Opportunistic fungal infections have become problematic in recent years. There has been an emergence of new fungal pathogens, development of antifungal resistance, and increasing prevalence. In addition, toxicity, resistance, and lack of efficacy as a single agent limit the effectiveness of current antifungal drugs. Therefore, new drugs targeting new pathways are greatly needed. An analog derived from the marine natural product phloeodictine, analog PHL5-34A, was identified as having strong in vitro antifungal activity. In this study, its mechanism of action (MOA) was investigated in the model yeast, Saccharomyces cerevisiae. This study made use of a collection of 166 yeast mutants …

The obesity epidemic is a social issue in that critics are quick to blame business and government for failing the consumers. The truth is that all three parties have failed each other by allowing such a cultural change to occur. The issue is similar to the tobacco public health crisis following the spike in lung cancer in the 1950s and 60s. The disease Systemic Lupus Erythematosus is a chronic inflammatory, autoimmune disorder that primarily affects women during their reproductive years. Women with SLE are at a greater risk of developing hypertension, which increases their risk of mortality from a cardiac related event. A proposed mechanism for SLE hypertension suggests that inflammation in the kidneys causes renal dysfunction, presumably resulting in less water excretion, increased plasma volume, and thus high blood pressure. This experiment tests the hypothesis that the T-cells from a mouse model of SLE hyper secrete the inflammatory cytokines IL-17, IL-10, and DFN- y, and are more sensitive to the …

Within a population, susceptibility to parasitism may vary. Individuals possessing certain intrinsic or extrinsic traits are often more likely to be parasitized than individuals within that population that do not possess those certain traits. As a result, parasites can have profound effects of the population dynamics of theirs hosts. Much research has been done on the host-parasite interaction of the eastern grey squirrel {Sciurus caroUnensis) and the tree squirrel hot fly {Cuterebra emasculator). During 4 weeks of the spring of 2011 and 5 weeks of the fall of 2011, interviews were conducted with medical professionals practicing in Mississippi regarding the diagnosis of brown recluse {Loxosceles reclusa) and black widow {Latrodectus mactans) spider bites. During the spring, a total of 19 interviews were held with emergency room physicians and dermatologists practicing in urban and rural, non-medically underserved areas of Mississippi. This study defines a critical role for Btk in regulating TLR4-induced crosstalk between antigen presenting cells (APCs) and natural killer (NK) cells. Reduced levels of IL-12, IL- 18 and IFN-γ were observed in Btk-deficient mice and ex vivo generated macrophages and dendritic cells (DCs) following acute LPS administration, whilst enhanced IL-10 production was observed. In addition, upregulation of activation markers and antigen presentation molecules on APCs was also impaired in the absence of Btk. APCs, by virtue of their ability to produce IL-12 and IL-18, are strong inducers of NK-derived IFN-γ. Co-culture experiments demonstrate that Btk-deficient DCs were unable to …

Brain cholesterol homeostasis has been shown to be disrupted in neurodegenerative conditions such as Alzheimer's and Huntington's diseases. Investigations in animal models of seizure-induced brain injury suggest that brain cholesterol levels are altered by prolonged seizures (status epilepticus) and are a feature of the pathophysiology of temporal lobe epilepsy. The present study measured hippocampal sterol levels in a model of unilateral hippocampal injury triggered by focal-onset status epilepticus, and in chronically epileptic mice. Status epilepticus was induced by intra-amygdala microinjection of kainic acid and ipsilateral and contralateral hippocampus analyzed. The most commonly used test for monitoring heparin therapy is the activated partial thromboplastin time (aPTT). The response of available aPTT reagents to heparin varies significantly. The aim of this study was to highlight the differences between aPTT reagents stored in a dried format to select the most suitable formulations to be used for the development of point-of-care diagnostic devices used for monitoring of unfractionated heparin dose response.

