Enteroviruses like coxsackievirus B3 (CVB3) are common suspects in myocarditis/dilated cardiomyopathy patients. Autoimmunity has been proposed as an underlying mechanism, but direct evidence of its role is lacking. To delineate autoimmune response in CVB3 myocarditis, we used IAk dextramers for cardiac myosin heavy chain (Myhc)-α 334–352. We have demonstrated that myocarditis-susceptible A/J mice infected with CVB3 generate Myhc-α-reactive CD4 T cells and such a repertoire was absent in naïve mice as measured by proliferative response to Myhc-α 334–352 and IAk dextramer staining. We also detected Myhc-α 334–352 dextramer+ cells in the hearts of CVB3-infected mice. The autoreactive …

The mechanism of the Golgi-to-ER transport of Golgi glycosyltransferases is not clear. We utilize a cell line expressing the core 2 N-acetylglucosaminyltransferase-M (C2GnT-M) tagged with c-Myc to explore this mechanism. By immunoprecipitation using anti-c-Myc antibodies coupled with proteomics analysis, we have identified several proteins including non-muscle myosin IIA (NMIIA), heat shock protein (HSP)-70 and ubiquitin activating enzyme E1 in the immunoprecipitate. Employing yeast-two-hybrid analysis and pulldown experiments, we show that the C-terminal region of the NMIIA heavy chain binds to the 1–6 amino acids in the cytoplasmic tail of C2GnT-M. BACKGROUND: Non-high-density lipoprotein (HDL) cholesterol is recommended as a secondary lipid goal treated initially with lifestyle modification. However, the relationship between non-HDL and subclinical atherosclerosis is unknown. We examined the independent relationships between coronary artery calcium (CAC), lipids including non-HDL, exercise, and diet among healthy male participants of the Prospective Army Coronary Calcium (PACC) Project.

METHODS: Male participants from the PACC Project (n = 1637, mean age 42.8 years; no history of coronary heart disease) were studied. We used validated surveys to measure dietary quality and habitual physical exercise. Fasting lipid concentrations and other cardiovascular risk variables …

Supplements and growth promotants containing steroid hormones are routinely administered to beef cattle to improve feeding efficiency, reduce behavioral problems, and enhance production. As a result, beef cattle manure will contain both synthetic steroids as well as a range of endogenous steroids including androgens, estrogens, and progestogens. A two-year controlled study was conducted in which beef cattle were administered steroid hormones via subcutaneous implants and feed additives and the occurrence of 16 endogenous and synthetic steroid hormones and metabolites was evaluated in runoff from beef cattle feedlots and in manure and soil collected from feedlot surfaces. We report on the ultrafast photoionization of pyridine, pyridazine, pyrimidine, and pyrazine. These four molecules represent a systematic series of perturbations into the structure of a benzene ring which explores the substitution of a C–H entity with a nitrogen atom, creating a heterocyclic structure. Data are recorded under intense-field, single-molecule conditions. The pulses (50 fs, 800 nm) are focused into the molecular vapor, and ion mass spectra are recorded for intensities of ∼1013 W/cm2 to ∼1015 W/cm2. In recent years, a voluminous amount of genomic data has been generated for several plant pathogenic fungi. Multiple studies have utilized these genomic data to advance our knowledge about the molecular mechanisms of plant pathogenesis. However, not all plant pathogenic fungi share the same infection strategies, and several genes have been identified that are crucial for plant pathogenesis in one fungus, but dispensable in others. Water flowing from forested catchments, both unmanaged and managed, is highly valued in terms of both quantity and quality. Increasing energy production using wood feedstocks produces varying degrees of impacts and thereby raises concerns about environmental impacts on the highly valued water supply resources of forest ecosystems. The term water supply encompasses both key components of water quantity and water quality. Water quantity considers the amount of increases or decreases, timing, consistency, and relative magnitude of water yields. Question: Seed banks often serve as reservoirs of taxonomic and genetic diversity that buffer plant populations and influence post-disturbance vegetation trajectories; yet evaluating their importance requires understanding how their composition varies within and across spatial and temporal scales (α- and β-diversity). Shifts in seed bank diversity are strongly governed by the deterministic role of differential seed longevities. Nevertheless, spatio-temporal variability in propagule depletion from and recruitment into seed banks may alter both α- and β-diversity. Here, we ask to what degree deterministic seed exhaustion, stochasticity in recruitment and mortality, or both, shape α- and β-diversity?

