Soft Microenvironments Promote The Early Neurogenic Differentiation But Not Self-Renewal Of Human Pluripotent Stem Cells, Albert Keung, Prashanth Asuri, Sanjay Kumar, David Schaffer

Human pluripotent stem cells (hPSCs) are of great interest in biology and medicine due to their ability to self-renew and differentiate into any adult or fetal cell type. Important efforts have identified biochemical factors, signaling pathways, and transcriptional networks that regulate hPSC biology. However, recent work investigating the effect of biophysical cues on mammalian cells and adult stem cells suggests that the mechanical properties of the microenvironment, such as stiffness, may also regulate hPSC behavior. While several studies have explored this mechanoregulation in mouse embryonic stem cells (mESCs), it has been challenging to extrapolate these findings and thereby explore their …

Multiplex real-time PCR detection of Escherichia coli O157:H7 is an efficient molecular tool with high sensitivity and specificity for meat safety assurance. The Biocontrol GDS and DuPont Qualicon BAX-RT rapid detection systems are two commercial tests based on real-time PCR amplification with potential applications for quantification of specific E. coli O157:H7 gene targets in enriched meat samples. However, there are arguments surrounding the use of these tests to predict pre-enrichment concentrations of E. coli O157:H7, as well as arguments pertaining to the influence of non-viable cells causing false positive results. Application Of Seawinds Scatterometer Data To The Study Of Antarctic Icebergs, Keith Mitchell Stuart

Knowledge of iceberg location and size is important for safety reasons as well as for understanding many geophysical and biological processes. This dissertation analyzes large tabular icebergs in the Southern Ocean using the SeaWinds scatterometer. SeaWinds is a spaceborne radar designed to measure the microwave backscatter from the Earth's surface. Using resolution-enhancement techniques, backscatter measurements are processed into backscatter images in which icebergs can be observed. An iceberg detection methodology is formalized using daily scatterometer images. The percentage of higher education students enrolled in online courses has increased from 9.6 percent in fall 2002 to 33 percent in fall 2010. Due to the increased importance of online courses and programs on university campuses there is a need to better prepare novice technology faculty for the delivery of these courses. This article provides a description of the process through which a group of faculty with low to high technology skills prepared to deliver an online masters program. Choosing The Best Plant For The Job: A Cost-Effective Assay To Prescreen Ancient Plant Remains Destined For Shotgun Sequencing, Nathan Wales, J. Alberto Romero-Navarro, Enrico Cappellini, M. Thomas P. Gilbert

DNA extracted from ancient plant remains almost always contains a mixture of endogenous (that is, derived from the plant) and exogenous (derived from other sources) DNA. The exogenous ‘contaminant’ DNA, chiefly derived from microorganisms, presents significant problems for shotgun sequencing. In some samples, more than 90% of the recovered sequences are exogenous, providing limited data relevant to the sample. However, other samples have far less contamination and subsequently yield much more useful data via shotgun sequencing. Given the investment required for high-throughput sequencing, whenever multiple samples are available, it is most economical to sequence the least contaminated sample. We present …
