NAD(P)H:quinone oxidoreductase type I (NQO1) is a target enzyme for triggered delivery of drugs at inflamed tissue and tumor sites, particularly those that challenge traditional therapies. Prodrugs, macromolecules, and molecular assemblies possessing trigger groups that can be cleaved by environmental stimuli are vehicles with the potential to yield active drug only at prescribed sites. Furthermore, quinone propionic acids (QPAs) covalently attached to prodrugs or liposome surfaces can be removed by application of a reductive trigger stimulus, such as that from NQO1; their rates of reductive activation should be tunable via QPA structure. We explored in detail the recombinant human NAD(P)H:quinone …

We report on the synthesis and characterization of a PEGylated IR786 GUMBOS (Group of Uniform Materials Based on Organic Salts). The synthesis of this material was accomplished using a three step protocol: (1) substitution of chloride on the cyclohexenyl ring in the heptamethine chain of IR786 by 6-aminohexanoic acid, (2) grafting of methoxy polyethylene glycol (MeOPEG) onto the 6-aminohexanoic acid via an esterification reaction, and (3) anion exchange between [PEG786][I] and lithium bis(trifluoromethylsulfonyl)imide (LiNTf 2) or sodium bis(2-ethylhexyl)sulfosuccinate (AOT) in order to obtain PEG786 GUMBOS. Examination of spectroscopic data for this PEG786 GUMBOS indicates a large stokes shift (122 nm). …

Many polymers, such as PMMA, are very susceptible to swelling or dissolution by organic solvents. Growing covalently attached polymer brushes from these surfaces by atom-transfer radical polymerization (ATRP) is challenging because of the typical requirement of organic solvent for initiator immobilization. We report an unprecedented, aqueous-based route to graft poly(N-isopropylacrylamide), PNIPAAm, from poly(methyl methacrylate), PMMA, surfaces by ATRP, wherein the underlying PMMA is unaffected. Successful attachment of the ATRP initiator, N-hydroxysuccinimidyl-2-bromo-2- methylpropionate, on amine-bearing PMMA surfaces was confirmed by XPS. From this surface-immobilized initiator, thermoresponsive PNIPAAm brushes were grown by aqueous ATRP to yield optically transparent PNIPAAm-grafted PMMA surfaces. This …

Late-Life Body Mass Index And Dementia: An Integrative Literature Review, Sarah Slade, Patricia K. Ravert Oct 2012

Late-Life Body Mass Index And Dementia: An Integrative Literature Review, Sarah Slade, Patricia K. Ravert

Faculty Publications

Dementia and obesity are significant public health concerns. Alzheimer's disease affects 5.3 million adults, while 72.5 million adults are obese. Emerging evidence linking body mass index (BMI) and dementia suggest that, although a high BMI in midlife is associated with a greater risk for dementia, a high BMI in late life is considered protective and should not necessarily be considered a risk factor for dementia. Beginning in midlife, practitioners should trend patient BMI numbers. Major fluctuations during this time should be monitored and nutritional counseling and cognitive screenings offered to help patients maintain a healthy BMI and detect early cognitive …

The Role Of Micrornas In Breast Cancer Migration, Invasion And Metastasis, Joy Tang, Fazlul H. Sarkar, Aamir Ahmad

Honors College Theses

MicroRNAs (miRNAs) are a major class of small, noncoding RNA molecules that regulate gene expression by targeting mRNAs to trigger either translational repression or mRNA degradation. They have recently been more widely investigated due to their potential role as targets for cancer therapy. Many miRNAs have been implicated in several human cancers, including breast cancer. miRNAs are known to regulate cell cycle and development, and thus may serve as useful targets for exploration in anticancer therapeutics. The link between altered miRNA signatures and breast cancer development and metastasis can be observed either through the loss of tumor suppressor miRNAs, such …

School Food Environment The Frontline For Childhood Obesity Prevention: A Mixed-Method Study Of Nutritional Competencies And Skills Of School Nutrition Professionals In Nebraska, Zainab Rida

