Long-Term Survival Of Anoxia Despite Rapid Atp Decline In Embryos Of The Annual Killifish Austrofundulus Limnaeus., Jason Podrabsky, Michael Menze, Steven Hand
Long-Term Survival Of Anoxia Despite Rapid Atp Decline In Embryos Of The Annual Killifish Austrofundulus Limnaeus., Jason Podrabsky, Michael Menze, Steven Hand
Faculty and Staff Scholarship
Embryos of the annual killifish Austrofundulus limnaeus can survive for months in the complete absence of oxygen. Survival of anoxia is associated with entry into a state of metabolic dormancy known as diapause. However, extreme tolerance of anoxia is retained for several days of post-diapause development. Rates of heat dissipation in diapause II and 4 days post-diapause II embryos were measured under aerobic conditions and during the transition into anoxia. Phosphorylated adenylate compounds were quantified in embryos during entry into anoxia and after 12 hr of aerobic recovery. Rates of heat dissipation were not affected by exposure to anoxia in …

Aloe Striata Plant Defense Compounds Produced In Response To Jasmonic Acid, Salicylic Acid, And Eriophyid Mite Salivary Extract Elicitor Compounds, Fritz Light
Master's Theses
Aloe mites are herbivores of the genus Aloe (ALOACEAE) and are associated with hyperplastic growth in various aloe species, but the biochemistry of this interaction is poorly understood. In an effort to characterize plant defense responses to herbivory in the genus Aloe, a salivary extract was isolated from aloe mites (Aceria aloinis Keifer) and its bioactivity was tested using a hypocotyl elongation assay. Subsequently, Aloe striata plants were treated with jasmonic acid (JA), salicylic acid (SA), and the mite salivary extract. Using water and methanol, compounds of different polarity were extracted from aloe tissues that had been frozen …

Advancing Global Tobacco Control: Exploring Worldwide Youth Attitudes And Behaviors Toward Tobacco Use And Control, Sreenivas Phanikumar Veeranki
Electronic Theses and Dissertations
Tobacco use continues to be the leading cause of preventable death in the world. The disproportionate increase in tobacco use in low- and middle-income countries needs immediate attention. Many smokers begin smoking as adolescents and are most likely to become permanent smokers. Moreover, youth are highly targeted by tobacco industry strategies. However, a gap exists in literature to understand worldwide youth tobacco use and control. The purpose of this study is to 1) identify factors that influence never-smoking youths‟ smoking susceptibility, 2) explore characteristics that influence youth exposure to environmental tobacco smoke (ETS) and 3) to delineate key determinants of …
Impact Of Hybrid Rice On Food Security: A Spatial Equilibrium Analysis Of Global Adoption And Diffusion Of Hybrid Rice Varieties, Till Ludwig
Graduate Theses and Dissertations
Rice is the staple crop for most consumers in Asia. More than half of the global population depends on rice. However, scarce resources for agricultural production and unfavorable conditions will make it hard to meet future demand in rice and threaten future food security. Hybrid rice technology is a method to increase the productivity of resources needed for rice production. Current developments show not only yield improvements in comparison to existing conventional and hybrid varieties, but also fuel hopes for new abiotic and biotic stress tolerance. The objective of this study is to determine what impact hybrid rice varieties can …

Assessing Knowledge Acquisition Through Lecture And Cooperative Learning: Traditional And Non-Traditional Secondary Students In Agriculture, Beth Ann Jean Bills-Hunt
Graduate Theses and Dissertations
Currently, students entering secondary agricultural education programs have less knowledge and hands-on experience about agriculture than previous generations. Agricultural educators are challenged to vary teaching practices to meet student needs. The purpose of this study was to determine if there is a difference in knowledge acquisition between students enrolled in secondary agricultural education classes taught by cooperative learning and lecture instruction. Additionally, this study explored the knowledge acquisition of traditional and non-traditional secondary agricultural education students and perceptions of instructional methods presented. Eight schools participated in the study. No significant difference was seen in test scores between methods of instruction, …

