William Drummond And The Battle Of Fort Erie, Donald E. Graves Jan 2012

William Drummond And The Battle Of Fort Erie, Donald E. Graves

Canadian Military History

The officers and men of the British army that defended Canada from American invasion during the War of 1812 knew they were “forgotten soldiers.” Fighting in a distant and secondary theatre, far from the gaze of a government and public pre-occupied with events on the continent, especially in Spain, they took a somewhat perverse pride in their status as outcasts. As one quipped about the Duke of Wellington—“thank God he managed to do without us” at Waterloo. But they also took a particular pride in their own local heroes including such men as Gordon of the 1st Regiment of Foot, …

Spartin Protein Associates With Phospholipids Via Its Plant-Related Senescence Domain And Functions As A Lipid Transfer Protein, Maureen Ashlee Shaw Jan 2012

Spartin Protein Associates With Phospholipids Via Its Plant-Related Senescence Domain And Functions As A Lipid Transfer Protein, Maureen Ashlee Shaw

Theses (1 year embargo)

Troyer syndrome is a hereditary spastic paraplegia caused by a mutation that leads to a complete loss of expression of spartin protein. The plant-related senescence domain in spartin is highly conserved and present in over 150 proteins, but its function is unknown. Our results indicate that spartin associates with phospholipids via its senescence domain.

Phospholipids are important components of intracellular membranes and play roles in many cellular processes. Knock-down of spartin results in impaired cell division. The phospholipid phosphatidylinositol 3-phosphate (PI3P) is present at midbodies, where it recruits proteins for cytokinesis. Our findings demonstrate that spartin colocalizes with PI3P at …

On The Move: Games And Gaming Figures In Nineteenth-Century U.S. Literature, Douglas Anthony Guerra Jan 2012

On The Move: Games And Gaming Figures In Nineteenth-Century U.S. Literature, Douglas Anthony Guerra

Dissertations (1 year embargo)

In this dissertation, I analyze the way that gameplay, considered broadly, both facilitated and informed representations of agency in the nineteenth century. In effect, games were archives of possibility made procedurally discrete, rendering modes of action and agency legible in ways that are suggested, although differently engaged, in their somewhat less ephemeral cousins: books. Understanding that both games and books emerged from within similar fields of cultural production and often marketed themselves to the same audiences, I argue that taking a technical and materially historical approach to games helps us to explore major literary works of the period to startlingly …

The Synaptic Vesicle Snare Neuronal Synaptobrevin Promotes Endolysosomal Degradations And Prevents Neurodegeneration, Adam Haberman, W. Ryan Williamson, Daniel Epstein, Dong Wang, Srisha Rina, Ian A. Meinertzhagen, P. Robin Hiesinger

Biology: Faculty Scholarship

Soluble NSF attachment protein receptors (SNAREs) are the core proteins in membrane fusion. The neuron-specific synaptic v-SNARE n-syb (neuronal Synaptobrevin) plays a key role during synaptic vesicle exocytosis. In this paper, we report that loss of n-syb caused slow neurodegeneration independent of its role in neurotransmitter release in adult Drosophila melanogaster photoreceptor neurons. In addition to synaptic vesicles, n-Syb localized to endosomal vesicles. Loss of n-syb lead to endosomal accumulations, transmembrane protein degradation defects, and a secondary increase in autophagy. Our evidence suggests a primary defect of impaired delivery of vesicles that contain degradation proteins, including the acidification-activated …

Expression Of 14-3-3 Protein Isoforms In Mouse Oocytes, Eggs And Ovarian Follicular Development, Santanu De, Jennifer Marcinkiewicz, Srinivasan Vijayaraghavan, Douglas Kline

Biology Faculty Articles

Background

The 14-3-3 (YWHA) proteins are a highly conserved, ubiquitously expressed family of proteins. Seven mammalian isoforms of 14-3-3 are known (β, γ, ε, ζ, η, τ and, σ). These proteins associate with many intracellular proteins involved in a variety of cellular processes including regulation of the cell cycle, metabolism and protein trafficking. We are particularly interested in the role of 14-3-3 in meiosis in mammalian eggs and the role 14-3-3 proteins may play in ovarian function. Therefore, we examined the expression of 14-3-3 proteins in mouse oocyte and egg extracts by Western blotting after polyacrylamide gel electrophoresis, viewed fixed …

