Fecal Indicator Bacteria In Knitting Mill Creek With A Comparison Of Methods Used To Enumerate E. Coli, Manju Nagarajan

OES Theses and Dissertations

This research seeks to evaluate the impact of an urban, multi-use neighborhood on the bacteriological quality of water in Knitting Mill Creek, a blind arm of the Lafayette River, a sub-estuary of the lower Chesapeake Bay. A principal objective was to determine the effects of rainfall, a surrogate for run-off, on water-column concentrations of fecal-indicator bacteria (FIB). I collected water samples weekly (September 2009-December 2010) and biweekly (January 2010-December 2011) at a storm-sewer outfall and a nearby marina and quantified their E. coli and enterococci concentrations using Colilert-18 and Enterolert, most-probable-number assays. In addition, I took monthly samples for these …

Life History Of Stranded Bottlenose Dolphins (Tursiops Truncatus) In Virginia, Margaret Cook Lynott

Biological Sciences Theses & Dissertations

Little is known about the life history and overall ecology of bottlenose dolphins that strand in Virginia. In this study, I examined archived life history samples and stranding data from bottlenose dolphins found in Virginia waters to (1) develop growth curves based on ages from sectioned teeth, (2) document characteristics of male and female reproductive organs, and (3) define the relationship between age/length and presence of neonatal characteristics during the first year of life. The Gompertz growth model provided the best fit of growth data for stranded T. truncatus in Virginia. Length at birth and asymptotic length for males were …

Helicobacter Pylori Hydrogenase Accessory Protein Hypa And Urease Accessory Protein Ureg Compete With Each Other For Uree Recognition, Stéphane L. Benoit, Jonathan L. Mcmurry, Stephanie A. Hill, Robert J. Maier

Faculty Articles

Background: The gastric pathogen Helicobacter pylori relies on nickel-containing urease and hydrogenase enzymes in order to colonize the host. Incorporation of Ni2+ into urease is essential for the function of the enzyme and requires the action of several accessory proteins, including the hydrogenase accessory proteins HypA and HypB and the urease accessory proteins UreE, UreF, UreG and UreH. Methods: Optical biosensing methods (biolayer interferometry and plasmon surface resonance) were used to screen for interactions between HypA, HypB, UreE and UreG. Results: Using both methods, affinity constants were found to be 5nM and 13nM for HypA–UreE and 8μM and 14μM for …

Evolution On The Hawaiian Hotspot: Biogeography And Divergence Time Estimation Of Kadua (Rubiaceae), Kenneth L. Parker Oct 2012

Evolution On The Hawaiian Hotspot: Biogeography And Divergence Time Estimation Of Kadua (Rubiaceae), Kenneth L. Parker

Biological Sciences Theses & Dissertations

Recent studies on the colonization time of Hawaiian plant and animal lineages have suggested certain lineages arrived to the archipelago before the formation of the extant islands. Kadua, a recently resurrected genus, is mostly endemic to the Hawaiian Archipelago, with many member taxa autochthonous to individual islands. This work investigates the colonization time of Kadua to the Hawaiian Islands. Sequence analyses of the quickly evolving nuclear regions ITS, ETS and 5s-NTS regions for 20 Hawaiian and 7 French Polynesian species of Kadua were used to create dated phylogeny calibrated with island ages. The phylogeny suggests a single colonization of Hawaii …

School of Medicine Publications

Background: Lipopolysaccharides (LPSs) up-regulate proinflammatory cytokines in macrophages, partly through a NF-κB-dependent process.

Results: Blocking neddylation, which helps regulate NF-κB, represses LPS-induced up-regulation of proinflammatory cytokines.

Conclusion: Neddylation plays a role in the up-regulation of NF-κB-regulated proinflammatory cytokines produced by macrophages in response to LPS.

Significance: Inhibition of neddylation represents a novel and effective method for the prevention of LPS-induced proinflammatory cytokines.

