The polyanionic nature of oligonucleotides and their enzymatic degradation present challenges for the use of siRNA in research and therapy; among the most notable of these is clinically relevant delivery into cells. To address this problem, we designed and synthesized the first members of a new class of guanidinium-rich amphipathic oligocarbonates that noncovalently complex, deliver, and release siRNA in cells, resulting in robust knockdown of target protein synthesis in vitro as determined using a dual-reporter system. The organocatalytic oligomerization used to synthesize these co-oligomers is step-economical and broadly tunable, affording an exceptionally quick strategy to explore chemical space for optimal …

Assessment Of The Role Of Solute Carrier Drug Transporters In The Systemic Disposition Of Fluoroquinolones: An In Vitro - In Vivo Comparison, Aditi Mulgaonkar Aug 2012

Assessment Of The Role Of Solute Carrier Drug Transporters In The Systemic Disposition Of Fluoroquinolones: An In Vitro - In Vivo Comparison, Aditi Mulgaonkar

Theses and Dissertations

Fluoroquinolones (FQ) are broad-spectrum charged antimicrobials exhibiting excellent tissue/fluid permeation. Thus, FQ disposition depends essentially on active transport and facilitative diffusion. Although most early transporter studies investigating renal elimination of FQs have focused on apical efflux of FQs from renal proximal tubule cell (RPTC) into urine, their basolateral uptake mechanism(s) from blood into RPTC (i.e., first step to tubular secretion) has not yet been explored in detail. Renally expressed SLC22 members: organic anion (OATs) and cation (OCTs) transporters are known to transport such small organic ionic substrates (molecular weight ~400 Da). Hence it is of interest to explore the role …

Obstetrics and Gynaecology, East Africa

Background: Globally, studies report a high prevalence of intimate partner sexual violence (IPSV) and an association with HIV infection. Despite the criminalisation of IPSV and deliberate sexual HIV infection in Zimbabwe, IPSV remains common. This study explored women’s and health workers’ perspectives and experiences of sexuality and sexual violence in pregnancy, including in relation to HIV testing.

Methods: This qualitative study was part of a larger study of the dynamics of intimate partner violence and HIV in pregnancy in Zimbabwe. Key informant interviews were conducted with health workers and focus group discussions were held with 64 pregnant or nursing mothers …

The Geographical Dimension Of The Development Effects Of Natural Resources, Abdur Chowdhury, Fabrizio Carmignani Aug 2012

The Geographical Dimension Of The Development Effects Of Natural Resources, Abdur Chowdhury, Fabrizio Carmignani

Economics Faculty Research and Publications

We study the contribution of natural resource intensity to long-term development along different dimensions: per-capita income, institutional quality, and education. We allow natural resources to affect these dimensions differently in different regions of the world. The evidence suggests that natural resources are generally a positive driver of development, but in Sub-Saharan Africa (SSA) their contribution is almost negligible, if not even negative. We explain these cross-regional differences with the fact that in SSA more than anywhere else large resource endowments are combined with a particularly bad disease environment. Some historical evidence and formal econometric results support this hypothesis.

Experiences With Social Services Among Homeless Lgbtq Young Adults, Fallon Kadel Aug 2012

Experiences With Social Services Among Homeless Lgbtq Young Adults, Fallon Kadel

HIM 1990-2015

My findings imply that homeless LGBTQ young adults are very limited in where they can go to find LGBTQ-accepting services. Even though all the shelters that were interviewed claimed they did not discriminate against LGBTQ persons, they do not provide and are not affiliated with services or programs that are considered to be LGBTQ- accepting. Based on the research, shelters would rather have clients conform to their pre-existing structure rather than the shelters conforming to the specific needs of the clients.; The goal of this research is to explore what programs and services are being provided at various homeless shelters …

Treatment of cancer often involves uses of multiple therapeutic strategies with different mechanisms of action. In this study we investigated combinations of nanosecond pulsed electric fields (nsPEF) with low concentrations of gemcitabine on human oral cancer cells. Cells (Cal-27) were treated with pulse parameters (20 pulses, 100 ns in duration, intensities of 10, 30 and 60 kV/cm) and then cultured in medium with 0.01 mu g/ml gemcitabine. Proliferation, apoptosis/necrosis, invasion and morphology of those cells were examined using MTT, flow cytometry, clonogenics, transwell migration and TEM assay. Results show that combination treatments of gemcitabine and nsPEFs exhibited significant synergistic activities …

