Thermophilic microbial communities that are active in a high-solids environment offer great potential for the discovery of industrially relevant enzymes that efficiently deconstruct bioenergy feedstocks. In this study, finished green waste compost was used as an inoculum source to enrich microbial communities and associated enzymes that hydrolyze cellulose and hemicellulose during thermophilic high-solids fermentation of the bioenergy feedstocks switchgrass and corn stover. Methods involving the disruption of enzyme and plant cell wall polysaccharide interactions were developed to recover xylanase and endoglucanase activity from deconstructed solids. Water temperatures off the south-western coast of Western Australia rose to unprecedented levels during February and March 2011, and this warming event has been termed a “marine heat wave”. While surface temperatures were more than 3°C above the long-term monthly average over an extended area in February 2011, the temperature in some localised areas in coastal waters exceeded the long-term monthly average by 5°C for periods of a day or two in late February/early March

The article discusses intellectual property law in light of the 10 structural elements of the Western cultural model of creativity. The author mentions Article I of the U.S. Constitution which grants the U.S. Congress the power to promote science and the useful arts. He argues that the law governing joint inventions and authorship is designed to cover collaborations, but it is not expansive enough to cover editors and literary agents.

The relationship between protein conformational dynamics and enzymatic reactions has been a fundamental focus in modern enzymology. Using single-molecule fluorescence resonance energy transfer (FRET) with a combined statistical data analysis approach, we have identified the intermittently appearing coherence of the enzymatic conformational state from the recorded single-molecule intensity-time trajectories of enzyme 6-hydroxymethyl-7,8-dihydropterin pyrophosphokinase (HPPK) in catalytic reaction. The coherent conformational state dynamics suggests that the enzymatic catalysis involves a multistep conformational motion along the coordinates of substrate-enzyme complex formation and product releasing, presenting as an extreme dynamic behavior intrinsically related to the time bunching effect that we have reported previously. …

Energy transfer plays a significant role in numerous chemical, physical, and biological processes. While the use of fluorescent proteins in Forster resonance energy transfer (FRET) studies of biomolecules is common, energy transfer between fluorescent proteins and inorganic nanoparticles has not been explored in detail. In this study, energy transfer from fluorescent phycobiliproteins to noble metal nanoparticles was analyzed. Solutions of B-phycoerythrin (B-PE) were mixed with colloidal Au and Ag nanoparticles and were characterized by steady-state and time-resolved fluorescence spectroscopy to determine the magnitude and mechanism of the energy transfer. This research focused on the experiences and views of lesbian parents and adult children of lesbian parents. The experiences of the members of lesbian-parented families determined the main areas that were explored. The lack of international and Irish research on the lived experiences of lesbian parents and their children prompted this investigation. The study was conducted using qualitative, semi-structured interviews. The sample consisted of three lesbian parents and two adult children of lesbian parents from two parent lesbian families. Quantifying the current carbon cycle of terrestrial ecosystems requires that we translate spatially sparse measurements into consistent, gridded flux estimates at the regional scale. This is particularly challenging in heterogeneous regions such as the northern forests of the United States. We use a network of 17 eddy covariance flux towers deployed across the Upper Midwest region of northern Wisconsin and Michigan and upscale flux observations from towers to the regional scale. Climate change scenarios suggest an increase in tropical ocean temperature by 1–3°C by 2099, potentially killing many coral reefs. But Arabian/Persian Gulf corals already exist in this future thermal environment predicted for most tropical reefs and survived severe bleaching in 2010, one of the hottest years on record. Exposure to 33–35°C was on average twice as long as in non-bleaching years. Gulf corals bleached after exposure to temperatures above 34°C for a total of 8 weeks of which 3 weeks were above 35°C. This is more heat than any other corals can survive, providing an insight into the present limits …

There is an absence of drought tolerant herbaceous perennial forage legume and herb options other than lucerne (Medicago sativa L.) for environments with Mediterranean-like climates common in extensive areas of Southern Australia, the Mediterranean basin, and Chile. Therefore, a collection of 174 forage perennial legume and herb entries from 103 species and 32 genera was evaluated for adaptation in a diverse range of Mediterranean climatic environments in Southern Australia. The seasonal rainfall distribution varied from moderately to highly winter dominant with long term average annual rainfall ranging from 318 to 655 mm. The entries were rated for productivity and …

This study reports an alternative approach to achieve vitrification where cells are pre-desiccated prior to cooling to cryogenic temperatures for storage. Chinese Hamster Ovary (CHO) cells suspended in a trehalose solution were rapidly and uniformly desiccated to a low moisture content (< 0.12 g of water per g of dry weight) using a spin-drying technique. Trehalose was also introduced into the cells using a high-capacity trehalose transporter (TRET1). Fourier Transform Infrared Spectroscopy (FTIR) was used to examine the uniformity of water concentration distribution in the spin-dried samples. 62% of the cells were shown to survive spin-drying in the presence of trehalose following immediate rehydration. The spin-dried samples were stored in liquid nitrogen (LN2) at a vitrified state. It was shown that following re-warming to room temperature and re-hydration with a fully complemented cell culture medium, 51% of the spin-dried and vitrified cells survived and demonstrated normal growth characteristics. Spin-drying is a novel strategy that can be used to improve cryopreservation outcome by promoting rapid vitrification.

The recent RAF inhibitor trial with PLX4032/RG7204 in late-stage mutant B-RAF melanoma patients has been lauded as a success story for personalized cancer therapy since short-term clinical responses were observed in the majority of patients. However, initial responses were followed by subsequent tumor re-growth, and a subset of patients showed intrinsic resistance. Bi-directional translational efforts are now essential to determine the mechanisms underlying acquired/secondary and intrinsic resistance to RAF inhibitors.

This article explores the evolving ways in which anthropologists, archaeologists, and the United States government have viewed Native American cultural heritage, especially in terms of burials and grave goods. I begin with a historical view of the looting and racism that plagued the disciplines since their inception, and move into the present, while examining how NAGPRA has enabled indigenous communities to have a voice concerning what happens to their heritage, thus transforming archaeology in beneficial and productive ways that were previously not thought possible.
