DNA double-strand breaks (DSB) with chemically modified end-termini are the most significant lesions resulting from radio/chemotherapeutic intervention of cancer and non homologous end-joining (NHEJ) factor Artemis nuclease has been implicated in the repair of such breaks. To examine whether the resolution of terminally blocked DNA DSBs is the biologically relevant function of Artemis, Artemis deficient fibroblasts were stably complemented with wild type or an endonuclease deficient D165N mutant Artemis. Physiological levels of wild type (WT) Artemis completely restored DSB repair proficiency and resistance to γ-radiation, bleomycin, and neocarzinostatin. Introduction: Fibromyalgia (FMS) is a chronic pain syndrome characterized by non-restorative sleep, and fatigue. Over 75% of individuals with FMS complain of poor sleep quality and fatigue. These have been ranked by patients with FMS as having great impact on quality of life. A literature review suggested that poor sleep quality may be a predictor of increased stress and FMS symptom onset in those affected. However, no experimental studies have demonstrated a causal relationship between poor sleep and stress in people with FMS. In these studies we sought to determine the role of Fyn kinase and ADAM10 in B cell biology. A disintegrin and metalloproteinase 10 (ADAM10) is a zinc dependent proteinase related to matrix metalloproteinases. ADAM10 has emerged as a key regulator of cellular processes by cleaving and shedding extracellular domains of multiple transmembrane receptors and ligands. In particular, ADAM10 has been identified as a key regulator of lymphocyte development. Here we report that ADAM10 is dispensable for early B cell development within the bone marrow. Tobacco dependence is high in women who suffer from anxiety disorders yet little is known about the contributions of nicotinic acetylcholine receptors (nAChRs) on anxiety-like behavior. β2*nAChRs (*denotes assembly with other subunits) are the most abundantly expressed nAChRs in the brain yet little is known about the contributions of β2*nAChRs on anxiety-like behavior in female mice. In this study, antagonism and nicotine effects on anxiety-like behavior was investigated across the life span in 6, 12 and 24-month-old drug-naïve knockout (KO), heterozygous (HET) and a gain of function α6L9S mice and wild type (WT). The signal transducer and activator of transcription 1 (STAT1) and 2 (STAT2) are primarily activated by interferons and play a central role in orchestrating responses to a variety of pathogens by activating the transcription of nuclear encoded genes that mediate antiviral, antigrowth and immune surveillance responses. In addition to their nuclear effects, we report that STAT1 and STAT2 inhibit the expression of mitochondrial encoded mRNAs by activating type I interferons (IFNβ) under basal conditions. STAT1-/- livers also exhibit elevated levels of nuclear encoded components of the electron transport chain (ETC). Treatment of wild-type mice with IFNβ inhibits both mitochondrial and …

Despite external bilateral symmetry, the internal organs of vertebrates are asymmetrically arranged. The development of Left-Right (LR) asymmetry is dependent on the activity of a ciliated organ known as the Kupffer’s vesicle (KV) or the embryonic node, in zebrafish and mouse, respectively. Cilia beating within the KV are essential to generate a counterclockwise flow of fluid, which leads to left-sided Ca2+ elevation. Ca2+ is a crucial morphogenic second messenger during vertebrate development. Ca2+ elevation activates targets, such as CaMK-II, which is also known as the multifunctional Ca2+/calmodulin-dependent protein kinase. Plants respond with a sink stimulation of photosynthesis when colonized by bacterial and fungal mutualists, which compensates for costs of carbohydrate allocation to the microbes. Problems may arise when light is limited and plants cannot increase photosynthesis. We hypothesize that under such conditions the costs for maintaining the mutualism outweigh the benefits, which ultimately turns the beneficial microbes into parasites exploiting resources and reducing host fitness. We study these plant-­‐microbe interactions under different light availabilities using lima bean plants, rhizobia (nitrogen-­‐fixing bacteria), and mycorrhizal fungi. The alien nonmarine gastropods of the Hawaiian islands were cataloged by Cowie (1997) and were reviewed by Cowie (1998a). Subsequent new records (both for the archipelago and for individual islands) were reported by Cowie (1998b, 1999, 2000), Kraus (2003), and, specifically associated with the horticultural industry between 2004 and 2006, by Hayes et al. (2007), reviewed and analyzed by Cowie et al. (2008). Subsequent surveys, covering 196 sites across the six largest main Hawaiian islands during 2006–2010, have resulted in the additional new records reported here. Surfactant-Sensitized Spectrophotometric Determination Of Hg(Ii) In Water Samples Using 2-(2-Thiazolylazo)- P-Cresol As Ligand And Cetylpyridinium Chloride As Cationic Surfactant, Ramazan Gürkan, Tuğba Çepken, Hali̇l İbrahi̇m Ulusoy

