A Comparative Study On Alkaline Lipase Production By A Newly Isolated Aspergillus Fumigatus Mtcc 9657 In Submerged And Solid-State Fermentation Using Economically And Industrially Feasible Substrate, Akhila Rajan, A. Jayakumaran Nair

Turkish Journal of Biology

A comparative study of solid-state fermentation (SSF) and submerged fermentation (SmF) for the production of alkaline lipase from a new isolate, Aspergillus fumigatus MTCC 9657, using defatted rice bran (DFRB) as a substrate was studied. Different process parameters, such as incubation period, initial pH and incubation temperature, were optimised to achieve the maximum yield of the alkaline lipase. Maximum enzyme production (8.13 U/mL) was obtained by day 7 of incubation in SSF compared to day 4 in SmF at an optimum pH of 8.5 and ambient temperature of 30 °C. Lipase produced by SSF was stable over a period of …

Characterization Of Poly Phenol Oxidase In Two In Vitro Regenerated Cultivars Of Mucuna: Mucuna Pruriens L. And Mucuna Prurita H., Raghavendra Sathyanarayana, Vadlapudi Kumar, Chapeyil Kumaran Ramesh, Mahadevappa Parmesha, Mahaboob Habeebulla Moinuddin Khan

Turkish Journal of Biology

Polyphenol oxidases (PPOs EC 1.14.18.1) were isolated from Mucuna pruriens and Mucuna prurita and confirmed as tyrosinases involved in L-DOPA production. PPOs were extracted by using a 0.05 M phosphate buffer, pH 7.0. The purified enzyme was resolved into a single band by PAGE, the enzyme was confirmed as PPO by activity staining, and SDS-PAGE analysis revealed that the purified PPO of both the species is a tetramer. Substrate specificity experiments were carried out with catechol, L-DOPA, L-tyrosine, and p-cresol. Of these, catechol was evaluated as the most suitable substrate based on the determined K_{m} and V_{max} values. The optimum …

Turkish Journal of Biology

The chemical composition and nutritional value of a wild edible ectomycorrhizal mushroom from southwestern Anatolia, Tricholoma anatolicum, were analyzed. Moisture, crude oil, protein, ash, total carbohydrate content, and mineral composition of the mushrooms studied, including Fe, Na, K, Zn, Cu, Ca, Cd, and Pb, were determined. The energy values of the samples were also calculated. The analyses were conducted during the 3 different growing stages of the mushrooms: mycelium, young fruiting bodies, and mature fruiting bodies. The highest values for moisture and crude oil contents were found to be in the mycelium, ash, and carbohydrate content of young fruiting bodies. …

The Optimization Of Adna Extraction Protocol And Sex Determination Of Bronze Age Individuals From Oylum Höyük (Kilis, Turkey), Serdal Arslan, Ayşen Açikkol, Ertan Mahi̇r Korkmaz

Turkish Journal of Biology

Oylum Höyük falls within the borders of the province of Kilis, located in the southern part of Turkey. It is an important Bronze Age settlement dating back to 5500 BC. In this study, the sex of 36 individuals found in Oylum Höyük was examined using morphological and genetic methods. In the morphological method, sex was identified in 14 female and 13 male individuals. The remaining 8 individuals could not be identified due to the poor preservation of their skeletal remains. Genetic sex was determined with the amplification of sex-specific X, Y centromeric alphoid repeats and ZFX-ZYF gene regions of DNA, …

Detection Of Prokaryotic Microbial Communities Of Çamaltı Saltern, Turkey, By Fluorescein In Situ Hybridization And Real-Time Pcr, Mehmet Burçi̇n Mutlu, Kiymet Güven

Turkish Journal of Biology

Knowledge of the functions, interactions, and diversity of extremely halophilic microorganisms mostly comes from the results of studies performed in different salterns throughout the world. As model habitats, salterns allow the comparison of different techniques used for qualitative and quantitative analysis of halophilic communities in these hypersaline environments. Çamaltı Saltern is the biggest coastal solar saltern located on the Aegean coast of Turkey, and it produces most of the salt consumed in the country. In the present study, detection of prokaryotic communities of the Çamaltı Saltern was performed using 2 culture-independent methods. Real-time polymerase chain reaction (RT-PCR) and fluorescein in …

Production Of A Novel Bifunctional Catalase-Phenol Oxidase Of Scytalidium Thermophilum In The Presence Of Phenolic Compounds, Yonca Yüzügüllü, Zümrüt Begüm Ögel, Ufuk Bölükbaşi, Nursen Çoruh, Gürkan Karakaş

Turkish Journal of Biology

A novel bifunctional catalase with additional phenol oxidase activity (CATPO) is produced by the thermophilic fungus, Scytalidium thermophilum, in a growth-associated manner. In order to study the biological significance of this dual enzyme in relation to phenolic compounds, 14 phenolics were tested for their effect on growth and CATPO production. It was determined that some phenolics exerted a negative effect on growth (catechol, coumaric acid, hydroquinone, kaempferol, myricetin), while others either did not influence growth in a negative manner or enhanced growth by a maximum of 50% increase in biomass (caffeic acid, chlorogenic acid, catechin, gallic acid, resorcinol, vanillic acid). …

Neurorehabilitation Of The Hand Using The Cybergrasp[Tm] And Mirror Image, Amy Frances Boos Jan 2011

Neurorehabilitation Of The Hand Using The Cybergrasp[Tm] And Mirror Image, Amy Frances Boos

Theses

In recent years, researchers have explored the use of a mirror image as a means of rehabilitation for individuals suffering from hemiparesis. Through neuroimaging and functional testing, neurological improvement has been demonstrated in those that engage in mirror therapy. Bilateral training, or simultaneous movement of both sides of the body, has also been studied as a treatment method to improve function after cerebral vascular accident. The development of robotic systems to assist movement of the human body has played a major role in the fabrication of bilateral training devices.

In this experiment, the CyberGrasp™ robotic exoskeleton was used to assist …
