Investigation Into The Mechanism Of Salicylate-Associated Genotypic Antibiotic Resistance In Staphylococcus Aureus, Nada Salah Helal
Investigation Into The Mechanism Of Salicylate-Associated Genotypic Antibiotic Resistance In Staphylococcus Aureus, Nada Salah Helal
USF Tampa Graduate Theses and Dissertations
Growth of Staphylococcus aureus with the NSAID salicylate increases phenotypic resistance (SAPAR), and the frequency at which heritable resistance occurs to various antibiotics (SAGAR). This study describes the effect of salicylate on heritable and phenotypic resistance to a set of antibiotics for laboratory and multi-drug resistant strains of S. aureus and investigates the link between resistance and SAGAR. Drug gradient plates were used to determine phenotypic resistance to antibiotics targeting DNA replication, transcription, translation and the cell wall in the presence or absence of salicylate. To measure heritable resistance, mutation frequencies were determined for each antibiotic in the presence and …

The research project is centered on determining the evolving meaning of the term “polytechnic”. As a standard class assignment, freshman students will be required to execute video interviews of upper classmen to find out what they think “polytechnic” means. This assignment will be repeated with each group of incoming freshmen so that all students will have the opportunity to interview and be interviewed. By interviewing the students each year, the researchers will be able to understand the changing definition of the term “polytechnic” as the students progress through school. By comparing responses from differing freshmen classes, the researchers will be …

Transforming Growth Factor-Β Suppresses Metastasis In A Subset Of Human Colon Carcinoma Cells., Neka A.K. Simms, Ashwani Rajput, Elizabeth A. Sharratt, Melanie Ongchin, Carol A. Teggart, J. Wang, Michael G. Brattain
Journal Articles: Eppley Institute
BACKGROUND: TGFβ signaling has typically been associated with suppression of tumor initiation while the role it plays in metastasis is generally associated with progression of malignancy. However, we present evidence here for an anti-metastatic role of TGFβ signaling.
METHODS: To test the importance of TGFβ signaling to cell survival and metastasis we compared human colon carcinoma cell lines that are either non-tumorigenic with TGFβ response (FET), or tumorigenic with TGFβ response (FETα) or tumorigenic with abrogated TGFβ response via introduction of dominant negative TGFβRII (FETα/DN) and their ability to metastasize. Metastatic competency was assessed by orthotopic transplantation. Metastatic colony formation …

Bacteriophages Phi Mr299-2 And Phi Nh-4 Can Eliminate Pseudomonas Aeruginosa In The Murine Lung And On Cystic Fibrosis Lung Airway Cells, Aidan Coffey, Et. Al.
Department of Biological Sciences Publications
Pseudomonas aeruginosa is a common cause of infection in the lungs of patients with cystic fibrosis (CF). In addition, biofilm formation and antibiotic resistance of Pseudomonas are major problems that can complicate antibiotic therapy. We evaluated the efficacy of using bacteriophages to kill the pathogen in both biofilms and in the murine lung. We isolated and characterized two phages from a local wastewater treatment plant, a myovirus (ϕNH-4) and a podovirus (ϕMR299-2). Both phages were active against clinical isolates of P. aeruginosa. Together, the two phages killed all 9 clinical isolate strains tested, including both mucoid and nonmucoid strains. An …
Towards A Better Specification Of The Store Environment Stimulus: An Augmented Stimulus-Organism-Response (Sor) Model That Captures Brand Expressiveness, John Murray
Conference proceedings
Abstract: This paper proposes an augmented SOR model which facilitates design/architect practitioners when they review store concepts. The paper contributes to the knowledge base of designers/architects when making deliberate brand expressions in development of the store environment. The global nature of the SOR model, it is argued, does not allow for discrimination between consumer interpretations of store brands; nor does it propose a realistic means of engaging design-architect practitioners at the concept proofing stage of development. This conceptual paper argues that retail branding studies benefit from inclusion of more flexible frameworks founded on separable and integral design-architecture and brand communicative …