Department of Nutrition and Health Sciences: Dissertations, Theses, and Student Research

The purpose of this statewide study was to address the nutrition knowledge, attitudes, and perceptions of school foodservice personnel in Nebraska regarding offering/serving healthy school meals. Moreover, this study identified some potential barriers and avenues of action for offering/serving healthy school meals. Data obtained from this study indicates that there is a strong correlation (r= .103, p r .237, p< .01) between the foodservice staff’s self-efficacy and their practices of offering/serving healthy school meals. Fortunately, the relationship between foodservice staff practices of offering/serving healthy school meals and their self-efficacy was positive and significantly predicted practices scores, β =.237, P< 0.01. The finding of the present study also identifies many barriers including lack of time and support that face the foodservice personnel in offering/serving healthy school meals. The findings suggested that there is an urgent need of a full school approach to promote and encourage healthy eating habits among students. Future research is needed to evaluate school wellness policies regarding healthy eating practices in schools. Moreover, establish partnerships with communities and universities for intervention that target students and their parents.

Dartmouth Scholarship

The antiapoptotic Bcl-2 family member Mcl-1 is a PEST protein (containing sequences enriched in proline, glutamic acid, serine, and threonine) and is subject to rapid degradation via multiple pathways. Impaired degradation leading to the maintenance of Mcl-1 expression is an important determinant of drug resistance in cancer. Phosphorylation at Thr 163 in the PEST region, stimulated by 12-O-tetradecanoylphorbol acetic acid (TPA)-induced activation of extracellular signal-regulated kinase (ERK), is associated with Mcl-1 stabilization in BL41-3 Burkitt lymphoma cells. This contrasts with the observation that Thr 163 phosphorylation in normal fibroblasts primes glycogen synthase kinase (GSK3)-induced phosphorylation at Ser 159, producing a …

Racial And Ethnic Disparities In Food Insufficiency: Evidence From A Statewide Probability Sample Of White, African American, American Indian, And Hispanic Infants, Yunju Nam, Jin Huang, Colleen Heflin, Michael Sherraden

Center for Social Development Research

This study examines racial and ethnic disparities in the experience of food insufficiency among families with infants, focusing on the roles of socioeconomic characteristics. It uses the SEED for Oklahoma Kids baseline survey data collected from a probability sample of white, African American, American Indian, and Hispanic caregivers of infants randomly selected from Oklahoma’s birth certificates. Fairlie’s extension of the Blinder-Oaxaca decomposition is employed to analyze these data. Results suggest that whites experience food insufficiency at a statistically significantly lower rate than do the three minority groups. Compositional gaps in economic and noneconomic resources are found to explain much of …

Oncology Faculty Publications

There is a large body of scientific evidence suggesting that 3,39-Diindolylmethane (DIM), a compound derived from the digestion of indole-3-carbinol, which is abundant in cruciferous vegetables, harbors anti-tumor activity in vitro and in vivo. Accumulating evidence suggests that AMP-activated protein kinase (AMPK) plays an essential role in cellular energy homeostasis and tumor development and that targeting AMPK may be a promising therapeutic option for cancer treatment in the clinic. We previously reported that a formulated DIM (BR-DIM; hereafter referred as B-DIM) with higher bioavailability was able to induce apoptosis and inhibit cell growth, angiogenesis, and invasion of prostate cancer cells. …

Neutralizing Antibodies And Pathogenesis Of Hepatitis C Virus Infection, Samira Fafi-Kremer, Catherine Fauvelle, Daniel J. Felmlee, Mirjam B. Zeisel, Quentin Lepiller, Isabel Fofana, Laura Heydmann, Françoise Stoll-Keller, Thomas F. Baumert

