Daugherty Water for Food Global Institute: Faculty Publications
This study aims to assess the link between fluoride content in groundwater and its impact on dental health in rural communities of the Ethiopian Rift. A total of 148 water samples were collected from two drainage basins within the Main Ethiopian Rift (MER). In the Ziway-Shala basin in particular, wells had high fluoride levels (mean: 9.4 ± 10.5 mg/L; range: 1.1 to 68 mg/L), with 48 of 50 exceeding the WHO drinking water guideline limit of 1.5 mg/L. Total average daily intake of fluoride from drinking groundwater (calculated per weight unit) was also found to be six times higher than …

Abstract
Background
Magnetic resonance imaging is a noninvasive method of evaluating embryonic development. Diffusion tensor imaging (DTI), based on the directional diffusivity of water molecules, is an established method of evaluating tissue structure. Yet embryonic motion degrades the in vivo acquisition of long-duration DTI. We used a dual-cooling technique to avoid motion artifact and aimed to investigate whether DTI can be used to monitor chick embryonic skeletal muscle development in ovo, and to investigate the correlation between quantitative DTI parameters fractional anisotropy (FA) and fiber length and quantitative histologic parameters fiber area percentage (FiberArea%) and limb length.

Abstract
Background
Protein phosphatase 1 (PP1) is one of the major phosphatases responsible for protein dephosphorylation in eukaryotes. Protein phosphatase 1 regulatory subunit 12B (PPP1R12B), one of the regulatory subunits of PP1, can bind to PP1cδ, one of the catalytic subunits of PP1, and modulate the specificity and activity of PP1cδ against its substrates. Phosphorylation of PPP1R12B on threonine 646 by Rho kinase inhibits the activity of the PP1c-PPP1R12B complex. However, it is not currently known whether PPP1R12B phosphorylation at threonine 646 and other sites is regulated by insulin. We set out to identify phosphorylation sites in PPP1R12B and to …

Abstract
MicroRNAs (miRNAs) are short non-coding RNAs that are involved in several important biological processes through regulation of genes post-transcriptionally. Carcinogenesis is one of the key biological processes where miRNAs play important role in the regulation of genes. The miRNAs elicit their effects by binding to the 3' untranslated region (3'UTR) of their target mRNAs, leading to the inhibition of translation or the degradation of the mRNA, depending on the degree of complementary base pairing. To-date more than 1,000 miRNAs are postulated to exist, although the field is moving rapidly. Currently, miRNAs are becoming the center of interest in a …

Abstract
Background
Spy1 is a novel 'cyclin-like' activator of the G1/S transition capable of enhancing cell proliferation as well as inhibiting apoptosis. Spy1 protein levels are tightly regulated during normal mammary development and forced overexpression in mammary mouse models accelerates mammary tumorigenesis.
Methods
Using human tissue samples, cell culture models and in vivo analysis we study the implications of Spy1 as a mediator of mammary transformation and breast cancer proliferation.
Results
We demonstrate that this protein can facilitate transformation in a manner dependent upon the activation of the G2/M Cdk, Cdk1, and the subsequent inhibition of the anti-apoptotic regulator FOXO1. …

ALTERATIONS IN REGIONAL CEREBRAL BLOOD FLOW WITH PROPOFOL ANESTHESIA COMPARED WITH SEVOFLURANE. Tejas Manchandia, Maolin Qiu, Margaret Rose, Todd Constable, and Ramachandran Ramani. Department of Anesthesiology and Department of Diagnostic Radiology, Yale University School of Medicine, New Haven, CT.
Alterations in regional cerebral blood flow (rCBF) are a reflection of neuronal activity in the brain because of the property of flow-metabolism coupling. Our group has previously reported a non-uniformity in rCBF changes with 0.25 MAC sevoflurane. The purpose of this project was to measure the alteration in rCBF with 0.5 MAC equivalent propofol and compare the changes in rCBF with …
Long-Term Patterns Of An Estuarine Pathogen Along A Salinity Gradient, David Bushek, Susan E. Ford, Iris Burt
Long-Term Patterns Of An Estuarine Pathogen Along A Salinity Gradient, David Bushek, Susan E. Ford, Iris Burt
Journal of Marine Research
Parasitic, disease-causing pathogens can exert strong control over marine populations yet few long-term studies exist that describe these relationships. Understanding the connections to long-term large-scale processes relative to local short-term processes should facilitate better planning for disease impacts in the management of marine resources. We describe a 21-yr dataset of dermo disease (Perkinsus marinus) in eastern oysters (Crassostrea virginica) in Delaware Bay, USA. Analyses indicated (1) a strong positive association between disease and mortality that was non-linear and defined by thresholds, (2) a clear spatial gradient of increasing disease and mortality with increasing salinity, (3) an …

