Domestic animals are unique in that they have been organised into managed populations called breeds. The strength of genetic divergence between breeds may vary dependent on the age of the breed, the scenario under which it emerged and the strength of reproductive isolation it has from other breeds. In this study, we investigated the Gulf Coast Native breed of sheep to determine if it contains lines of animals that are sufficiently divergent to be considered separate breeds. Allele sharing and principal component analysis (PCA) using nearly 50,000 SNP loci revealed a clear genetic division that corresponded with membership of either …

Bait Effects In Sampling Coral Reef Fish Assemblages With Stereo-Bruvs, Stacey R. Dorman, Euan S. Harvey, Stephen J. Newman
Fisheries Research Articles
Baited underwater video techniques are increasingly being utilised for assessing and monitoring demersal fishes because they are: 1) non extractive, 2) can be used to sample across multiple habitats and depths, 3) are cost effective, 4) sample a broader range of species than many other techniques, 5) and with greater statistical power. However, an examination of the literature demonstrates that a range of different bait types are being used. The use of different types of bait can create an additional source of variability in sampling programs. Coral reef fish assemblages at the Houtman Abrolhos Islands, Western Australia, were sampled using …

Dartmouth Scholarship
Pseudomonas aeruginosa secretes an epoxide hydrolase virulence factor that reduces the apical membrane expression of ABC transporters such as the cystic fibrosis transmembrane conductance regulator (CFTR). This virulence factor, named CFTR inhibitory factor (Cif), is regulated by a TetR-family, epoxide-responsive repressor known as CifR via direct binding and repression. We identified two sites of CifR binding in the intergenic space between cifR and morB, the first gene in the operon containing the cif gene. We have mapped these binding sites and found they are 27 bp in length, and they overlap the -10 and +1 sites of both the cifR …
Towards Understanding The Mechanism Of Action Of Abused Cathinones, Rakesh Vekariya
Towards Understanding The Mechanism Of Action Of Abused Cathinones, Rakesh Vekariya
Theses and Dissertations
The dopamine transporter (DAT) mediates reuptake of dopamine from the synaptic cleft into the presynaptic terminus and plays a critical role in maintaining the normal function of dopaminergic neurons. DAT is the major target of widely abused psychostimulant drugs, including cocaine and amphetamine. DAT also figures into disease states, and it is a target for therapeutic drugs. It is known that cathinone and methcathinone, β-keto analogs of amphetamine and methamphetamine, respectively, produce pharmacological actions similar to amphetamine. Cathinone and methcathinone analogs are recently gaining in popularity on the clandestine market (e.g. ‘bath salts’). Cathinone and methcathinone analogs as well as …

Development Of Magnetic Fabrics With Tunable Hydrophobicity, Thu Ho
Theses and Dissertations
Polystyrene (PS) fiber mats incorporating iron (Fe) particles were fabricated by electrospinning and the hydrophobicity of the resulting magnetic fabrics was investigated with and without an applied magnetic field. The results show that the hydrophobicity (as measured using water droplet contact angle) increases in the presence of a magnetic field and the hysterisis in the advancing/receding contact angle (a measure of the stickiness of the surface) decreases in the presence of a magnetic field. It is also shown that the contact angle and hysterises increase with decreasing fiber diameter and mat thickness.

Rethinking Children As Property, Kevin Noble Maillard
Rethinking Children As Property, Kevin Noble Maillard
College of Law - Faculty Scholarship
Despite the collective view in law and social practice that it is intrinsically taboo to consider human beings as chattel, the law persists in treating children as property. Applying principles of property, this Article examines paternity disputes to explain and critique the law’s view of children as property of their parents. As evidenced in these conflicts, I demonstrate that legal paternity exposes a rhetoric of ownership, possession, and exchange. The law presumes that a child born to a married woman is fathered by her husband, even when irrefutable proof exists that another man fathered the child. Attempts by the non-marital …

Wills that prioritize the interests of nontraditional families over collateral heirs test courts’ dedication to observing the posthumous wishes of testators. Collateral heirs who object to will provisions that redraw the contours of “family” are likely to profit from the incompatibility of testamentary freedom and social deviance. Thus, the interests of married, white adults may claim priority over nonwhite, unmarried others. Wills that acknowledge the existence of moral or social transgressions—namely, interracial sex and reproduction—incite will contests by collateral heirs who leverage their status as white and legitimate in order to defeat testamentary intent. This Article turns to antebellum and …