Optimal Population Of Foxp3+ T Cells In Tumors Requires An Antigen Priming-Dependent Trafficking Receptor Switch., Chuanwu Wang, Jee H. Lee, Chang H. Kim

Department of Comparative Pathobiology Faculty Publications

FoxP3+ T cells populate tumors and regulate anti-tumor immunity. The requirement for optimal population of FoxP3+ regulatory T cells in tumors remains unclear. We investigated the migration requirement and stability of tumor-associated FoxP3+ T cells. We found that only memory, but not naïve, FoxP3+ T cells are highly enriched in tumors. Almost all of the tumor-infiltrating FoxP3+ T cells express Helios, an antigen associated either with thymus-generated FoxP3+ T cells or activated T cells in the periphery. The tumor-infiltrating FoxP3+ T cells largely lack CD62L and CCR7, two trafficking receptors required for T cell migration into secondary lymphoid tissues. Instead, …

Epigenetic Regulation Of Genes Involved In Vascular Dysfunction In Preeclamptic Women, Ahmad Mousa Jan 2012

Epigenetic Regulation Of Genes Involved In Vascular Dysfunction In Preeclamptic Women, Ahmad Mousa

Theses and Dissertations

DNA methylation is the most recognizable epigenetic mechanism. In general, DNA hypomethylation is associated with increased gene expression whereas DNA hypermethylation is associated with decreased gene expression. To date, little is known about the role of DNA methylation in the pathophysiology of preeclampsia. In this study, we examined the differences in DNA methylation in omental arteries of normal pregnant and preeclamptic women using the high throughput Illumina HumanMethylation27 BeadChip assay. We found 1,685 genes with a significant difference in DNA methylation at a false discovery rate of < 10% with many inflammatory genes having reduced methylation. The thromboxane synthase gene was the most hypomethylated gene in preeclamptic women as compared to normal pregnant women. When we examined the expression of thromboxane synthase in omental arteries of normal pregnant and preeclamptic women we found it to be significantly increased in preeclamptic women. The increased expression was observed in vascular smooth muscle cells, endothelial cells and infiltrating neutrophils. Experimentally induced DNA hypomethylation increased the expression of thromboxane synthase in the neutrophil-like HL-60 cell line, whereas tumor necrosis factor α (TNFα), a neutrophil product, increased its expression in cultured human vascular smooth muscle cells (VSMC). These finding suggest that DNA methylation and release of TNFα by infiltrating neutrophils could contribute to the increased expression of thromboxane synthase in systemic blood vessels of preeclamptic women, contributing to the hypertension and coagulation abnormalities. We also explored the possible contribution of DNA methylation to the altered expression of genes involved in collagen metabolism in preeclampsia. Several matrix metalloproteinase (MMP) genes, including MMP1 and MMP8, were significantly less methylated in preeclamptic women, whereas TIMP and COL genes were either significantly more methylated or had no significant change in their DNA methylation status. Experimentally induced DNA hypomethylation increased the expression of MMP-1, but not TIMP-1 or COL1A1, in cultured VSMCs and increased the expression of MMP-1 and MMP-8 in HL-60 cells. These findings suggest that DNA methylation contributes to the imbalance in genes involved in collagen metabolism in blood vessels of preeclamptic women.

Genome-Wide Analysis Of Syntenic Gene Deletion In The Grasses, James C. Schnable, Michael Freeling, Eric Lyons Jan 2012

Genome-Wide Analysis Of Syntenic Gene Deletion In The Grasses, James C. Schnable, Michael Freeling, Eric Lyons

Department of Agronomy and Horticulture: Faculty Publications

The grasses, Poaceae, are one of the largest and most successful angiosperm families. Like many radiations of flowering plants, the divergence of the major grass lineages was preceded by a whole-genome duplication (WGD), although these events are not rare for flowering plants. By combining identification of syntenic gene blocks with measures of gene pair divergence and different frequencies of ancient gene loss, we have separated the two subgenomes present in modern grasses. Reciprocal loss of duplicated genes or genomic regions has been hypothesized to reproductively isolate populations and, thus, speciation. However, in contrast to previous studies in yeast and teleost …
