Hispanic. Then we created and tested alternative measures with a biracial classification, based on biological parentage from birth certificates or parent report of infant biracial identity in the survey. Agriculture is an economically important form of landuse in the North American Great Plains. Since 19th Century European settlement, conversion of grasslands to rowcrops has increased food and bioenergy production, but has decreased wildlife habitat. Future agricultural landuse changes may be driven by alternative energy demands and regional climatic changes. Landuse change and its drivers could affect bioenergy production, wildlife populations and natural resources, and considering the potential impacts of impending changes in advance could assist with preparations for an uncertain future.

Despite a worldwide push for the conversion of conventional agriculture to organic agriculture, organic farming itself is not a very popular vocational choice for young people. Brattleboro, one of the towns in Vermont, is no exception. Although several local organizations are promoting local agriculture and food security, in reality, not many young people in Brattleboro want to choose organic agriculture as their career.
With a total of 17 in-depth qualitative interviews with young adults in the Brattleboro area, this inquiry sought to identify a range of factors that would motivate them to choose organic agriculture as their career. The results …

This thesis aims at the evaluation of the best amide bonding strategy that places a fluorescence aminomethyl coumarin tag onto a LMW organic acid in aqueous solution. A three-stage study was conducted. First, a suitable aminomethyl coumarin fluorophore was synthesized. Second, a library of acid-derivatized standards was synthesized for use in optimizing the anticipated HPLC separation of a mixture of LMW acids expected to be found in marine samples. Otoacoustic Emissions (OAEs) are sounds generated by an active process in the auditory system’s cochlea. It has been widely accepted that the generation of OAEs is a precursor for healthy hearing. The measurement of evoked OAEs can be used to determine the general health of the cochlea and basilar membrane's response and sound transmission forward and backwards through the inner ear. OAEs are commonly used for newborn infant hearing screening where many middle ear pathologies are first detected. In most cases, secondary screening tests such as tympanometry are not conducted unless the patient has failed the OAE screening first. Increases …

The purpose of this study was to determine the reliability of measuring attachment through subjective measures, as well as the inter-rater reliability of those using the subjective measures. Participants were volunteers who had worked with the Division of Youth and Family Services (DYFS). They included social workers, lawyers, psychologists, etc. Participants were asked to answer questionnaires regarding the DYFS children they had worked with, as well as demographics about themselves. The questionnaires also included an alternative informed consent. The surveys were mailed to participants and included return envelopes to ensure ease of mailing them back.
