Voltage-dependent potassium (Kv) channels provide the repolarizing power that shapes the action potential duration and helps control the firing frequency of neurons. The K(+) permeation through the channel pore is controlled by an intracellularly located bundle-crossing (BC) gate that communicates with the voltage-sensing domains (VSDs). During prolonged membrane depolarizations, most Kv channels display C-type inactivation that halts K(+) conduction through constriction of the K(+) selectivity filter. Besides triggering C-type inactivation, we show that in Shaker and Kv1.2 channels (expressed in Xenopus laevis oocytes), prolonged membrane depolarizations also slow down the kinetics of VSD deactivation and BC gate closure during the …

In voltage-sensitive phosphatases (VSPs), a transmembrane voltage sensor domain (VSD) controls an intracellular phosphoinositide phosphatase domain, thereby enabling immediate initiation of intracellular signals by membrane depolarization. The existence of such a mechanism in mammals has remained elusive, despite the presence of VSP-homologous proteins in mammalian cells, in particular in sperm precursor cells. Here we demonstrate activation of a human VSP (hVSP1/TPIP) by an intramolecular switch. By engineering a chimeric hVSP1 with enhanced plasma membrane targeting containing the VSD of a prototypic invertebrate VSP, we show that hVSP1 is a phosphoinositide-5-phosphatase whose predominant substrate is PI(4,5)P(2). In the chimera, enzymatic activity …

Using Grounded Theory To Reveal Tacit Knowledge For Nurse Educators In An Unfamiliar African Setting, Ramona B. Patterson
Using Grounded Theory To Reveal Tacit Knowledge For Nurse Educators In An Unfamiliar African Setting, Ramona B. Patterson
Theses and Dissertations (ETD)
There is a gap in evidence to guide individuals as they prepare for a nursing education partnership in an unfamiliar setting. Much of the existing knowledge base is contained within institutional or organizational confines. Individuals new to the field need access to existing expertise and tacit knowledge. The purpose of this research was to generate a grounded theory to guide aspiring nursing faculty in developing the necessary knowledge base to fulfill the nurse educator role in an unfamiliar setting. Ghana, Africa, was specified as the unfamiliar setting and context for this study.
Purposive sampling identified individuals from North America and …

Cross Regulation Of Sirtuin 1, Ampk, And Ppary In Conjugated Linoleic Acid Treated Adipocytes, Shan Jiang, Wei Wang, Jess Miner, Michael E. Fromm
Department of Biochemistry: Faculty Publications
Trans-10, cis-12 conjugated linoleic acid (t10c12 CLA) reduces triglyceride (TG) levels in adipocytes through multiple pathways, with AMP-activated protein kinase (AMPK) generally facilitating, and peroxisome proliferator-activated receptor γ (PPARγ) generally opposing these reductions. Sirtuin 1 (SIRT1), a histone/protein deacetylase that affects energy homeostasis, often functions coordinately with AMPK, and is capable of binding to PPARγ, thereby inhibiting its activity. This study investigated the role of SIRT1 in the response of 3T3-L1 adipocytes to t10c12 CLA by testing the following hypotheses: 1) SIRT1 is functionally required for robust TG reduction; and 2) SIRT1, …

The majority of bird species co-express two functionally distinct hemoglobin (Hb) isoforms in definitive erythrocytes as follows: HbA (the major adult Hb isoform, with α-chain subunits encoded by the αA-globin gene) and HbD (the minor adult Hb isoform, with α-chain subunits encoded by the αD-globin gene). The αD-globin gene originated via tandem duplication of an embryonic α-like globin gene in the stem lineage of tetrapod vertebrates, which suggests the possibility that functional differentiation between the HbA and HbD isoforms may be attributable to a retained ancestral character state in HbD that harkens back to …

