Cytotoxic And Antibacterial Activities Of Leaf Extracts Of Astragalus Gombiformis Pomel (Fabaceae) Growing Wild In Tunisia, Hassen Teyeb, Nahla Zanina, Mohamed Neffati, Wahiba Douki, Mohamed Fadhel Najjar
Turkish Journal of Biology
Many species of the genus Astragalus have been long used in folk medicine because of their biological properties. The aim of this study was to investigate the antibacterial and cytotoxic activities of extracts from the leaves of wild Astragalus gombiformis Pomel. Antibacterial activity was assayed against several common human pathogenic bacterial strains using the paper disk-agar diffusion method. Cytotoxicity was measured against human lung carcinoma cells using the colorimetric MTT assay. The strongest cytotoxic activity against the human A549 lung epithelial carcinoma cell line was detected for dichloromethane extracts at IC_{50} = 85 ± 21.7 µg/mL. The best incubation time …

In tropical Bolivia coffee plantations, the plant community can be separated into high (trees), middle (coffee), and low (weed) strata. Understanding the importance of each stratum is critical for improving the sustainability of the system. The objective of this study was to determine the importance of strata on nutrient recycling. Litter falls from the upper and middle strata were collected monthly using cone-shaped traps and divided by species into leaves, branches, flowers, and fruits. Dry biomass additions to the soil from high and middle strata totaled 12,655 kg (ha yr)−1 annually. About 76% of the biomass was provided by …

The study herein determined the role of nuclear factor erythoid 2–related factor 2 (Nrf2) in the pathogenesis of hepatic steatosis, insulin resistance, obesity, and type 2 diabetes. Lepob/ob-Keap1-knockdown (KD) mice, which have increased Nrf2 activity, were generated. Markers of obesity and type 2 diabetes were measured in C57Bl/6J, Keap1-KD, Lepob/ob, and Lepob/ob-Keap1-KD mice. Lepob/ob-Keap1-KD mice exhibited less lipid accumulation, smaller adipocytes, decreased food intake, and reduced lipogenic gene expression. Enhanced Nrf2 activity impaired insulin signaling, prolonged hyperglycemia in response to glucose challenge, and induced insulin resistance in Lepob/ob background. Nrf2 augmented …

T cell-dependent development of anti-Factor VIII (FVIII) antibodies that neutralize FVIII activity is a major obstacle to replacement therapy in hemophilia A. To create a less immunogenic therapeutic protein, recombinant FVIII can be modified to reduce HLA binding of epitopes based on predicted anchoring residues. Here, we used immunoinformatic tools to identify C2 domain HLA DR epitopes and predict site-specific mutations that reduce immunogenicity. Epitope peptides corresponding to original and modified sequences were validated in HLA binding assays and in immunizations of hemophilic E16 mice, DR3 and DR4 mice and DR3 × E16 mice. Consistent with immunoinformatic predictions, original epitopes …

Babies born with Pompe disease require life-long treatment with enzyme-replacement therapy (ERT). Despite the human origin of the therapy, recombinant human lysosomal acid α glucosidase (GAA, rhGAA), ERT unfortunately leads to the development of high titers of anti-rhGAA antibody, decreased effectiveness of ERT, and a fatal outcome for a significant number of children who have Pompe disease. The severity of disease, anti-drug antibody (ADA) development, and the consequences thereof are directly related to the degree of the enzyme deficiency. Babies born with a complete deficiency GAA are said to have cross-reactive immunologic material (CRIM)–negative Pompe disease and are highly likely …

Development Of An Integrated Economic And Ecological Framework For Ecosystem-Based Fisheries Management In New England, D. Jin, P. Hoagland, Tracey Dalton, E. M. Thunberg
Marine Affairs Faculty Publications
We present an integrated economic-ecological framework designed to help assess the implementation of ecosystem-based fisheries management (EBFM) in New England. We develop the framework by linking a computable general equilibrium (CGE) model of a coastal economy to an end-to-end (E2E) model of a marine food web for Georges Bank. We focus on the New England region using coastal county economic data for a restricted set of industry sectors and marine ecological data for three top level trophic feeding guilds: planktivores, benthivores, and piscivores. We undertake numerical simulations to model the welfare effects of changes in alternative combinations of yields from …