Functions Of The Arabidopsis Kinesin Superfamily Of Microtubule-Based Motor Proteins, Chuanmei Zhu, Ram Dixit Oct 2012

Functions Of The Arabidopsis Kinesin Superfamily Of Microtubule-Based Motor Proteins, Chuanmei Zhu, Ram Dixit

Biology Faculty Research

Plants possess a large number of microtubule-based kinesin motor proteins. While the kinesin-2, 3, 9, and 11 families are absent from land plants, the kinesin-7 and 14 families are greatly expanded. In addition, some kinesins are specifically present only in land plants. The distinctive inventory of plant kinesins suggests that kinesins have evolved to perform specialized functions in plants. Plants assemble unique microtubule arrays during their cell cycle, including the interphase cortical microtubule array, preprophase band, anastral spindle and phragmoplast. In this review, we explore the functions of plant kinesins from a microtubule array viewpoint, focusing mainly on Arabidopsis kinesins. …

Neural Responses To Infants Linked With Behavioral Interactions And Testosterone In Fathers, Patty X. Kuo, Joshua Carp, Kathleen C. Light, Karen M. Grewen Oct 2012

Neural Responses To Infants Linked With Behavioral Interactions And Testosterone In Fathers, Patty X. Kuo, Joshua Carp, Kathleen C. Light, Karen M. Grewen

Department of Child, Youth, and Family Studies: Faculty Publications

Few fMRI studies have investigated the brain-behavioral basis of parenting in human fathers. Ten fathers were videotaped and gave salivary testosterone samples while interacting with their 2–4 month old infants, and viewed video clips of their own infant and an unfamiliar age-, ethnicityand sex-matched other infant during an fMRI protocol. Infant stimuli activated a network of prefrontal and subcortical brain regions. Furthermore, a subset of these regions activated significantly more to own (OWN) than other (OTHER) infants. Finally, neural responses to OWN versus OTHER were linked with paternal sensitivity, paternal reciprocity, and testosterone. In sum, our results provide a novel …

Dpy-30 Domain And Its Flanking Sequence Mediate The Assembly Modulation Of Flagellar Radial Spoke Complexes, Radhika Gopal, Kenneth W. Foster, Pinfen Yang Oct 2012

Dpy-30 Domain And Its Flanking Sequence Mediate The Assembly Modulation Of Flagellar Radial Spoke Complexes, Radhika Gopal, Kenneth W. Foster, Pinfen Yang

Biological Sciences Faculty Research and Publications

RIIa is known as the dimerization and docking (D/D) domain of the cyclic AMP (cAMP)-dependent protein kinase. However, numerous molecules, including radial spoke protein 2 (RSP2) in Chlamydomonas flagella, also contain an RIIa or a similar DPY-30 domain. To elucidate new roles of D/D domain-containing proteins, we investigated a panel of RSP2 mutants. An RSP2 mutant had paralyzed flagella defective in RSP2 and multiple subunits near the spokehead. New transgenic strains lacking only the DPY-30 domain in RSP2 were also paralyzed. In contrast, motility was restored in strains that lacked only RSP2’s calmodulin- binding C-terminal region. These cells swam normally …

Deep Diversification And Long-Term Persistence In The South American ‘Dry Diagona’: Integrating Continent-Wide Phylogeography And Distribution Modeling Of Geckos, Fernanda P. Werneck, Tony Gamble, Guarino R. Colli, Miguel T. Rodrigues, Jack W. Sites Jr. Oct 2012

Deep Diversification And Long-Term Persistence In The South American ‘Dry Diagona’: Integrating Continent-Wide Phylogeography And Distribution Modeling Of Geckos, Fernanda P. Werneck, Tony Gamble, Guarino R. Colli, Miguel T. Rodrigues, Jack W. Sites Jr.

Biological Sciences Faculty Research and Publications

The relative influence of Neogene geomorphological events and Quaternary climatic changes as causal mechanisms on Neotropical diversification remains largely speculative, as most divergence timing inferences are based on a single locus and have limited taxonomic or geographic sampling. To investigate these influences, we use a multilocus (two mitochondrial and 11 nuclear genes) range‐wide sampling of Phyllopezus pollicaris, a gecko complex widely distributed across the poorly studied South American ‘dry diagonal’ biomes. Our approach couples traditional and model‐based phylogeography with geospatial methods, and demonstrates Miocene diversification and limited influence of Pleistocene climatic fluctuations on P. pollicaris. Phylogeographic structure and …

Anaesthetic Management Of Neonatal Primary Hyperparathyroidism, Anwar-Ul-Huda, Sanam Dost Mohammad, Tauseef Ahmed