A new, reliable, surfactant-sensitized method is described for the spectrophotometric determination of total and dissolved inorganic mercury at trace levels. The method is based on the ternary complex formation of Hg(II) with 2-(2-thiazolylazo)-p-cresol and a cationic surfactant, cetylpyridinium chloride, at pH 9.5. The detection and quantification limits of the method with surfactant were 6 and 19 \mu g L^{-1}, while those of the method without surfactant were 22 and 67 \mu g L^{-1}, respectively. Flow injection cold vapor atomic absorption spectrometry (FI-CVAAS) was used for determination of cadmium in high salt matrices such as dialysis concentrates and textile leach solutions. The parameters such as acidity, concentration of reducing agent, reaction coil length, and carrier gas flow rate were investigated to obtain the best sensitivity. No significant background signal was observed even at high salt concentrations. Under the optimized conditions the limit of detection value (3s_b m^{-1}, where s_b is the standard deviation of the blank signals and m is the slope of the calibration graph) was found to be 0.05 ng mL^{-1} for an …

Two new triterpene glycosides, 3-O-\beta-D-glucopyranosyl- (1\to 4)-\beta-D-xylopyranosyl-(1\to 3)-\alpha-L-rhamnopyranosyl- (1\to 3)-\alpha-L-arabinopyranosyl olean-12-ene 28-O-\beta -D- glucopyranosyl-(1\to 6)-\beta-D-glucopyranosyl ester (gazipashoside A, 1) and 3-O-\beta-D-glucopyranosyl-(1\to 4)-\beta-D-xylopyranosyl- (1\to 3)-\alpha-L-rhamnopyranosyl-(1\to 2)-\alpha-L-arabinopyranosyl hederagenin 28-O-\beta -D-glucopyranosyl-(1\to 6)-\beta-D-glucopyranosyl ester (gazipashoside B, 2), were isolated together with 11 known compounds including 7 triterpene, 1 flavonoid, and 3 iridoidal metabolites from the aerial parts of Cephalaria gazipashensis (Dipsacaceae). The structures of all compounds were elucidated by extensive evaluation of the spectroscopic data (1D- and 2D- NMR, HRESI-MS), as well as chemical examinations. Students’ Attributions Of Instructor Credibility As A Function Of Instructors’ Out-Of-Class Support, Adam C. Jones, Paul Schrodt

Basic Communication Course Annual

This investigation examined the impact that instructor out-of-class support (OCS) and sex differences have on students’ perceptions of instructor credibility. Participants (N = 634) were randomly assigned to one of six experimental conditions manipulating the degree to which an instructor responds with a highly supportive, moderately supportive, or non-supportive message following a hypothetically stressful situation. Multivariate analyses revealed a two-way interaction effect of instructor OCS by student sex on perceptions of instructor credibility. Significant and meaningful main effects for instructor OCS on all three dimensions of credibility (i.e., competence, trustworthiness, and caring) were also obtained, with students attributing more credibility …

A web-based survey of 357 respondents enrolled in basic communication courses was conducted to examine communication center usage, communication apprehension, and help seeking behaviors. There was no significant difference between students who attended the communication center and those who did not in their communication apprehension and help seeking behaviors. There were significant correlations between help seeking behaviors and communication apprehension. Demographics, communication apprehension, and communication center usage and awareness predicted multiple types of help seeking behaviors.

How should religion and ethics be studied if we want to understand what people believe and why they act the way they do? In the 1980s and '90s postmodernist worries about led to debates that turned on power, truth, and relativism. Since the turn of the century scholars impressed by 'cognitive science' have introduced concepts drawn from evolutionary biology, neurosciences, and linguistics in the attempt to provide 'naturalist' accounts of religion. Understanding the genetic structure of parasite populations on the natural landscape can reveal important aspects of disease ecology and epidemiology and can indicate parasite dispersal across the landscape. Myxobolus cerebralis (Myxozoa: Myxosporea), the causative agent of whirling disease in the definitive host Tubifex tubifex, is native to Eurasia and has spread to more than 25 states in the USA. The small amounts of data available to date suggest that M. cerebralis has little genetic variability. Computational prediction of protein subcellular localization can greatly help to elucidate its functions. Despite the existence of dozens of protein localization prediction algorithms, the prediction accuracy and coverage are still low. Several ensemble algorithms have been proposed to improve the prediction performance, which usually include as many as 10 or more individual localization algorithms. However, their performance is still limited by the running complexity and redundancy among individual prediction algorithms.

Intercropping cereal grains and legumes has potential as an alternative cool-season forage crop in low-input farming systems. The objectives of this study were to quantify the effects of density, species proportion, and nitrogen (N) supply on the biomass accumulation of field pea and spring oat grown as sole or intercrops. Greenhouse experiments were conducted using an additive series experimental design. Treatments included three fertilizer N rates and 20 density/proportion combinations in a randomized complete block with four replications. The determination of the ideal breeding size of beef replacement females is traditionally centered on maximizing pregnancy rate. Relevant physical and economic relationships were combined into a bioeconomic systems model that identified key profit factors. This system-wide approach encapsulated the physical relationships with relevant costs and revenues, including annual and seasonal variations and measures relative to profitability through the application of an incomplete or modified profit function.