Researchers measure biomarkers as a reflection of patient health status or intervention outcomes. While blood is generally regarded as the best body fluid for evaluation of systemic processes, substitution of saliva samples for blood would be less invasive and more convenient. The concentration of specific biomarkers may differ between blood and saliva. The objective of this study was to compare multiple biomarkers (27 cytokines) in plasma samples, passive drool saliva samples, and filter paper saliva samples in 50 healthy adults. Demographic data and three samples were obtained from each subject: saliva collected on filter paper over 1 minute, saliva collected …

Toothbrush Contamination: A Review Of The Literature, Michelle R. Frazelle, Cindy L. Munro
School of Nursing Publications
Toothbrushes are commonly used in hospital settings and may harbor potentially harmful microorganisms. A peer-reviewed literature review was conducted to evaluate the cumulative state of knowledge related to toothbrush contamination and its possible role in disease transmission. A systematic review was conducted on adult human subjects through three distinct searches. The review resulted in seven experimental and three descriptive studies which identified multiple concepts related to toothbrush contamination to include contamination, methods for decontamination, storage, design, and environmental factors. The selected studies found that toothbrushes of healthy and oral diseased adults become contaminated with pathogenic bacteria from the dental plaque, …

Background Ventilator-associated pneumonia (VAP) is a common infection in the intensive care unit (ICU) and associated with a high mortality.
Methods A quasi-experimental study was conducted in a medical-surgical ICU. Multiple interventions to optimize VAP prevention were performed from October 2008 to December 2010. All of these processes, including the Institute for Healthcare Improvement’s (IHI) ventilator bundle plus oral decontamination with chlorhexidine and continuous aspiration of subglottic secretions (CASS), were adopted for patients undergoing mechanical ventilation.
Results We evaluated a total of 21,984 patient-days, and a total of 6,052 ventilator-days (ventilator utilization rate of 0.27). We found VAP rates of …

Blockade Of Electron Transport At The Onset Of Reperfusion Decreases Cardiac Injury In Aged Hearts By Protecting The Inner Mitochondrial Membrane, Qun Chen, Thomas Ross, Ying Hu, Edward J. Lesnefsky
Internal Medicine Publications
Myocardial injury is increased in the aged heart following ischemia-reperfusion (ISC-REP) compared to adult hearts. Intervention at REP with ischemic postconditioning decreases injury in the adult heart by attenuating mitochondrial driven cell injury. Unfortunately, postconditioning is ineffective in aged hearts. Blockade of electron transport at the onset of REP with the reversible inhibitor amobarbital (AMO) decreases injury in adult hearts. We tested if AMO treatment at REP protects the aged heart via preservation of mitochondrial integrity. Buffer-perfused elderly Fischer 344 24 mo. rat hearts underwent 25 min global ISC and 30 min REP. AMO (2.5 mM) or vehicle was given …

Interactions Between Bile Acids And Nuclear Receptors And Their Effects On Lipid Metabolism And Liver Diseases, David Q.-H. Wang, Brent A. Neuschwander-Tetri, Piero Portincasa, William M. Pandak
Internal Medicine Publications
In this special issue of Journal of Lipids, we acknowledge the contributions by several experts offering timely perspectives on the complex interactions between bile acids and nuclear receptors (NRs) on lipid metabolism and liver diseases at different levels and contexts in the body.
NRs are found within the interior of cells and are defined as ligand-activated transcriptional regulators of several key aspects of body physiology and pathophysiology. NRs regulate gene transcription through interaction with cellular coactivators and corepressors. In the liver, NRs play a key role in a large variety of metabolic processes such as cholesterol, bile acid, fatty acid, …