Dissolute Pluralism: An Ethics Of Queer Thought, Kristin P. Mccartney Aug 2012

Dissolute Pluralism: An Ethics Of Queer Thought, Kristin P. Mccartney

College of Liberal Arts & Social Sciences Theses and Dissertations

Every practice has a theory, but multiracial queer social movements have centered theory in their praxis. In consciousness-raising and affinity groups, listservs and electronic forums, zines and community action research projects, participants frame the task of thought itself as collective and interactive, subject not just to evolving norms, but also to vicissitudes of duration, location, and timing, medium, ritual, and personality. I argue that the ephemeral character of these interventions is not incidental or regrettable, but designed to encourage organic participation and obstruct institutional codification. In turn, these values and tactics call academics to interrogate how they enter into this …

Yeast Surface Display Of Cd154, Iaryna Masniuk Aug 2012

Yeast Surface Display Of Cd154, Iaryna Masniuk

Graduate Theses and Dissertations

The CD154 (CD40L) is a member of tumor necrosis factor (TNF) family that plays a crucial role in regulation of both cell-mediated and humoral immunity by activating antigen presenting cells (APCs). Previously, studies in the laboratory demonstrated that yeast surface display system allows the expression of biologically functional antigen, such as subunit HA, against influenza virus. Yeast surface display system seems to be an excellent system to be utilized to develop anti-influenza virus vaccine. Thus, it is of interest to study the adjuvant function of CD154 on the subunit HA vaccine against influenza virus. In this project, the CD154 encoding …

Comparing Single-Sex And Reformed Coeducation: A Constitutional Analysis, Nancy Chi Cantalupo Aug 2012

Comparing Single-Sex And Reformed Coeducation: A Constitutional Analysis, Nancy Chi Cantalupo

San Diego Law Review

One of the most enduring educational debates of the past three decades has dealt with the legality and advisability of sex-segregated education. This debate can often look confusing, given a large number of debaters and the diversity of their perspectives and agendas. More than this diversity, however, the debate is confusing because the debate has been structured as a contest between the "innovation" of sex-segregated education and status quo coeducation. Missing from the debate is a comparison between reformed coeducation and a single-sex alternative, a comparison that is markedly more useful in determining what ought to be done about the …

Adolescent Identity Versus The First Amendment: Sexuality And Speech Rights In The Public Schools, Steven J. Macias Aug 2012

Adolescent Identity Versus The First Amendment: Sexuality And Speech Rights In The Public Schools, Steven J. Macias

San Diego Law Review

This Article examines the legal tensions that currently exist between public schools' attempts to protect gay students from bullying and harassment and antigay students' First Amendment rights to engage in allegedly harassing speech. First, it looks at the popular conception of the relationship between childhood or adolescence and sexuality by considering the political and legal fallout of the same-sex marriage debate. Then this Article turns to a conception of public schooling that would allow for the constitutionally permissible regulation of antigay-identity speech. Next, through the use of critical theory, this Article explains why First Amendment defenses to gay harassment have …

Long-Term Survival Of Anoxia Despite Rapid Atp Decline In Embryos Of The Annual Killifish Austrofundulus Limnaeus., Jason Podrabsky, Michael Menze, Steven Hand

Faculty and Staff Scholarship

Embryos of the annual killifish Austrofundulus limnaeus can survive for months in the complete absence of oxygen. Survival of anoxia is associated with entry into a state of metabolic dormancy known as diapause. However, extreme tolerance of anoxia is retained for several days of post-diapause development. Rates of heat dissipation in diapause II and 4 days post-diapause II embryos were measured under aerobic conditions and during the transition into anoxia. Phosphorylated adenylate compounds were quantified in embryos during entry into anoxia and after 12 hr of aerobic recovery. Rates of heat dissipation were not affected by exposure to anoxia in …