Can Oysters Crassostrea Virginica Develop Resistance To Dermo Disease In The Field: The Impediment Posed By Climate Cycles, Eric N. Powell, John M. Klinck, Ximing Guo, Eileen E. Hofmann, Susan E. Ford, David Bushek
Journal of Marine Research
Populations of eastern oysters, Crassostrea virginica, are commonly limited by mortality from dermo disease. Little development of resistance to Perkinsus marinus, the dermo pathogen, has occurred, despite the high mortality rates and frequency of epizootics. Can the tendency of the parasite to exhibit cyclic epizootics limit the oyster's response to the disease despite the presence of alleles apparently conferring disease resistance? We utilize a gene-based population dynamics model to simulate the development of disease resistance in Crassostrea virginica populations exposed to cyclic mortality encompassing periodicities expected of dermo disease over the geographic range at which epizootics have been …
Spatial And Temporal Variability Of Disease Refuges In An Estuary: Implications For The Development Of Resistance, Susan E. Ford, Emily Scarpa, David Bushek
Spatial And Temporal Variability Of Disease Refuges In An Estuary: Implications For The Development Of Resistance, Susan E. Ford, Emily Scarpa, David Bushek
Journal of Marine Research
Although the concept of genetic refuge has long been employed in ecological and paleoecological context, it has only rarely been used to identify regions where organisms are protected from diseases that affect the rest of a population. The refuges harbor individuals that have not been exposed to selective mortality and remain susceptible to the disease. They represent a reservoir of susceptibility alleles that can mix with those from resistant survivors of disease and can retard the development of resistance in the population as a whole. Two water-borne protistan parasites affect oysters along the east coast of the United States: Haplosporidium …

Experimental Assessment Of Particle Mixing Fingerprints In The Deposit-Feeding Bivalve Abra Alba (Wood), Guillaume Bernard, Antoine Grémare, Olivier Maire, Pascal Lecroart, Filip J. R. Meysman, Aurélie Ciutat, Bruno Deflandre, Jean Claude Duchêne
Journal of Marine Research
Particle mixing induced by the deposit-feeding bivalve Abra alba was assessed using a new experimental approach allowing for the tracking of individual particle displacements. This approach combines the adaptation of existing image acquisition techniques with new image analysis software that tracks the position of individual particles. This led to measurements of particle mixing fingerprints, namely the frequency distributions of particle waiting times, and of the characteristics (i.e. direction and length) of their jumps. The validity of this new approach was assessed by comparing the so-measured frequency distributions of jump characteristics with the current qualitative knowledge regarding particle mixing in the …

Comparison Of Three Cidr Based Fixed-Time Ai Protocols For Beef Heifers, G.A. Perry, J.K. Grant, J.A. Walker, G.A. Bridges, S.G. Kruse, S. Bird, K. Heaton, R. Arias, S.L. Lake
South Dakota Beef Report, 2012
Several effective fixed-time AI (FTAI) protocols have been developed to facilitate AI while eliminating the need for estrus detection. Among these are the 5-d CO-Synch+CIDR (5d), PG 6-d CIDR (PG-CIDR), and 14-d CIDR-PG (CIDR-PG) protocols. While each of these protocols varies in duration and approach to synchronizing estrus and ovulation, each has been reported as an effective method to facilitate FTAI in beef heifers. Therefore, the objective of this study was to compare FTAI pregnancy rates in beef heifers synchronized with these three CIDR based protocols. Virgin beef heifers (n = 801) at four locations were synchronized with one of …