Jean-Baptiste Étienne, C.M. And The Restoration Of The Daughters Of Charity, Edward R. Udovic C.M., Ph.D.
Vincentian Studies
The nineteenth-century superior general Jean-Baptiste Étienne has often been given the title of “Second Founder” of the Congregation and the Daughters of Charity. Edward Udovic argues that this title is deserved, not because of any similarity to Vincent de Paul, but because of Étienne’s faithfulness to the communities’ primitive spirit. Étienne made that the guiding principle of the communities’ re-establishment. His background, experience, agenda for restoration and reform, and worldview are all examined. According to Udovic, Étienne and his leadership are best described as “Vincentian-centric, Romantic, Gallican, and authoritarian.” A French nationalist and imperialist, he was particularly concerned with remaining …

The Era Of Boundlessness At St. Mary’S Of The Barrens, 1818-1843: A Brief Historical Analysis, Richard J. Janet Ph.D.
Vincentian Studies
Richard Janet analyzes the early history of Saint Mary’s of the Barrens according to the historian Philip Gleason’s view of the development of American Catholicism. Gleason sees the arc of American Catholicism as moving from a period of “boundlessness,” in which institutions engaged in all types of works, to the mid-nineteenth-century period of “consolidation,” in which leaders tried to manage growth in a way that would stabilize those institutions. Gleason’s thesis is discussed in detail. The work of Felix De Andreis, Joseph Rosati, and John Timon is examined. They faced many problems inherent in maintaining a Vincentian community while ministering …

Measuring Parvalbumin Levels In Fish Muscle Tissue: Relevance Of Muscle Locations And Storage Conditions, Poi-Wah Lee, Julie A. Nordlee, Stef J. Koppelman, Joseph L. Baumert, Steve L. Taylor
Department of Food Science and Technology: Faculty Publications
Fish is an allergenic food capable of provoking severe anaphylactic reactions. Parvalbumin is the major allergen identified in fish and frog muscles. Antibodies against fish and frog parvalbumin have been used to quantify parvalbumin levels from fish. However, these antibodies react variably with parvalbumin from different fish species. Several factors might be responsible for this variation including instability of parvalbumin in fish muscle as a result of frozen storage and differential parvalbumin expression in muscles from various locations within the whole fish. We aimed to investigate whether these factors contribute to the previously observed variable immunoreactivity of the anti-parvalbumin antibodies. …

Distribution And Quantification Of Antibiotic Resistant Genes And Bacteria Across Agricultural And Non- Agricultural Metagenomes, Lisa M. Durso, Daniel N. Miller, Brian J. Wienhold
United States Department of Agriculture-Agricultural Research Service / University of Nebraska-Lincoln: Faculty Publications
There is concern that antibiotic resistance can potentially be transferred from animals to humans through the food chain. The relationship between specific antibiotic resistant bacteria and the genes they carry remains to be described. Few details are known about the ecology of antibiotic resistant genes and bacteria in food production systems, or how antibiotic resistance genes in food animals compare to antibiotic resistance genes in other ecosystems. Here we report the distribution of antibiotic resistant genes in publicly available agricultural and non-agricultural metagenomic samples and identify which bacteria are likely to be carrying those genes. Antibiotic resistance, as coded for …
Using Dual Tasks To Test Immediate Transfer Of Training Between Naturalistic Movements: A Proof-Of-Principle Study, Sydney Y. Schaefer, Catherine E. Lang
Using Dual Tasks To Test Immediate Transfer Of Training Between Naturalistic Movements: A Proof-Of-Principle Study, Sydney Y. Schaefer, Catherine E. Lang
Kinesiology and Health Science Faculty Publications
Theories of motor learning predict that training a movement reduces the amount of attention needed for its performance (i.e. more automatic). If training one movement transfers, then the amount of attention needed for performing a second movement should also be reduced, as measured under dual task conditions. The purpose of this study was to test whether dual task paradigms are feasible for detecting transfer of training between two naturalistic movements. Immediately following motor training, subjects improved performance of a second untrained movement under both single and dual task conditions. Subjects with no training did not. Improved performance in the untrained …