Aloe Striata Plant Defense Compounds Produced In Response To Jasmonic Acid, Salicylic Acid, And Eriophyid Mite Salivary Extract Elicitor Compounds, Fritz Light

Master's Theses

Aloe mites are herbivores of the genus Aloe (ALOACEAE) and are associated with hyperplastic growth in various aloe species, but the biochemistry of this interaction is poorly understood. In an effort to characterize plant defense responses to herbivory in the genus Aloe, a salivary extract was isolated from aloe mites (Aceria aloinis Keifer) and its bioactivity was tested using a hypocotyl elongation assay. Subsequently, Aloe striata plants were treated with jasmonic acid (JA), salicylic acid (SA), and the mite salivary extract. Using water and methanol, compounds of different polarity were extracted from aloe tissues that had been frozen …

Multivariate and univariate statistical analyses of morphometric data and examination of morphological characters revealed the existence of four undescribed species from populations previously considered to belong to C. longirostris; these were located on the southern extents of the range of C. longirostris. Three of these new species were morphologically similar to the group of species containing C. coosawattae, C. chasmodactylus, C. elkensis, C. longirostris, C. longulus, and C. manningi, while one was morphologically similar to the C. fasciatus, C. …

Lignin Modification In Arabidopsis And Populus For Studies Of Gene Function And Improving Lignin Degradation, Yi Xu Aug 2012

Lignin Modification In Arabidopsis And Populus For Studies Of Gene Function And Improving Lignin Degradation, Yi Xu

All Dissertations

Lignin is one of the most abundant biopolymers in plants and plays an important role in plant structure and stress defense. Lignin is also considered to be a hallmark of vascular plants because of its crucial role in plant terrestrialization. However, lignin is an undesired component in the pulp and paper industry, bioethanol production, and forage digestibility. Thus, understanding the functions and the evolution of lignin biosynthesis genes can not only advance our knowledge of the evolution of land-adaptation for vascular plants but also help guide the effort to exploit the potential for genetic manipulation of lignin for desirable traits …

The Influence Of Flagship Species On In Situ And Ex Situ Wildlife Tourists' Connection To Wildlife And Pro-Conservation Behaviors, Jeffrey Skibins Aug 2012

The Influence Of Flagship Species On In Situ And Ex Situ Wildlife Tourists' Connection To Wildlife And Pro-Conservation Behaviors, Jeffrey Skibins

All Dissertations

Annually, millions of tourists visit natural areas and zoos primarily to view flagship species such as lions and elephants. Venues rely on the inherent charisma of these species to increase visitation and anchor conservation efforts. Expected visitor outcomes from the use of flagships include raised levels of awareness and pro-conservation behaviors. However, the role of flagships in wildlife tourism has been criticized for not delivering conservation benefits for species of interest or biodiversity, and producing negative site impacts. Furthermore, little is known about how the connection to a species influences conservation behaviors. This dissertation addresses this gap in knowledge by …

Essays In Family Economics, Bing Bai

All Dissertations

The first chapter examines the effect of the number of years children spend living with a single-parent family instead of a two-parent family on children's completed schooling, based on a sample of children from the PSID. To deal with the endogeneity of mothers' family structure decisions, I exploit the variation across states and over time in unilateral divorce laws, unmarried fertility ratios, welfare rules, earned income tax credit rates, and labor market conditions that generate plausibly exogenous changes in mothers' family structure choices. I construct a set of extensive measures for these contextual variables and use them as instruments to …

Ecology And Phylogeny Of The Biting-Midge Genus Culicoides (Diptera: Ceratopogonidae), Dustin Swanson

All Dissertations

Flies of the genus Culicoides are small, biting midges in the family Ceratopogonidae. These flies are of medical, veterinary, and economic importance because their blood-feeding behavior can cause stress to hosts and transmit disease agents. Despite the importance of these flies, little is known about their biology, especially the ecology of the immature stages and the phylogenetic relationships among taxa. The objectives of my study were to address these two areas of Culicoides biology.
Larval Culicoides were collected from aquatic habitats in four ecoregions of South Carolina, USA. Eleven ecological variables were recorded for each sample. Larvae were identified by …
