As in other eukaryotes, protein kinases play major regulatory roles in filamentous fungi. Although the genomes of many plant pathogenic fungi have been sequenced, systematic characterization of their kinomes has not been reported. The wheat scab fungus Fusarium graminearum has 116 protein kinases (PK) genes. Although twenty of them appeared to be essential, we generated deletion mutants for the other 96 PK genes, including 12 orthologs of essential genes in yeast. All of the PK mutants were assayed for changes in 17 phenotypes, including growth, conidiation, pathogenesis, stress responses, and sexual reproduction. Overall, deletion of 64 PK genes resulted in …

The Atlantic killifish (Fundulus heteroclitus) is an environmental sentinel organism used extensively for studies on environmental toxicants and salt (NaCl) homeostasis. Previous research in our laboratory has shown that rapid acclimation of killifish to seawater is mediated by trafficking of CFTR chloride channels from intracellular vesicles to the plasma membrane in the opercular membrane within the first hour in seawater, which enhances chloride secretion into seawater, thereby contributing to salt homeostasis. Acute transition to seawater is also marked by an increase in both mRNA and protein levels of serum glucocorticoid kinase 1 (SGK1) within 15 minutes of transfer. …

Drinking shallow groundwater with naturally elevated concentrations of arsenic is causing widespread disease in many parts of South and Southeast Asia. In the Bengal Basin, growing reliance on deep (>150 m) groundwater has lowered exposure. In the most affected districts of Bangladesh, shallow groundwater concentrations average 100 to 370 μg L−1, while deep groundwater is typically < 10 μg L−1. Groundwater flow simulations have suggested that, even when deep pumping is restricted to domestic use, deep groundwater in some areas of the Bengal Basin is at risk of contamination. However, these simulations have neglected the impedance of As migration by adsorption to aquifer sediments. Here we quantify for the first time As sorption on deeper sediments in situ by replicating the intrusion of shallow groundwater through injection of 1,000 L of deep groundwater modified with 200 μg L−1 of As into a deeper aquifer. Arsenic concentrations in the injected water were reduced by 70% due to adsorption within a single day. Basin-scale modelling indicates that while As adsorption extends the sustainable use of deep groundwater, some areas remain vulnerable; these areas can be prioritized for management and monitoring.

In the tropical Indo-Pacific, most phylogeographic studies have focused on the shallow-water taxa that inhabit reefs to approximately 30 m depth. Little is known about the large predatory fishes, primarily snappers (subfamily Etelinae) and groupers (subfamily Epinephelinae) that occur at 100–400 m. These long-lived, slow-growing species support fisheries across the Indo-Pacific, yet no comprehensive genetic surveys within this group have been conducted. Here we contribute the first range-wide survey of a deepwater Indo-Pacific snapper, Pristipomoides filamentosus, with special focus on Hawai'i. Although viruses have been implicated in central nervous system (CNS) diseases of unknown etiology, including multiple sclerosis and amyotrophic lateral sclerosis, the reproducible identification of viral triggers in such diseases has been largely unsuccessful. Here, we explore the hypothesis that viruses need not replicate in the tissue in which they cause disease; specifically, that a peripheral infection might trigger CNS pathology. To test this idea, we utilized a transgenic mouse model in which we found that immune cells responding to a peripheral infection are recruited to the CNS, where they trigger neurological damage. This study investigated the potential impact of soil moisture perturbations on the statistical spread of an ensemble forecast for three different synoptic events during the summer of 2006. Soil moisture was perturbed from a control simulation to generate a 12 member ensemble with six drier and six moister soils. The impacts on the near-surface atmospheric conditions and on precipitation were analysed. It was found, as previous studies have confirmed, that soil moisture can change the spatial and temporal distribution of precipitation and of the overlying circulation. In the current environment of military operations requesting faster delivery schedules to counter insurgent tactics, the engineering team often searches for how to quickly deliver the 80% solution, typically in 6-12 months. These are labeled rapid development projects. A content analysis of best practices in commercial product development literature, where time to market is often a driving factor, was accomplished showing varying emphasis of systems engineering technical and technical management processes. This article examines the theoretical and practical implications of three methods of materials acquisitions in an academic library. First, it evaluates how traditional collection development, electronic patron driven acquisitions (PDA) and other older forms of PDA affect materials storage, preservation, purchase speed and usage. Then this paper employs Foucault’s theories about power to discuss the ramifications of these acquisitions methods for librarians and three major user groups: faculty, graduate students and undergraduates. The article concludes that each method presents different practical and theoretical advantages and disadvantages.

Indexing images has always been problematic due to their richness of content and innate subjectivity. Three traditional approaches to indexing images are described and analyzed. An introduction of the contemporary use of social tagging is presented along with its limitations. Traditional practices can continue to be used as a stand-alone solution, however deficiencies limit retrieval. A collaborative technique is supported by current research and a model created by the authors for its inception is explored. The information environment is rapidly changing, affecting the ways in which information is organized and accessed. User needs and expectations have also changed due to the overwhelming influence of Web 2.0 tools. Conventional information systems no longer support evolving user needs. Based on current research, we explore a method that integrates the structure of controlled languages with the flexibility and adaptability of social tagging. This article discusses the current research and usage of social tagging and Web 2.0 applications within the academic library. Types of tags, the semiotics of tagging and its influence on indexing are covered.