Department of Anaesthesia

Neonatal primary hyperparathyroidism (NPHP) is a rare disease that presents in the first 6 months of life. It is almost invariably fatal unless a prompt diagnosis is made and urgent surgical intervention is instituted. Patients with hyperparathyroidism require special preoperative considerations. They require proper work-up from cardiac, renal and neurological point of view, correction of intravenous volume and electrolytes irregularities. We are presenting a case of an infant who started to have cyanosis and lethargy at 21st day of life. Initial diagnosis of pneumonia was made but later on diagnosed as hyperparathyroidism. He was planned for parathyroidectomy under general anaesthesia. …

Characterisation Of The Colour Doppler "Twinkle" Artefact, Deirdre Cassidy, Andrew Fagan, Jacinta Browne Oct 2012

Characterisation Of The Colour Doppler "Twinkle" Artefact, Deirdre Cassidy, Andrew Fagan, Jacinta Browne

Articles

This investigation involved the development of a range of Colour Doppler Twinkle Artefact phantoms to characterise and quantify the “Twinkle” artefact which is often present when an irregular structure is encountered in the imaged field of view. The artefact occurs in both colour and power Doppler ultrasound imaging and manifests as a false depiction of colour velocity information in stationary soft tissue and therefore can cause significant misdiagnosis of areas of flow within the patient. It has been hypothesised that it is generated due to a strongly reflecting medium composed of individual reflectors and therefore becomes a clinical concern when …

Book Review: Slow Violence And The Environmentalism Of The Poor, By Rob Nixon, Dayna Nadine Scott Oct 2012

Book Review: Slow Violence And The Environmentalism Of The Poor, By Rob Nixon, Dayna Nadine Scott

Osgoode Hall Law Journal

IN SEEKING TO UNDERSTAND the relationships of the local to the global in contemporary environmental justice struggles, we have yearned for a way to trace the “occluded relationships” between transnational economic actors and the things that tie them to particular places, such as labour, land, resources, and commodity dynamics. The bodies caught in the middle have been ‘raced’ and erased, made invisible, and wiped away. With gripping urgency, Rob Nixon’s book Slow Violence and the Environmentalism of the Poor brings those bodies back into view by exposing the violence perpetrated against them across time and space.

Journal Articles: Ophthalmology

Reprogramming somatic cells to a pluripotent state by nucleic acid based (NAB) approaches, involving the ectopic expression of transcription factors, has emerged as a standard method. We recently demonstrated that limbal progenitors that regenerate cornea are reprogrammable to pluripotency by a non-NAB approach through simple manipulation of microenvironment thus extending the possible therapeutic use of these readily accessible cells beyond the proven treatment of corneal diseases and injury. Therefore, to determine the validity and robustness of non-cell autonomous reprogramming of limbal progenitors for a wider clinical use, here, we have compared their reprogramming by non-NAB and NAB approaches. We observed …

The endogenous opioid system is involved in modulating a number of behavioral and physiological systems, including the hypothalamic-pituitary-adrenal (HPA) axis. In humans, a functional variant in the OPRM1 gene (OPRM1 A118G) is associated with a number of outcomes, including attenuated HPA axis responses to stress. A nonsynonymous variant (OPRM1 C77G) in the rhesus macaque has been shown to have similar effects in vivo to the human variant. The current study investigated whether OPRM1 C77G influences HPA axis response to stress in rhesus macaques. We analyzed plasma adrenocorticotropic hormone (ACTH) and cortisol levels measured in response to three different stressors: 1) …

Ringing The Bell On Concussions: The Rise Of Head Injuries And Cognitive Decline In Football Players, And The Nfl's Obligation To Improve Safety Measures, Jarryd Werts

Cardozo Public Law, Policy & Ethics Journal

The NFL faces significant legal and ethical challenges in addressing the long-term health consequences of concussions among its players. While the league has implemented various safety measures and policies, such as rule changes and educational initiatives, these efforts may be insufficient to fully protect players from the risks associated with repeated head trauma. The NFL must continue to improve its safety protocols not only to safeguard its current and future players but also to set a precedent for other contact sports leagues. The legal battles over negligence and fraud highlight the need for a more robust approach to player health …