An Investigation Of Nanos1 Function During Primordial Germ Cell Development In Xenopus Laevis., Fangfang Lai
An Investigation Of Nanos1 Function During Primordial Germ Cell Development In Xenopus Laevis., Fangfang Lai
Open Access Dissertations
Nanos is expressed in multipotent cells, stem cells, and primordial germ cells (PGCs) of organisms as diverse as jellyfish and humans. It functions together with Pumilio to translationally repress targeted mRNAs. Here we show by loss-of-function experiments that Xenopus Nanos1 is required to preserve PGC fate. Morpholino knockdown of maternal Nanos1 resulted in a striking decrease in PGCs and loss of germ cells from the gonads. Lineage tracing and TUNEL staining reveals that Nanos1 deficient PGCs fail to migrate out of the endoderm. They appear to undergo apoptosis rather than convert to normal endoderm. Whereas normal PGCs do not become …

The Pseudomonas Syringae Hrpj Protein Controls The Secretion Of Type Iii Translocator Proteins And Has A Virulence Role Inside Plant Cells, Emerson Crabill, Andrew Karpisek, James R. Alfano
Center for Plant Science Innovation: Faculty Publications
The bacterial plant pathogen Pseudomonas syringae injects effector proteins into plant cells via a type III secretion system (T3SS), which is required for pathogenesis. The protein HrpJ is secreted by P. syringae and is required for a fully functional T3SS. A hrpJ mutant is non-pathogenic and cannot inject effectors into plant cells or secrete the harpin HrpZ1. Here we show that the hrpJ mutant also cannot secrete the harpins HrpW1 and HopAK1 or the translocator HrpK1, suggesting that these proteins are required in the translocation (injection) of effectors into plant cells. Complementation of the hrpJ mutant with secretion incompetent HrpJ …

High-Activity Mutants Of Butyrylcholinesterase For Cocaine Hydrolysis And Method Of Generating The Same, Chang-Guo Zhan, Hoon Cho, Hsin-Hsiung Tai
Pharmaceutical Sciences Faculty Patents
A novel computational method and generation of mutant butyrylcholinesterase for cocaine hydrolysis is provided. The method includes molecular modeling a possible BChE mutant and conducting molecular dynamics simulations and hybrid quantum mechanical/molecular mechanical calculations thereby providing a screening method of possible BChE mutants by predicting which mutant will lead to a more stable transition state for a rate determining step. Site-directed mutagenesis, protein expression, and protein activity is conducted for mutants determined computationally as being good candidates for possible BChE mutants, i.e., ones predicted to have higher catalytic efficiency as compared with wild-type BChE. In addition, mutants A199S/A328W/Y332G, A199S/F227A/A328W/Y332G, A199S/S287G/A328W/Y332G, …

Human Impact On Atolls Leads To Coral Loss And Community Homogenisation: A Modeling Study, Bernhard Riegl, Charles R. C. Sheppard, Samuel J. Purkis
Marine & Environmental Sciences Faculty Articles
We explore impacts on pristine atolls subjected to anthropogenic near-field (human habitation) and far-field (climate and environmental change) pressure. Using literature data of human impacts on reefs, we parameterize forecast models to evaluate trajectories in coral cover under impact scenarios that primarily act via recruitment and increased mortality of larger corals. From surveys across the Chagos, we investigate the regeneration dynamics of coral populations distant from human habitation after natural disturbances. Using a size-based mathematical model based on a time-series of coral community and population data from 1999–2006, we provide hind- and forecast data for coral population dynamics within lagoons …

"Nevermericans?": How Communication Issues Shape The Perceptions Of Self And The Perceptions Of American Identity Among The International Students, Lyudmyla Pustelnyk
Masters Theses
This study of Intercultural communication and identity uses Cultural Contracts theory and Narrative theory to explore how international students communicate about their understanding of self (identity) and how this understanding is influenced and changes during their studies in the United States. Research participants who have, or are currently studying in the U.S., from Eastern and Central European countries were interviewed about their communication experiences while in the U.S., resulting in different expressions of identity - in-between identity, feeling Americanized, global citizen, and crystallization of native identity - which developed as the result of their U.S. university studies. Their narratives also …