Mass Latex Balloon Releases And The Potential Effects On Wildlife, Stephan Irwin
All Dissertations
The first component of the study examined the fate of balloons after release to determine aspects of where, in what state, and how long they persist in the environment to assess potential risk of exposure to wildlife. Tagged balloons from sporting events gave estimates of mean distances traveled from releases. Effects on the structural integrity of the latex balloon as it reaches the upper atmosphere were also tested to determine the physical state when fragments landed. Degradation studies were conducted to determine the length of time latex can persist in the environment after exposure to various environmental conditions (sun, shade, …

All Dissertations
A taxonomic study of the crayfish species Cambarus (Hiaticambarus) longirostris was conducted. Multivariate and univariate statistical analyses of morphometric data and examination of morphological characters revealed the existence of four undescribed species from populations previously considered to belong to C. longirostris; these were located on the southern extents of the range of C. longirostris. Three of these new species were morphologically similar to the group of species containing C. coosawattae, C. chasmodactylus, C. elkensis, C. longirostris, C. longulus, and C. manningi, while one was morphologically similar to the C. fasciatus, C. …
Lignin Modification In Arabidopsis And Populus For Studies Of Gene Function And Improving Lignin Degradation, Yi Xu
All Dissertations
Lignin is one of the most abundant biopolymers in plants and plays an important role in plant structure and stress defense. Lignin is also considered to be a hallmark of vascular plants because of its crucial role in plant terrestrialization. However, lignin is an undesired component in the pulp and paper industry, bioethanol production, and forage digestibility. Thus, understanding the functions and the evolution of lignin biosynthesis genes can not only advance our knowledge of the evolution of land-adaptation for vascular plants but also help guide the effort to exploit the potential for genetic manipulation of lignin for desirable traits …

The Influence Of Flagship Species On In Situ And Ex Situ Wildlife Tourists' Connection To Wildlife And Pro-Conservation Behaviors, Jeffrey Skibins
The Influence Of Flagship Species On In Situ And Ex Situ Wildlife Tourists' Connection To Wildlife And Pro-Conservation Behaviors, Jeffrey Skibins
All Dissertations
Annually, millions of tourists visit natural areas and zoos primarily to view flagship species such as lions and elephants. Venues rely on the inherent charisma of these species to increase visitation and anchor conservation efforts. Expected visitor outcomes from the use of flagships include raised levels of awareness and pro-conservation behaviors. However, the role of flagships in wildlife tourism has been criticized for not delivering conservation benefits for species of interest or biodiversity, and producing negative site impacts. Furthermore, little is known about how the connection to a species influences conservation behaviors. This dissertation addresses this gap in knowledge by …

The first chapter examines the effect of the number of years children spend living with a single-parent family instead of a two-parent family on children's completed schooling, based on a sample of children from the PSID. To deal with the endogeneity of mothers' family structure decisions, I exploit the variation across states and over time in unilateral divorce laws, unmarried fertility ratios, welfare rules, earned income tax credit rates, and labor market conditions that generate plausibly exogenous changes in mothers' family structure choices. I construct a set of extensive measures for these contextual variables and use them as instruments to …

