Recent archaeological discoveries have revealed that pigment use, beads, engravings, and sophisticated stone and bone tools were already present in southern Africa 75,000 y ago. Many of these artifacts disappeared by 60,000 y ago, suggesting that modern behavior appeared in the past and was subsequently lost before becoming firmly established. Most archaeologists think that San hunter–gatherer cultural adaptation emerged 20,000 y ago. However, reanalysis of organic artifacts from Border Cave, South Africa, shows that the Early Later Stone Age inhabitants of this cave used notched bones for notational purposes, wooden digging sticks, bone awls, and bone points similar to those …

A new in vitro culture model for latency of tuberculosis pathogen suitable for screening chemical libraries was developed by subjecting the pathogen to multiple stresses that the pathogen is thought to encounter in the host. When subjected to these stress factors, the pathogen develops key features characteristic of latency, drug resistance and lipid storage. Using the culture, a method to screen chemicals was developed. This method is suitable for high through put screening.

The mechanism of microparticle shedding from the plasma membrane of calcium-loaded cells has been investigated in erythrocytes and platelets. Recent studies have revealed the physiological and clinical importance of microparticle release from nucleated cells such as lymphocytes and endothelium. The experiments of this study were designed to address whether simple mechanisms discovered in platelets and erythrocytes also apply to the more complex nucleated cells. Four such mechanisms were addressed: potassium efflux, transbilayer phosphatidylserine migration, cytoskeleton degradation, and membrane lipid order. The extraction of structural object metrics from a next generation remote sensing modality, namely waveform light detection and ranging (LiDAR), has garnered increasing interest from the remote sensing research community. However, a number of challenges need to be addressed before structural or 3D vegetation modeling can be accomplished. These include proper processing of complex, often off-nadir waveform signals, extraction of relevant waveform parameters that relate to vegetation structure, and from a quantitative modeling perspective, 3D rendering of a vegetation object from LiDAR waveforms. In this paper several astrodynamical formation flying models are assessed against the experimental results derived from the SPACE-SI formation flying experiments performed in September 2011 with the OHB Sweden developed Prisma satellites Mango and Tango. In these formation flying experiments critical manoeuvres for three types of missions were investigated with respect to in-orbit performances. The experiments included parallel flying with in track displacement demonstrating high-resolution optical dual satellite imaging and radar interferometric constellation, circumvolution as well as encircling of the target demonstrating debris observation and parallel flying with the radial displacement demonstrating fractionated spacecraft and accurate pointing of the formation. …

Host responsiveness to IFNg is critical for resolution of Listeria infection, but the identities and roles of IFNg responsive cells that initiate this process remain unclear. In this thesis, unique mice displaying conditional loss of the IFNg receptor: Ifngr1f/f mice) in different tissues have been generated and used to explore the role of this cytokine in initiating the anti-Listeria response.