Purpose
End-organ apoptosis is well-described in progressive sepsis and Multiple Organ Dysfunction Syndrome (MODS), especially where platelets accumulate (e.g. spleen and lung). We previously reported an acute sepsis-induced cytotoxic platelet phenotype expressing serine protease granzyme B. We now aim to define the site(s) of and mechanism(s) by which platelet granzyme B induces end-organ apoptosis in sepsis.
Methods
End-organ apoptosis in murine sepsis (i.e. polymicrobial peritonitis) was analyzed by immunohistochemistry. Platelet cytotoxicity was measured by flow cytometry following 90 minute ex vivo co-incubation with healthy murine splenocytes. Sepsis progression was measured via validated preclinical murine sepsis score.

Thinking Like Non-Lawyers: Why Empathy Is A Core Lawyering Skill And Why Legal Education Should Change To Reflect Its Importance, Ian Gallacher
College of Law - Faculty Scholarship
This article is an exploration of some of the issues raised by the recent Carnegie Report on legal education, and contains a recommendation that law schools change the way they teach especially first year law students in order to make them more empathetically aware of the circumstances by which the court opinions they study arose and the effects those opinions will have on others. This recommendation is made not just because it will make students better people, but also because it will make them better lawyers; the article analyses in depth the dangers inherent in an overemphasis on the “logical” …

Modeling Of Pharmacokinetics Of Cocaine In Human Reveals The Feasibility For Development Of Enzyme Therapies For Drugs Of Abuse, Fang Zheng, Chang-Guo Zhan
Pharmaceutical Sciences Faculty Publications
A promising strategy for drug abuse treatment is to accelerate the drug metabolism by administration of a drug-metabolizing enzyme. The question is how effectively an enzyme can actually prevent the drug from entering brain and producing physiological effects. In the present study, we have developed a pharmacokinetic model through a combined use of in vitro kinetic parameters and positron emission tomography data in human to examine the effects of a cocaine-metabolizing enzyme in plasma on the time course of cocaine in plasma and brain of human. Without an exogenous enzyme, cocaine half-lives in both brain and plasma are almost linearly …

Hepatic Mitochondrial Efficiency In Lines Of Mice Differing In Feed Intake, Thomas Murphy
Department of Animal Science: Dissertations, Theses, and Student Research
Selection was practiced for high (MH) and low (ML) heat loss using direct calorimetry to create lines of mice differing in feed intake per unit BW (FI/BW). Selection occurred for both criteria in each of 3 replications (R); an unselected control (MC) was also maintained in each replicate. A total of 25 generations of selection was practiced; the 9 lines are presently maintained without selection. As a percentage of MC, mice of MH and ML lines differed in heat loss and FI/BW by 56 and 34%, respectively, when selection ceased. The 218 male mice measured in the present study came …

PURPOSE: Ceria engineered nanomaterials (ENMs) have current commercial applications and both neuroprotective and toxic effects. Our hypothesis is that ceria ENMs can associate with brain capillary cells and/or cross the blood-brain barrier.
METHODS: An aqueous dispersion of ∼5 nm ceria ENM was synthesized and characterized in house. Its uptake space in the Sprague Dawley rat brain was determined using the in situ brain perfusion technique at 15 and 20 mL/minute flow rates; 30, 100, and 500 μg/mL ceria perfused for 120 seconds at 20 mL/minute; and 30 μg/mL perfused for 20, 60, and 120 seconds at 20 mL/minute. The capillary …