Ecology And Phylogeny Of The Biting-Midge Genus Culicoides (Diptera: Ceratopogonidae), Dustin Swanson
All Dissertations
Flies of the genus Culicoides are small, biting midges in the family Ceratopogonidae. These flies are of medical, veterinary, and economic importance because their blood-feeding behavior can cause stress to hosts and transmit disease agents. Despite the importance of these flies, little is known about their biology, especially the ecology of the immature stages and the phylogenetic relationships among taxa. The objectives of my study were to address these two areas of Culicoides biology.
Larval Culicoides were collected from aquatic habitats in four ecoregions of South Carolina, USA. Eleven ecological variables were recorded for each sample. Larvae were identified by …
Avian Phenology, Climate, And Land-Use Conservation Assessed At Broad Spatial And Temporal Scales Using Interdisciplinary Approaches And Citizen Science, Jason Courter
All Dissertations
Current natural resource challenges include global alteration of land cover, loss of biodiversity, impacts from increasing demands for agricultural products, and climate change. Birds are often used to assess the effects of ecological stressors because they are sensitive to environmental changes, ubiquitous and charismatic, and long-term monitoring programs have been in place for more than a century. I demonstrate the effects of climate change on avian migration and nesting dates and the potential value of using interdisciplinary approaches and citizen science to address contemporary ecological challenges. I examine Ruby-throated Hummingbird (Archilochus colubris) migration in the eastern United States from 1880-2010 …
Laboratory Characterization And Theoretical Analysis Of Contaminant Mass Transfer, Xiaoling Liu
Laboratory Characterization And Theoretical Analysis Of Contaminant Mass Transfer, Xiaoling Liu
All Dissertations
Chlorinated volatile organic compounds (CVOCs) in fractured, low permeability material can be a long-term source of groundwater contaminants and they are among the most difficult remediation challenges. Thermal methods are a possible remediation option, where heat is used to boil water and the volatile compounds are stripped out by partitioning into the resulting water vapor. Theoretical analysis indicates that only a small fraction of pore water removal may lead to a significant geometric reduction in the dissolved concentration [(Udell(, 1998]. Previous modeling studies [(Falta and Murdoch(, 2011; (Pruess(, 1983] suggest that a lower permeability limit exists for effective mass transfer …
Propagation Of Uncertainty In Light-Frame Wood Buildings, Seyed Masood Hassanzadehshirazi
Propagation Of Uncertainty In Light-Frame Wood Buildings, Seyed Masood Hassanzadehshirazi
All Dissertations
Light-frame wood construction is the most common form of construction for residential and low-rise commercial buildings (e.g. hotels and motels) in North America. In these buildings, shearwalls are the primary systems resisting lateral loads induced by earthquakes and winds. Past earthquake events have revealed that the structural performance of nominally identical light-frame wood buildings varied significantly. Although much research has been conducted investigating the uncertainty in the performance of wood shearwalls and buildings under earthquake loading, the focus of many of these studies was on the uncertainty due to the earthquake motions, referred as earthquake-to-earthquake uncertainty, the influences of variability …

Life on Sanak is remarkable—from the brilliant orange of lichens clinging to life high on wave-beaten and barren rocks to the raspy bark of an introduced arctic fox or a vole's abstract bouquet of drying tundra flowers. In this chapter we will explore a limited selection of the diverse cast of characters that inhabit the Sanak Islands, ranging from the tiny and cryptic to the large and charismatic.
The Ecological Interactions Of Sanak, Nancy Huntly, Spencer Wood, Roly Russell
The Ecological Interactions Of Sanak, Nancy Huntly, Spencer Wood, Roly Russell
Biology Faculty Publications
Ecology is not only about the interesting biology of organisms, as described for Sanak Island in the last chapter, but also about interactions: interactions between animals (including people), plants, microbes, the habitats that they live in, and the environments that surround them. The ecology of Sanak Island today reflects not only today's conditions, but also shows the imprint of the distant history of interactions of the island, especially, as we will see below, the imprint of the Aleut people that have made this island their home.

Almost everyone learns about the concept of ecological "food chains" in grade school. An example of a marine food chain is shown on the left-hand side of the chart to the left: phytoplankton are eaten by mussels, which are eaten by octopus, which are eaten by sea otters, which are eaten by humans. These feeding interactions can be abstractly represented as five "nodes" connected in a single chain by four links, as shown on the right-hand side of the diagram to the left. The food chain shown is one of the thousands of interlinked food chains that make up the …

