A Conserved Two-Component Regulatory System, Pids/Pidr, Globally Regulates Pigmentation And Virulence-Related Phenotypes Of Burkholderia Glumae, Hari Sharan Karki, Inderjit Kaur Barphagha, Jong Hyun Ham

Burkholderia glumae is a rice pathogenic bacterium that causes bacterial panicle blight. Some strains of this pathogen produce dark brown pigments when grown on casamino-acid peptone glucose (CPG) agar medium. A pigment-positive and highly virulent strain of B.glumae, 411gr-6, was randomly mutagenized with mini-Tn5gus, and the resulting mini-Tn5gus derivatives showing altered pigmentation phenotypes were screened on CPG agar plates to identify the genetic elements governing the pigmentation of B.glumae. In this study, a novel two-component regulatory system (TCRS) composed of the PidS sensor histidine kinase and the PidR response regulator was identified as an essential regulatory factor for pigmentation. Inferring The Evolutionary History Of Divergence Despite Gene Flow In A Lizard Species, Scincella Lateralis (Scincidae), Composed Of Cryptic Lineages, Nathan D. Jackson, Christopher C. Austin

Although recent radiations are fruitful for studying the process of speciation, they are difficult to characterize and require the use of multiple loci and analytical methods that account for processes such as gene flow and genetic drift. Using multilocus sequence data, we combine hierarchical cluster analysis, coalescent species tree inference, and isolation-with-migration analysis to investigate evolutionary relationships among cryptic lineages of North American ground skinks. We also estimate the extent that gene flow has accompanied or followed diversification, and also attempt to account for and minimize the influence of gene flow when reconstructing relationships. Cryopreservation of sperm from Xiphophorus fishes has produced live young in three species: X. hellerii, X. couchianus, and X. maculatus. In this study, the goal was to establish protocols for sperm cryopreservation and artificial insemination to produce live young in X. variatus, and to identify needs for repository development. The objectives were to: 1) collect basic biological characteristics of males; 2) cryopreserve sperm from X. variatus, 3) harvest live young from cryopreserved sperm, and 4) discuss the requirements for establishment of sperm repositories. Background: Co-occurring disorders present a challenge for providers who often fail to diagnose them with accuracy. This study explores the assessment process of co-occurring depressive and substance use disorders in community health clinics serving ethnically diverse patients. The goals are to identify how symptoms discussed in intake, as well as patient and provider characteristics, are associated with receiving a diagnosis of co-occurring disorders or not. Methods: Participation in the study consisted of videotaping the intake, conducting a semi-structured interview, and completing demographic and clinical measures. The goal of the ICEFISH 2004 cruise, which was conducted on board RVIB Nathaniel B. Palmer and traversed the transitional zones linking the South Atlantic to the Southern Ocean, was to compare the evolution, ecology, adaptational biology, community structure, and population dynamics of Antarctic notothenioid fishes relative to the cool/temperate notothenioids of the sub-Antarctic. To place this work in a comprehensive ecological context, cruise participants surveyed the benthos and geology of the biogeographic provinces and island shelves on either side of the Antarctic Polar Front (or Antarctic Convergence). The Borrelia burgdorferi BpaB proteins of the spirochete's ubiquitous cp32 prophages are DNA-binding proteins, required both for maintenance of the bacteriophage episomes and for transcriptional regulation of the cp32 erp operons. Through use of DNase I footprinting, we demonstrate that BpaB binds the erp operator initially at the sequence 5′-TTATA-3′. Electrophoretic mobility shift assays indicated that BpaB also binds with high affinity to sites located in the 5′ noncoding regions of two additional cp32 genes. Characterization of the proteins encoded by those genes indicated that they are a single-stranded DNA-binding protein and a nuclease, which we named SsbP and NucP, …

