Macrophage migration inhibitory factor (MIF) is a catalytic cytokine and an upstream mediator of the inflammatory pathway. MIF has broad regulatory properties, dysregulation of which has been implicated in the pathology of multiple immunological diseases. Inhibition of MIF activity with small molecules has proven beneficial in a number of disease models. Known small molecule MIF inhibitors typically bind in the tautomerase site of the MIF trimer, often covalently modifying the catalytic proline. Allosteric MIF inhibitors, particularly those that associate with the protein by noncovalent interactions, could reveal novel ways to block MIF activity for therapeutic benefit and serve as chemical …

Previous work has identified Mesp1 as an important regulator of the epithelial–mesenchymal transition: EMT) and of cardiovascular cell fate in differentiating embryonic stem cells: ESCs). To understand the molecular mechanisms underlying the actions of Mesp1, we sought to identify transcription targets of Mesp1. Mesp1 rapidly induced expression of PDGFRα in differentiating ESCs and directly bound to evolutionary conserved E-boxes within the PDGFRα promoter. This result suggested that PDGFRα could be a direct target of Mesp1. However, we found that PDGFRα was not sufficient for the induction of EMT in ESCs or the induction of Flk1+ mesoderm, but that it may …

Challenges Of Permitting Ballast Water Discharges From Commercial Vessels In U.S. Waters, Paul Rodriguez

On February 5, 2009, the US Environmental Protection Agency (US EPA) issued the Vessel General Permit (VGP), under the Clean Water Act (CWA), for discharges incidental to the normal operation of vessels, including ballast water discharges. This set forth permit requirements for most commercial vessel discharges within 3 nautical miles of U.S. shore. Since it will expire in December 2013, the US EPA recently issued a draft 2013 VGP. One of the significant changes is the proposal of numerical ballast water discharge standards requiring most commercial vessels to install ballast water treatment systems onboard.

Early detection of diseased cells can greatly alter the course of a disease process and create a better outcome for the patient. Lymphangioleiomyomatosis (LAM) is a disease characterized by gradual destruction of lung tissue caused by infiltration and proliferation of smooth muscle-like cells. Currently little can be done to treat the disease other than to ease symptoms. Early LAM diagnosis can greatly impact the course of the disease and the patient’s quality of life. Background: Olive (Olea europaea L.) cultivation is rapidly expanding and low quality saline water is often used for irrigation. The molecular basis of salt tolerance in olive, though, has not yet been investigated at a system level. In this study a comparative transcriptomics approach was used as a tool to unravel gene regulatory networks underlying salinity response in olive trees by simulating as much as possible olive growing conditions in the field. Specifically, we investigated the genotype-dependent differences in the transcriptome response of two olive cultivars, a salt-tolerant and a salt-sensitive one.

Neuronal damage is a hallmark feature of HIV-associated neurological disorders (HANDs). Opiate drug abuse accelerates the incidence and progression of HAND; however, the mechanisms underlying the potentiation of neuropathogenesis by these drugs remain elusive. Opiates such as morphine have been shown to enhance HIV transactivation protein Tat-mediated toxicity in both human neurons and neuroblastoma cells. In the present study, we demonstrate reduced expression of the tropic factor platelet-derived growth factor (PDGF)-B with a concomitant increase in miR-29b in the basal ganglia region of the brains of morphine-dependent simian immunodeficiency virus (SIV)-infected macaques compared with the SIV-infected controls. Weight-loss interventions generally improve lipid profiles and reduce cardiovascular disease risk, but effects are variable and may depend on genetic factors. We performed a genetic association analysis of data from 2,993 participants in the Diabetes Prevention Program to test the hypotheses that a genetic risk score (GRS) based on deleterious alleles at 32 lipid-associated single-nucleotide polymorphisms modifies the effects of lifestyle and/or metformin interventions on lipid levels and nuclear magnetic resonance (NMR) lipoprotein subfraction size and number. During its somatic nuclear differentiation, the single cell eukaryote Tetrahymena thermophila undergoes genome-wide programmed DNA rearrangement to eliminate transposon-like elements from its future soma. This process involves small RNA-directed heterochromatin formation followed by extensive nuclear reorganization to form subnuclear domains. While more has been known about small RNAs and heterochromatin, the mechanisms and players involved in the process of nuclear reorganization and the subsequent removal of transposon-like elements from the somatic genome are just starting to unravel. Raman spectroscopy is used to gather information on the mineral and organic components of bone tissue to analyze their composition. By measuring the Raman signal of bone through spatially offset Raman spectroscopy the health of the bone can be determined. We’ve customized a system with 8 collection channels that consist of individual fibers, which are coupled to separate spectrometers and cooled CCDs. This parallel detection system was used to scan gelatin phantoms with Teflon inclusions of two sizes. Raman signals were decoupled from the autofluorescence background using channel specific polynomial fitting. Images with high contrast to background ratios of Raman …

The Characterization and Radiometric Calibration for Remote Sensing Annual Meeting (CALCON)
The National Ecological Observatory Network (NEON) is a continental-scale ecological observation facility currently under construction by the National Science Foundation (NSF). NEON’s mission is to enable understanding and forecasting of the impacts of land-use change and invasive species by providing the infrastructure and consistent methodologies in these areas. The Airborne Observation Platform (AOP) will play a unique role in scaling individual in-situ measurements to those collected by satellite-based remote sensing. The current payload consists of the NEON Imaging Spectrometer Design Verification Unit (NISDVU), a waveform LIDAR, and a high-resolution camera integrated into a platform integration mount (PIM). Much of the current research on global and local food insecurity – particularly among poor urban populations – strongly suggests that the impacts of industrial agriculture and the American fast food culture has been detrimental to both urban and rural populations, and to small farmers. Food insecurity and unequal access to healthy and affordable food are critical issues in the metropolis. In response to these concerns, there has been a great deal of mobilization around the concept of local and regional food systems. Human aging is characterized by both physical and physiological frailty. A key feature of frailty, sarcopenia is the age-associated decline in skeletal muscle mass, strength, and endurance that characterize even the healthy elderly. Increases in adiposity, particularly in visceral adipose tissue, are almost universal in aging individuals and can contribute to sarcopenia and insulin resistance by increasing levels of inflammatory cytokines known collectively as adipokines. Aging also is associated with declines in adaptive and innate immunity, known as immune senescence, which are risk factors for cancer and all-cause mortality.