Invasive plant species threaten biological communities globally. However, relatively little is known about how evolutionary processes vary over the course of an invasion. To evaluate the importance of historical and adaptive drivers of range expansion, we compare the performance of North American populations of invasive Lonicera japonica from areas established 100–150 years ago, now the southern core of the range, to populations from the northern range margin, established within the last 65 years. Growth and survival of individuals from 17 core and 14 margin populations were compared in common gardens at both regions. After three years, margin plants were larger …

Nest predation is the primary factor influencing grassland songbird reproductive success. Understanding factors driving spatial and temporal variation in nest survival requires that we identify the primary nest predators and factors influencing predator abundance and behavior. Predation events are rarely witnessed, and the identification of nest predators is inferred, often incorrectly, from nest remains or observations of potential predators. We used video photography to identify predators of Sprague's Pipit (Anthus spragueii) nests in Saskatchewan and Montana. Predation is the primary source of nest mortality for most passerines; thus, behaviors to reduce the impacts of predation are frequently quantified to study learning, adaptation, and coevolution among predator and prey species. Video surveillance of nests has made it possible to examine real-time parental nest defense. During 1999-2009, we used video camera systems to monitor 518 nests of grassland birds. We reviewed video of 48 visits by snakes to 34 nests; 37 of these visits resulted in predation of active nests. When adult birds encountered snakes at the nest (n = 33 visits), 76% of the encounters resulted …

A synthesis of old and new paleoclimatic data from the Pyramid and Winnemucca lake basins indicates that, between 48.0 and 11.5∙103 calibrated years BP (hereafter ka), the climate of the western Great Basin was, to a degree, linked with the climate of the North Atlantic. Paleomagnetic secular variation (PSV) records from Pyramid Lake core PLC08-1 were tied to the GISP2 ice-core record via PSV matches to North Atlantic sediment cores whose isotopic and(or) carbonate records could be linked to the GISP2δ18O record. Aims: To evaluate the effectiveness of two spray-based decontamination methods for surface contamination reduction and to determine the potential for contamination spread by these methods.

Methods and Results: Material coupons (treated plywood and concrete) were contaminated with c. 1 × 107 spores of Bacillus atrophaeus by aerosol deposition. Decontaminants (pH-adjusted bleach or Spor-Klenz® RTU) were applied to coupons by either backpack sprayer or gas-powered sprayer. Contact time, reapplication frequency and rinse method were also varied. In addition to surface removal efficacy, partitioning of contamination between the rinsate and aerosol fractions was determined. Results indicated that pH-adjusted bleach …

In this demo, a novel underground communication system and an online underground sensor network testbed is demonstrated. The underground communication system, developed in the Cyber-physical Networking (CPN) Laboratory at the University of Nebraska-Lincoln, includes an underground antenna that is tailored to mitigate the adverse effects of soil on underground communication. An online connection is established with the CPN underground sensor network testbed that is located at Clay Center, Nebraska. The underground sensor network testbed consists of a network of underground communication systems equipped with soil moisture sensors and a mobile data harvesting unit equipped with cellular communication capabilities. Cannabis has been around for thousands of years and has been used recreationally, medicinally, and for fiber. Over 400 compounds have been isolated from Cannabis sativa with approximately 100 being cannabinoids. Of those 100 compounds, Δ9-THC has been determined as the primary constituent, which is also responsible for the psychoactivity associated with Cannabis. Along with Δ9-THC, cannabidiol has been studied in depth for its therapeutic effects. This has currently been the focus of many researchers since cannabidiol does not cause psychotropic effects.

Hot melt extrusion (HME) was evaluated as a processing technology for the manufacture of immediate as well as controlled release formulations for oral delivery of two model compounds. For immediate release applications lower molecular weight grades of Hydroxypropyl cellulose polymers, EF and ELF, were utilized as carrier matrices to form solid solutions of a poorly soluble compound, Ketoprofen (KPR). Thermal characterization techniques were used to confirm thermal stability, miscibility and setting up processing conditions for extrusion. Antheridiogen systems, whereby notch-bearing, archegoniate gametophytes induce maleness in ameristic neighbors, have been detected in many core leptosporangiate ferns. Previous studies have failed to detect an antheridiogen system in Osmundales, which is sister to all other extant leptosporangiates; hence, antheridiogen systems are thought to have evolved after their divergence. Detailed studies of morphological development and patterns of gender expression in Osmundales and other basal leptosporangiate clades are needed to clarify how antheridiogen systems evolved. How the greening of Arctic landscapes manifests as a change in ecosystem structure and function remains largely unknown. This study investigates the likely implications of plant community change on ecosystem function in tundra near Barrow, Alaska. We use structural data from marked plots, established in 1972 and resampled in 1999, 2008 and 2010 to assess plant community change. Knowledge of how arctic plant communities will respond to change has been largely derived from plot level experimental manipulation, not from trends of decade time scale environmental observations. This study documents plant community change in 330 marked plots at 33 sites established during the International Biological Program near Barrow, Alaska in 1972. Plots were resampled in 1999, 2008 and 2010 for species cover and presence. Cluster analysis identified nine plant communities in 1972. Rhododendrons are an important, dominant, and primitive group of flowering plants with considerable ecological and economic importance found in the temperate, subalpine and alpine regions of western Arunachal Pradesh, India. In addition to aesthetic, sacred, and ethnomedicinal values, several species have commercial and social importance. Rhododendrons are one of the preferred plant species used by local inhabitants in the region. Anthropogenic disturbance associated with deforestation, unsustainable extraction, over-exploitation, and agricultural practices have collectively put pressure on Rhododendron species; as a result many species have become endangered, rare, or threatened.
