Journal of Electrochemistry

Nucleic acid as the carrier of genetic information and the functional molecules for molecular biology and bioanalytical chemistry has attracted increasing interest in electrochemical analysis. This review presents a brief outline of some electrochemical analytical assays based on molecular recognition of nucleic acids. Most of these methods are focused on the detection of nucleic acid sequence, genetic mutation and nucleic acids as functional molecules.

Fabrication Of Ito Micro-Pore Electrochemical Sensor And The Detection Of Exocytosis, Hui Zhao, Ling Li, Pin-Gang He, Yu-Zhi Fang

Exocytosis is one of the major means for cells to transmit messages. Amperometric detection of exocytosis is an important method in studying communication between cells. In this study a new type of ITO micro-pore electrochemical sensor was fabricated by means of ITO and UV light lithographic mask and was used in the detection of exocytosis with promising results. Compared with the conventional method, the ITO micro-pore sensor can not only replace the carbon fiber electrode, but be combined with fluorescent or chemiluminescent methods in detecting the physiological phenomena of cells.

Surface Diffusion Of Adsorptive Species On Gold Nanoelectrodes, Bao-Fa Su, Wei Wang, Dong-Ping Zhan, Bin Ren, Zhong-Qun Tian

Journal of Electrochemistry

Gold nanoelectrodes were prepared successfully by a programmed laser puller. The Faraday adsorptions of oxygen and iodine, and the underpotential deposition of lead on the gold nanoelectrodes were investigated. The results showed that the active areas of nanoelectrodes were dramatically higher than their appearant geometry areas, which is caused by the surface diffusion of adsorptive species from the nanoscale gold/electrolyte interface to the adjacent gold surface.

Ripe For The Plucking: Centralized And Consolidated Library Budgets As Revenue Streams For Profit., Roger Cross

The accumulation and centralization of fiscal and selection power into the hands of library professionals could have benefited all, but inherent flaws were bound to arise. In particular the failure to adapt to increased large profit-driven publishers business. Libraries and universities failed to adequately foresee the extent of publishing monopolization. The academic library, as provider of campus-wide journal subscription, reduced demand by reducing the need to maintain individual departmental and faculty subscriptions. The end result was that a small number of publishers were able to fix pricing according to desired profit margins.

The serpin plasminogen activator inhibitor-1 (PAI-1) is a crucial regulator in fibrinolysis and tissue remodeling. PAI-1 has been associated with several pathological conditions and is a validated prognostic marker in human cancers. However, structural information about the native inhibitory form of PAI-1 has been elusive because of its inherent conformational instability and rapid conversion to a latent, inactive structure. Here we report the crystal structure of PAI-1 W175F at 2.3 Å resolution as the first model of the metastable native molecule. Structural comparison with a quadruple mutant (14-1B) previously used as representative of the active state uncovered key differences. The …

The Runt-related transcription factor, Runx2, is essential for osteogenesis and is controlled by both distal (P1) and proximal (P2) promoters. To understand Runx2 function requires determination of the spatiotemporal activity of P1 and P2 to Runx2 protein production. We generated a mouse model in which the P1-derived transcript was replaced with a lacZ reporter allele, resulting in loss of P1-derived protein while simultaneously allowing discrimination between the activities of the two promoters. Loss of P1-driven expression causes developmental defects with cleidocranial dysplasia-like syndromes that persist in the postnatal skeleton. Lafora Disease (LD) is a fatal neurodegenerative epileptic disorder that presents as a neurological deterioration with the accumulation of insoluble, intracellular, hyperphosphorylated carbohydrates called Lafora bodies (LBs). LD is caused by mutations in either the gene encoding laforin or malin. Laforin contains a dual specificity phosphatase domain and a carbohydrate-binding module, and is a member of the recently described family of glucan phosphatases. In the current study, we investigated the functional and physiological relevance of laforin dimerization. African Americans (AA) tend to have heightened systemic inflammation and endothelial dysfunction. Endothelial microparticles (EMP) are released from activated/apoptotic endothelial cells (EC) when stimulated by inflammation. The purpose of our study was to assess EMP responses to inflammatory cytokine (TNF-α) and antioxidant (superoxide dismutase, SOD) conditions in human umbilical vein ECs (HUVECs) obtained from AA and Caucasians. EMPs were measured under four conditions: control (basal), TNF-α, SOD, and TNF-α + SOD. Culture supernatant was collected for EMP analysis by flow cytometry and IL-6 assay by ELISA. Induction of endotoxin tolerance leads to a reduced inflammatory response after repeated challenge by LPS and is important for resolution of inflammation and prevention of tissue damage. Enterobacterial LPS is recognized by the TLR4 signaling complex, whereas LPS of some non-enterobacterial organisms is capable of signaling independently of TLR4 utilizing TLR2-mediated signal transduction instead. In this study we report that Porphyromonas gingivalis LPS, a TLR2 agonist, fails to induce a fully endotoxin tolerant state in a human monocytic cell line (THP-1) and mouse bone marrow-derived macrophages. The largest instrumentation grant in Lawrence University’s history — $552,666 from the National Science Foundation — will fund the purchase of a confocal microscope system to support biological research and strengthen hands-on research training.

Confocal microscopy is a cutting edge technique that provides the best available resolution of microscopic images and allows the reconstruction of three-dimensional structures from images obtained through the microscope. Seven teams of faculty mentors and student researchers — six from Lawrence and one from the University of Wisconsin-Fox Valley — will use the microscope to advance understanding in developmental biology, cell biology, physiology and biochemistry.

BACKGROUND: Recent genome-wide association studies (GWAS) have identified single nucleotide polymorphisms (SNP)s that are essentially unequivocally associated with peripheral cholesterol. Since the alleles of the APOE gene, which modulate peripheral cholesterol metabolism, and midlife plasma cholesterol are both associated with Alzheimer's disease (AD) risk, we have evaluated the hypothesis that SNPs associated with plasma cholesterol are also associated with AD.

RESULTS: Seventeen non-APOE SNPs reproducibly associated with cholesterol per GWAS were tested for association with AD in ~2,000 AD and ~4,000 non-AD subjects. As a group, these SNPs are associated with AD. Two SNPs in particular, rs3846662 and rs1532085, are …