Whereas Ifngr1f/f mice displayed normal cellular levels of IFNGR1 and normal resistance to Listeria infection, Vav-icre+Ifngr1f/f mice lacking IFNg responsiveness selectively in hematopoietic cells exhibited highly increased susceptibility to Listeria infection comparable to that of Ifngr1-/- mice. Murine gammaherpesvirus 68 (MHV68) ORF73 (mLANA) has sequence homology to Kaposi’s sarcoma-associated herpesvirus (KSHV) latency-associated nuclear antigen (LANA). LANA acts on the KSHV terminal repeat (TR) elements to mediate KSHV episome maintenance. Disruption of mLANA expression severely reduces the ability of MHV68 to establish latent infection in mice, consistent with the possibility that mLANA mediates episome persistence. Here we assess the roles of mLANA and MHV68 TR (mTR) elements in episome persistence. mTR-associated DNA persisted as an episome in latently MHV68-infected tumor cells, demonstrating that the mTR elements can serve as a cis-acting element for MHV68 episome maintenance. Sexual desire is ubiquitous in all theater. It follows that virtually all theatrical traditions struggle with the issue of the representation of that desire onstage. The French stage of the 1630s and Jean Rotrou's theater of that period in particular constitute an unusual moment of relative freedom before the imposition of the bienséances banished sexual activity from the stage. The involvement of the gut microbiota in metabolic disorders, and the ability of whole grains to affect both host metabolism and gut microbial ecology, suggest that some benefits of whole grains are mediated through their effects on the gut microbiome. Nutritional studies that assess the effect of whole grains on both the gut microbiome and human physiology are needed. Rat exposure to 60% O2 for 7 days (hyper-60) or to >95% O2 for 2 days followed by a 24-hour in room air (hyper-95R) confers susceptibility or tolerance, respectively, of the otherwise lethal effects of subsequent exposure to 100% O2. The objective of this study was to determine if lung retention of the radiopharmaceutical agent 99mTc-HMPAO (hexamethylpropyleneamine oxime) is differentially altered in hyper-60 and hyper-95R rats. Tissue retention of HMPAO is dependent on intracellular content of the antioxidant glutathione (GSH) and mitochondrial function. A passive treatment alternative for the removal of metal and organic contaminants from runoff and effluent in CDFs was evaluated in this study. Activated carbon impregnated curtains have been used for different remediation projects; however, very little research has been conducted to understand its sorption behavior, breakthrough time, and exhaustion time of contaminants under high flow rates characteristic of CDF runoff and effluent. Regulatory T cells (Tregs), interleukin 17-producing T helper cells (Th17 cells), and other immune cells play important roles in the maintenance of pregnancy, and their function is impacted by HIV infection. I hypothesized that FIV-infection may likewise alter placental T cell gene expression causing aberrant immune function and compromised pregnancy. The purpose of this project was to evaluate the expression placental immunomodulators related to Treg and Th17 cell function in the FIV-infected cat model. Gene expression was quantified in placenta and serum using quantitative PCR and ELISA, respectively. A need exists for a more robust method of evaluating musculoskeletal injuries resulting from impact conditions, particularly blasts. Computational modeling is a promising method of achieving this goal. The accuracy of a model depends on high quality mechanical properties for each component. This study examined the mechanical properties of porcine muscle along with structure property relationships. Fresh muscle was tested in compression and tension at strain rates of 0.1 s-1, 0.01 s-1, and 0.001 s-1. This research presents a rail transportation cost analysis of bulk agricultural commodities (such as grain and wood chips) with similar characteristics as pre-processed biomass. This study analyzes the cost factors that affect rail pricing for shipments of bulk-commodities (such as grain) from the Midwest to various regions in the US using regression analysis theories. The rail cost equations developed from the regression analysis were used to compare the trade-offs that exist between truck, rail and barge transportation of pre-processed biomass.

Diesel and biodiesel combustion in a light duty compression ignition engine were simulated during a closed cycle, using a commercial computational fluid dynamics (CFD) code, CONVERGE. The corresponding computational domain was constructed for the engine based on combustion chamber geometry and compression ratio measurements. Submodels were calibrated for simulation. The results were able to capture the experimental pressure and apparent heat release rate trends for both fuels over a range of engine loads and fuel injection timings. NOx emissions trends were captured for diesel, while under-predicted for biodiesel. The NOx trends were also analyzed based on the thermal NO mechanism. …

Cyanobacterial algal bloom is a major water quality issue in inland lakes, reservoirs, and estuarine environments because of its scum and bad odor forming and toxin producing abilities. Health risks from cyanobacterial toxin can vary from skin irritations to fever, intestinal problems, and neurological disorders. Terminations of blooms also cause oxygen depletion leading to hypoxia and widespread fish kills. Smoking, physical activity, and insufficient fruit and vegetable consumption have been linked to increased instances of coronary heart disease, stroke, peripheral vascular disease, chronic obstructive lung disease, type 2 diabetes, osteoporosis, depression, hypertension, numerous cancers, and complications surrounding blood pressure and blood cholesterol. In the United States approximately 22% of college students have smoked at least one cigarette in the past 30 days; 70-85% are not meeting recommended levels of physical activity; and 75% are not consuming recommended levels of fruits and vegetables. Aeromonas hydrophila is a Gram-negative, rod shaped, facultative anaerobic bacterium that is ubiquitous to freshwater and slightly brackish aquatic environments and can cause infections in fish, humans, reptiles, and avian species. Recent severe outbreaks of disease in catfish aquaculture have been associated with a highly virulent Aeromonas hydrophila strain (VAH) that is genetically distinct from less virulent strains. Given that A. hydrophila is known to infect birds, we hypothesized that fish eating birds may serve as a reservoir for VAH and spread the pathogen by flying to uninfected ponds. Dens are a critical component of black bear (Ursus americanus) habitat, yet scale-dependent den-site selection has received limited attention. Natural and anthropogenic factors (e.g., vegetation, roads) may also influence bear den-site selection. I quantified black bear denning chronology and den use and evaluated multi-scale den-site selection in Mississippi, USA during 2005–2011. Females entered dens earlier than males and emerged later; multiple den use by both sexes in a single winter was common. Methylmercury (MeHg) production is magnified in the natural environment by geochemical influxes and an active sulfate reducing bacteria community. It has been found that the presence of sulfides, excess nutrients, and the type of depositional environment (normal marine or euxinic) greatly influences MeHg production and degree of pyritization (DOP). The aim of this research is to investigate a possible connection between MeHg accumulation and the DOP in Weeks Bay sediment.
