Provided are methods for screening for drugs effective for treating fibrotic conditions. One screening method comprises exposing a cell to a test compound, monitoring the effect of the test compound on the amount or form of a cell molecule, comparing the amount or form of the cell molecule with the result obtained by treatment of the cell with an anti-fibrotic -effective amount of a withanolide compound, and selecting a drug effective for treating a fibrotic disease based on the ability of the test compound to provide the effect obtained by the withanolide compound on the cell molecule. Many polymers, such as PMMA, are very susceptible to swelling or dissolution by organic solvents. Growing covalently attached polymer brushes from these surfaces by atom-transfer radical polymerization (ATRP) is challenging because of the typical requirement of organic solvent for initiator immobilization. We report an unprecedented, aqueous-based route to graft poly(N-isopropylacrylamide), PNIPAAm, from poly(methyl methacrylate), PMMA, surfaces by ATRP, wherein the underlying PMMA is unaffected. Successful attachment of the ATRP initiator, N-hydroxysuccinimidyl-2-bromo-2- methylpropionate, on amine-bearing PMMA surfaces was confirmed by XPS. From this surface-immobilized initiator, thermoresponsive PNIPAAm brushes were grown by aqueous ATRP to yield optically transparent PNIPAAm-grafted PMMA surfaces. This …

The antiapoptotic Bcl-2 family member Mcl-1 is a PEST protein (containing sequences enriched in proline, glutamic acid, serine, and threonine) and is subject to rapid degradation via multiple pathways. Impaired degradation leading to the maintenance of Mcl-1 expression is an important determinant of drug resistance in cancer. Phosphorylation at Thr 163 in the PEST region, stimulated by 12-O-tetradecanoylphorbol acetic acid (TPA)-induced activation of extracellular signal-regulated kinase (ERK), is associated with Mcl-1 stabilization in BL41-3 Burkitt lymphoma cells. MicroRNAs (miRNAs) are a major class of small, noncoding RNA molecules that regulate gene expression by targeting mRNAs to trigger either translational repression or mRNA degradation. They have recently been more widely investigated due to their potential role as targets for cancer therapy. Many miRNAs have been implicated in several human cancers, including breast cancer. miRNAs are known to regulate cell cycle and development, and thus may serve as useful targets for exploration in anticancer therapeutics. School Food Environment The Frontline For Childhood Obesity Prevention: A Mixed-Method Study Of Nutritional Competencies And Skills Of School Nutrition Professionals In Nebraska, Zainab Rida

The purpose of this statewide study was to address the nutrition knowledge, attitudes, and perceptions of school foodservice personnel in Nebraska regarding offering/serving healthy school meals. Moreover, this study identified some potential barriers and avenues of action for offering/serving healthy school meals. Data obtained from this study indicates that there is a strong correlation (r= .103, p r .237, p< .01) between the foodservice staff’s self-efficacy and their practices of offering/serving healthy school meals. Fortunately, the relationship between foodservice staff practices of offering/serving healthy school meals and their self-efficacy was positive and significantly predicted practices scores, β =.237, P< 0.01. The finding of the present study also identifies many barriers including lack of time and support that face the foodservice personnel in offering/serving healthy school meals. The findings suggested that there is an urgent need of a full school approach to promote and encourage healthy eating habits among students. Future research is needed to evaluate school wellness policies regarding healthy eating practices in schools. Moreover, establish partnerships with communities and universities for intervention that target students and their parents.

There is a large body of scientific evidence suggesting that 3,39-Diindolylmethane (DIM), a compound derived from the digestion of indole-3-carbinol, which is abundant in cruciferous vegetables, harbors anti-tumor activity in vitro and in vivo. Accumulating evidence suggests that AMP-activated protein kinase (AMPK) plays an essential role in cellular energy homeostasis and tumor development and that targeting AMPK may be a promising therapeutic option for cancer treatment in the clinic. Hepatitis C virus (HCV) infection is a major cause of chronic liver disease worldwide. The interplay between the virus and host innate and adaptive immune responses determines the outcome of infection. There is increasing evidence that host neutralizing responses play a relevant role in the resulting pathogenesis. We report the results of a study focused on identifying and articulating an ‘‘epistemic foundation’’ underlying a pre-collegiate focus on engineering. We do so in the context of UTeachEngineering (UTE), a program supported in part by funding by the National Science Foundation and designed to develop a model approach to address the systematic challenges facing this work—from identifying learning goals, to certifying pre- and in-service teachers for engineering courses to developing a research-based high school engineering course. This phenomenological study examined the impact of Research Experiences for Teachers (RET) teacher-developed curriculum on teaching styles and strategies at two RET sites with common Legacy Cycle training. The study was conducted to assess and document program-specific and National Science Foundation (NSF) goals related to classroom practices and outcomes. Impulsivity as a symptom during mania is well documented, but less is known about the driving factors behind these impulsive acts. Perhaps this is because impulsivity is a term that encompasses multiple facets. There is evidence for a type of impulsivity in which extreme sensitivity to positive affective states can influence cognitive processes, such as error detection and identification of threatening cues. This can ultimately lead to engagement in impulsive actions during elevated mood states. Stochastic interventions are a powerful tool to define parameters that measure the causal effect of a realistic intervention that intends to alter the population distribution of an exposure. In this paper we follow the approach described in D\'iaz and van der Laan (2011) to define and estimate the effect of an intervention that is expected to cause a truncation in the population distribution of the exposure. The C-terminal nuclear localization sequence of FUsed in Sarcoma (FUS-NLS) is critical for its nuclear import mediated by transportin (Trn1). Familial amyotrophic lateral sclerosis (ALS) related mutations are clustered in FUS-NLS. We report here the structural, biochemical and cell biological characterization of the FUS-NLS and its clinical implications. The crystal structure of the FUS-NLS/Trn1 complex shows extensive contacts between the two proteins and a unique α-helical structure in the FUS-NLS. The binding affinity between Trn1 and FUS-NLS (wide-type and 12 ALS-associated mutants) was determined. Influenza A virus (IAV) is a leading cause of respiratory tract disease worldwide. Anti-viral CD8+ T lymphocytes responding to IAV infection are believed to eliminate virally infected cells by direct cytolysis but may also contribute to pulmonary inflammation and tissue damage via the release of pro-inflammatory mediators following recognition of viral antigen displaying cells. We have previously demonstrated that IAV antigen expressing inflammatory cells of hematopoietic origin within the infected lung interstitium serve as antigen presenting cells (APC) for infiltrating effector CD8+ T lymphocytes; however, the spectrum of inflammatory cell types capable of serving as APC was not …