Cytoprotective functions of a 20S proteasome activator were investigated. Saccharomyces cerevisiae Blm10 and human 20S proteasome activator 200 (PA200) are homologs. Comparative genome-wide analyses of untreated diploid cells lacking Blm10 and growing at steady state at defined growth rates revealed downregulation of numerous genes required for accurate chromosome structure, assembly and repair, and upregulation of a specific subset of genes encoding protein-folding chaperones. Blm10 loss or truncation of the Ubp3/Blm3 deubiquitinating enzyme caused massive chromosomal damage and cell death in homozygous diploids after phleomycin treatments, indicating that Blm10 and Ubp3/Blm3 function to stabilize the genome and protect against cell death. …
Impact Of Secondary Treatment Types And Sludge Handling Processes On Estrogen Concentration In Wastewater Sludge, Erica J. Marti
Impact Of Secondary Treatment Types And Sludge Handling Processes On Estrogen Concentration In Wastewater Sludge, Erica J. Marti
UNLV Theses, Dissertations, Professional Papers, and Capstones
Endocrine-disrupting compounds (EDCs), such as estrogen, are known to be present in the aquatic environment at concentrations that negatively affect fish and other wildlife. Wastewater treatment plants (WWTPs) are major contributors of EDCs into the environment. EDCs are released via effluent discharge and land application of biosolids. Estrogen removal in WWTPs has been studied in the aqueous phase; however, few researchers have determined estrogen concentration in sludge. This study focuses on estrogen concentration in wastewater sludge as a result of secondary treatment types and sludge handling processes. Grab samples were collected before and after multiple treatment steps at two WWTPs …
Examining The Role Of Family And Marital Communication In Understanding Resilience To Family-Of-Origin Adversity, Kristen Carr
Examining The Role Of Family And Marital Communication In Understanding Resilience To Family-Of-Origin Adversity, Kristen Carr
Department of Communication Studies: Dissertations, Theses, and Student Research
Resilience, or the “successful adaptation to adversity,” is often investigated as an individual response to stressful experiences (Zautra, Hall, & Murray, 2010, p. 4). After the experience of stress or adversity, most people seek to return to some degree of normalcy, but their ability to do so varies widely. To understand this variation in individuals’ responses to adversity, most researchers have focused on resilience as a process that occurs within individuals, rather than between them. However, in the current study, resilience is positioned as an interpersonal process as well as an individual one, in that people interpret and respond to …

Purification Of Fibrinogen From Human Plasma, Ayman E. Ismail
Purification Of Fibrinogen From Human Plasma, Ayman E. Ismail
Department of Chemical and Biomolecular Engineering: Dissertations, Theses, and Student Research
A solvent detergent treated fibrinogen was purified from human plasma by cryoprecipitation (cryo) followed by chemical precipitation using ethanol (EtOH) or ammonium sulfate (AS) as precipitating agents. Amounts of fibronectin (FN), factor XIII A-subunit (FXIIIA), factor XIII b-subunit (FXIIIB), and alpha2-antiplasmin (α2-AP) in the isolated fibrinogen were quantified. Thromboelastography (TEG) analysis was used to evaluate the clot strength of the isolated fibrinogen and to determine the ability of the ethanol and ammonium sulfate precipitations to eliminate the solvent detergent. Sodium dodecylsulfate-polyacrylamide gel analysis indicated that fibrinogen produced by each of these precipitation methods had similar purity. …

Effects Of Planting Date, Harvest Date, And Environmental Conditions On Germination Of Forage Kochia Accessions, Cody F. Creech
All Graduate Theses and Dissertations, Spring 1920 to Summer 2023
Forage kochia is used to re-seed areas in harsh environments that have been infested by annual weeds or disturbed by repetitive fires. It is an introduced perennial semi-shrub that is valuable to livestock and wildlife as a forage. The seed of forage kochia is very unpredictable and successful establishment in areas of low precipitation where it is utilized has often been erratic. Forage kochia seeding failures are costly and leave the land vulnerable to further degradation and erosion. This study was conducted to investigate how germination of forage kochia is affected by different harvest and planting dates, age of seed, …

An Analysis Of The Redox Properties And Stability Of Chlamydomonas Reinhardtii Cytochrome F, Cytochrome C6, And Mutants Thereof, Nicole Lynn Vanderbush
Graduate Theses and Dissertations
This body of work presents mutagenesis studies conducted on two c-type cytochromes from Chlamydomonas reinhardtii. Cytochrome f, a unique c-type cytochrome, is investigated in regards to its redox potential, the dependence of the redox potential, and the thermal stability of the protein. The mutations made were Y1F, Y9F, Y160F, Y160L, R156L, and R156K. The residues that were mutated surround the heme. It was found that, relative to the wild-type, only the Y160L and R156 mutants showed any difference in midpoint potential at pH 7. Wild-type and mutants both had a midpoint potential that was dependent upon pH indicating that none …