Peninsula Medical School

Hepatitis C virus (HCV) infection is a major cause of chronic liver disease worldwide. The interplay between the virus and host innate and adaptive immune responses determines the outcome of infection. There is increasing evidence that host neutralizing responses play a relevant role in the resulting pathogenesis. Furthermore, viral evasion from host neutralizing antibodies has been revealed to be an important contributor in leading both to viral persistence in acute liver graft infection following liver transplantation, and to chronic viral infection. The development of novel model systems to study HCV entry and neutralization has allowed a detailed understanding of the …

Head Start: A True Start To Getting Ahead: A Literature Review Of The Head Start Program As A Primary Poverty Prevention Strategy, Laporsche C. Smith Oct 2012

Head Start: A True Start To Getting Ahead: A Literature Review Of The Head Start Program As A Primary Poverty Prevention Strategy, Laporsche C. Smith

McNair Scholars Research Journal

The federal Head Start Program plays a vital role in increasing the likelihood that under-privileged children will become academically successful in school, attend college, and become successful citizens. Yet, Head Start continuously struggles to retain its funding and is challenged in its goal of assisting low income children. A review of the literature on Head Start and related early childhood education longitudinal studies will attest to the importance of this program as a primary prevention strategy to reduce poverty, raise public awareness of its benefits, and support the efforts of its advocates to retain and expand its funding at local …

Non-Suicidal Self-Injury: The Movie Industry's Influence On Its Stigma, Courtney M. Raymond Oct 2012

Non-Suicidal Self-Injury: The Movie Industry's Influence On Its Stigma, Courtney M. Raymond

McNair Scholars Research Journal

Non-Suicidal Self-Injury (NSSI) is a concerning behavior disproportionately affecting adolescents today. Rates of NSSI have increased, as have depictions of NSSI in the media. Therefore, some researchers believe that increased media exposure is contributing to increased rates of NSSI. Research has shown that NSSI is a coping mechanism and/or a cry for help among those who display such behaviors. However, studies also show that many adolescents with this behavior do not seek help. This may be because persons engaging in NSSI feel that their behavior, thoughts, or feelings are stigmatized by the general population. The purpose of this study is …

A Literature Review: How Language Parameter Deficits Impact Social Interaction In Children With Autism Spectrum Disorders During Middle Childhood, And Intervention Strategies, Carmen Nicole Grange

McNair Scholars Research Journal

Human growth is dependent upon social interactions, notably essential during the middle childhood years (ages 5-10). Although some children develop differently than others, all experience the same types of mental processes through which, due to a variety of factors, they may become typically achieving, or stunted in their development. Language is a key component of being human. Communication occurs through verbal and nonverbal behavior. People are able to understand what others are trying to encode through multiple processes of interpretation. Humans learn to use language through interaction with others, but are born with the ability to acquire language. Children with …

An Exploration Into The Reality Of Media Perceptions: The Depictions Of Women Public Relations Practitioners In Reality Television, Cherese E. Colston

McNair Scholars Research Journal

Reality television is a media staple for this generation. Reality programs including Jersey Shore, Keeping Up with the Kardashians, and Teen Mom 2 are among the most watched programs in the United States. In the last five years, reality television has highlighted professions including Public Relations. However, these new reality shows depict an unrealistic image of female practitioners in the PR field. These negative portrayals have been analyzed in film, yet there is a lack of research in regards to reality television. The purpose of this study is to analyze the portrayal of women in the Public Relations …

The Extent Of Political Participation In The United States Among Latino Non-Citizens And Citizens, Shanilinin M. Calderon Oct 2012

The Extent Of Political Participation In The United States Among Latino Non-Citizens And Citizens, Shanilinin M. Calderon

McNair Scholars Research Journal

In the United States, Hispanics, or Latino Americans, are individuals with Mexican, Puerto Rican, Cuban, or other Latin American origin. Undocumented and documented Latinos are two of the fastest growing populations in the United States. However, this demographic is still underrepresented in American politics today. The increase in this growing population is due to “push and pull” factors that attract immigrants; some of these factors include escaping from poverty in search for a better life, and better opportunities for employment. Factors contributing to lower voting turnouts include the belief that the Latino vote will not make a difference, mistrust in …