The authors offer two arguments for the inclusion of theism in natural science. First, an argument against excluding theism is offered. Though early roots of science promoted a view that it is a way to accumulate knowledge that is untainted by presuppositions and traditions, postmodern critiques call this into question. Scientists have sometimes rejected religion as a context-dependent, tradition-based way of knowing, yet science itself is also context-dependent and tradition-based. This chapter begins with general considerations for religiously and spiritually oriented psychotherapy, including the importance of seeing religion and spirituality as d.imensions of cultural diversity, considering clients' welfare and autonomy, and maintaining competence. Three types of religious and spiritual intervention approaches are then discussed: assimilative, accommodative, and collaborative. Assimilative approaches introduce spiritual interventions or considerations into a standard psychotherapy approach. Accommodative approaches involve adapting a standard psychotherapy regimen to include religious or spiritual matters. Second Step: A Violence Prevention Curriculum is a school-based program developed to prevent and reduce aggressive behavior. Three separate age-appropriate curricula are available: preschool/kindergarten level, elementary level, and middle school level. Designed to be teacher-friendly and convenient for classroom use, the curricula aim to prevent and decrease aggression by helping children develop habits of prosocial behavior and thought. Instructional strategies are evidence-based and are built on a broad foundation of research. Western literature provides an array of information regarding resilience within at-risk youth. Resilience research within non-Western contexts, and more specifically with exploited youth, is more limited. Despite exploitation, some youth develop a hardy ability to overcome adversity, allowing them more mastery over their environments and even increased psychological steadfastness. This project involved exploring the protective factors of resilience and psychological functioning in Cambodian youth, specifically a group of 24 survivors of sexual trafficking and another group of 24 rural youth without reported exploitation. Parents with severe mental illness are at greater risk than others of becoming involved in the child protection system, and their cases are more likely than others to result in termination of parental rights. Among women with severe mental illness, 26-75% lose custody to one or more of their children, rates far higher than for women without mental illness. Lawyers who represent mentally ill parents in child protection matters face a number of challenges, including maintaining a productive attorney-client relationship, advocating for appropriate services and reasonable accommodations for their clients’ disabilities, and refuting assumptions about their clients’ parenting abilities that …

Assets church members believed they needed to engage in effective HIV/AIDS prevention and control activities. We used the three-step forum focus group discussion (FFGD) methodology to elicit responses from 32 church leaders and lay members, representing five denominations in Aba, Nigeria. Concrete resources, health expertise, finances, institutional support, capacity building, and spiritual support connected to the collective interest of members were indicated as useful for church members to engage in HIV/AIDS prevention and control activities. Adequate planning and delivery of cost-effective, appropriate and sustainable health promotion programs require an understanding of perceived church-based assets.

This autoethnography is a constructed account of a co-exploration into the nature and effects of a longitudinal dyadic conversation process from a relational constructionist perspective. The conversations, between me as participant autoethnographer and a co-participant, aimed at maximising personal learning for both. Through co-created contexts of mutual engagement and respectful presence, we were able to focus our learning on the spontaneous process and content of the conversations. The purpose of this paper is to show how “realist synthesis” methodology (Pawson, 2002) was adapted to review a large sample of community based projects addressing alcohol and drug use problems. Our study drew on a highly varied sample of 127 projects receiving funding from a national non-government organisation in Australia between 2002 and 2008. Open and pattern coding led to the identification of 10 barrier and nine enabler mechanisms influencing project implementation across the sample. Eight case studies (four demonstrating successful implementation; four demonstrating less than successful implementation) were used for depth exploration of these mechanisms. High level theories …

Bereavement has been extensively studied over the years, yet scholarly work depicting, with the first-person perspective, the experience of childhood bereavement is severely lacking. The research question I set out to answer here is: What is it like as an Asian child to experience bereavement following grandparental death? As such, self-introspection was exercised, and this, together with the diaries and free writings generated at the time of my grandma’s death, was used as the basis for autoethnographic reflections.