The domestic dog offers a unique opportunity to study complex disorders similar to those seen in humans, but within the context of the much simpler genetic backgrounds of pure breeds, which represent closed populations. We performed a whole-genome search for genetic risk factors of adult-onset deafness in the Border Collie, a breed of herding dog that relies on acute hearing to perceive and respond to commands while working. Adult-onset deafness in Border Collies typically begins in early adulthood and is similar to age-related hearing loss in humans. Maximal Force Characteristics Of The Ca2+-Powered Actuator Of Vorticella Convallaria, Sangjin Ryu, Matthew J. Lang, Paul Matsudaira

The millisecond stalk contraction of the sessile ciliate Vorticella convallaria is powered by energy from Ca2+ binding to generate contractile forces of ~10 nN. Its contractile organelle, the spasmoneme, generates higher contractile force under increased stall resistances. By applying viscous drag force to contracting V. convallaria in a microfluidic channel, we observed that the mechanical force and work of the spasmoneme depended on the stalk length, i.e., the maximum tension (150–350 nN) and work linearly depended on the stalk length (~2.5 nN and ~30 fJ per 1 mm of the stalk). Endogenous Inhibitor Proteins That Connect Ser/Thr Kinases And Phosphatases In Cell Signaling., Masumi Eto, David L Brautigan

Protein phosphatase activity acts as a primary determinant of the extent and duration of phosphorylation of cellular proteins in response to physiological stimuli. Ser/Thr protein phosphatase-1 (PP1) belongs to the PPP superfamily, and is associated with regulatory subunits that confer substrate specificity, allosteric regulation, and subcellular compartmentalization. In addition, all eukaryotic cells contain multiple heat-stable proteins that originally were thought to inhibit phosphatase catalytic subunits released from the regulatory subunits, as a fail-safe mechanism. In recent years, trends in the medical industry have created a growing demand for implantable medical devices. In particular, the need to provide doctors a means to continuously monitor biometrics over long time scales with increased precision is paramount to efficient healthcare. To make medical implants more attractive, there is a need to reduce their size and power consumption. Small medical implants would allow for less invasive procedures, greater comfort for patients, and increased patient compliance. Reductions in power consumption translate to longer battery life. An Anatomical, Histological And Ultrastructural Study On The Placenta Of The Red-Shanked Douc Langur (Pygathrix Nemaeus), Damri Darawiroj, Natthaya Tuaprakone, Yongchai Utara
The Thai Journal of Veterinary Medicine
A red-shanked douc langur (Pygathrix nemaeus nemaeus) is currently enlisted as an endangered primatespecies. This study was undertaken to describe the gross anatomy, histology and ultrastructural morphology of thedouc langur term placenta by light, scanning, and transmission electron microscopy. The primary disc of bidiscoidalplacenta was separated to 4-6 small cotyledonary subdivisions and was attached by an umbilical cord. The chorionfrondosum of placenta was classified as villous, haemomonochorial and deciduate type. The interhaemal area wascomposed of a layer of syncytiotrophoblast that occasionally clustered as the syncytial knot, fetal capillaryendothelium and its basement membrane. In 2011, I set up an outdoor dyeing and weaving studio that also functioned as a sculptural installation, The Earthway Studio. The Ponseti method is a widely accepted and highly successful conservative treatment of pediatric clubfoot that relies on weekly manipulations and cast applications. However, the material behavior of the cast in the Ponseti technique has not been investigated. The current study sought to characterize the ability of two standard casting materials to maintain the Ponseti corrected foot position by evaluating creep response. A dynamic cast testing device (DCTD) was built to simulate a typical pediatric clubfoot. Characterisation Of Friction And Lubrication Regimes In Premium Tubular Connections, F Stewart, Hr Le, Ja Williams, Et Al

A friction test rig has been developed to carry out repeated sliding friction tests for premium tubular connections. The test rig enables accurate measurement of friction in various contact regimes which are relevant to the threaded connections between tubular components. Higher load tests can simulate the contact in metal-to-metal seals under very high contact pressures by using perpendicular pin-on-pin tests. The contact in the thread loading flank under intermediate pressures can be simulated by using larger radius coupon-on-coupon tests. The measured coefficient of friction is well correlated with a lubrication parameter combining lubricant film thickness and initial surface roughness.