Using Next Generation Sequencing (Ngs) To Identify And Predict Micrornas (Mirnas) Potentially Affecting Schizophrenia And Bipolar Disorder, Vernell Williamson
Theses and Dissertations
The last decade has seen considerable research focusing on understanding the factors underlying schizophrenia and bipolar disorder. A major challenge encountered in studying these disorders, however, has been the contribution of genetic, or etiological, heterogeneity to the so-called “missing heritability” [1-6]. Further, recent successes of large-scale genome-wide association studies (GWAS) have nonetheless seen only limited advancements in the delineation of the specific roles of implicated genes in disease pathophysiology. The study of microRNAs (miRNAs), given their ability to alter the transcription of hundreds of targeted genes, has the potential to expand our understanding of how certain genes relate to schizophrenia …
Synthesis Of Half-Loaded Nitroimidazole Indocyanine Green Dyes Attached To Carbon Nanotubes, Courtney L. Stanford
Synthesis Of Half-Loaded Nitroimidazole Indocyanine Green Dyes Attached To Carbon Nanotubes, Courtney L. Stanford
Master's Theses
Indocyanine green dye is the only FDA approved NIR fluorescent dye that can be used in humans. 2-Nitroimidazoles are very effective radiosensitizers, have low cytotoxicity, and target hypoxic tissue. By coupling the two together, hypoxic tumors can be targeted and imaged. Previous work has been done to couple 2-nitroimidazole to both sides of the indocyanine green dye. This work focuses on expanding that concept to couple 2-nitroimidazole to only one side of the dye, thus allowing for the other end to be available to couple to another compound, generating a half loaded 2-nitroimidazole indocyanine green dye. In theory, this half-loaded …
Dietary Protein Influences Paracellular Calcium Transport: Two Molecular Targets, Belinda M. Kotler
Dietary Protein Influences Paracellular Calcium Transport: Two Molecular Targets, Belinda M. Kotler
Master's Theses
The effect of dietary protein on bone health remains controversial. We have shown that increasing dietary protein increases calcium absorption in both humans and rats with no change in bone resorption. We used a rat model in which dietary protein increases intestinal calcium absorption and whole body calcium retention, to explore the molecular basis for this effect. Female Sprague-Dawley rats were fed a low (5%) or high (40%) protein diet for 7 days. On day 7, duodenal mucosa was harvested and total RNA isolated. Microarray analyses using these RNA samples identified 208 genes whose expression was at least 1.5 fold …

A Longitudinal Study Of The Effect Of At Birth Adoptions On Anxiety, Stress Hormones And Adolescent Alcohol Intake: A Nonhuman Primate Model, Whitney Faith Maxwell
Theses and Dissertations
Adopted individuals have an increased risk for a variety of psychopathological disorders. Studies of the effects adoption in humans are difficult to perform because of the difficulty separating genetic risk and treatment effects. This is a developmental study investigating the effects of at birth adoption using a nonhuman primate model. Three experimental paradigms were used to assess maternal treatment, stress-related behavior, and physiology late in infancy and again later in life. Rhesus monkeys were reared for their first six months of life by either their biological mother or an unrelated, lactating adult female. Adoptions occurred immediately following birth. At six …

All Theses and Dissertations (ETDs)
Multiple Sclerosis is a common disease, affecting 2.5 million people world-wide. The clinical course is heterogeneous, ranging from benign disease in which patients live an almost normal life to severe and devastating disease that may shorten life. Despite much research, a fully effective treatment for MS is still unavailable and diagnostic techniques for monitoring MS disease evolution are much needed.
As a non-invasive tool, Magnetic resonance imaging: MRI) plays a key role in MS diagnosis. Numerous MRI techniques have been proposed over the years. Among most widely used are conventional T1-weighted: T1W), T2-weighted: T2W) and FLuid Attenuated Inversion Recovery: FLAIR) …

Titanium Nitride And Nitrogen Ion Implanted Coated Dental Materials, Youssef S. Al Jabbari, Jennifer Fehrman, Ashley C. Barnes, Angela M. Zapf, David W. Berzins, Spiros Zinelis
School of Dentistry Faculty Research and Publications
Titanium nitride and/or nitrogen ion implanted coated dental materials have been investigated since the mid-1980s and considered in various applications in dentistry such as implants, abutments, orthodontic wires, endodontic files, periodontal/oral hygiene instruments, and casting alloys for fixed restorations. Multiple methodologies have been employed to create the coatings, but detailed structural analysis of the coatings is generally lacking in the dental literature. Depending on application, the purpose of the coating is to provide increased surface hardness, abrasion/wear resistance, esthetics, and corrosion resistance, lower friction, as well as greater beneficial interaction with adjacent biological and material substrates. While many studies have …

Understanding Free Radicals: Isolating Active Thylakoid Membranes And Purifying The Cytochrome B6f Complex For Superoxide Generation Studies, Jason Stofleth
The Journal of Purdue Undergraduate Research
All life persists in an environment that is rich in molecular oxygen. The production of oxygen free radicals, or superoxide, is a necessary consequence of the biogenesis of energy in cells. Both mitochondrial and photosynthetic electron transport chains have been found to produce superoxide associated with cell differentiation, proliferation, and cell death, thereby contributing to the effects of aging. Aerobic respiration in mitochondria consumes oxygen, whereas photosynthesis in chloroplasts or cyanobacteria produces oxygen. The increased concentration of molecular oxygen may serve to allow greater availability for the production of superoxide by cytochrome bc complexes in photosynthetic membranes compared to those …
