A Tight Coupling Between Β2Y97 And Β2F200 Of The GabaA Receptor Mediates Gaba Binding, Phu N. Tran, Kurt T. Laha, David A. Wagner
Biological Sciences Faculty Research and Publications
The GABAA receptor is an oligopentameric chloride channel that is activated via conformation changes induced upon the binding of the endogenous ligand, GABA, to the extracellular inter-subunit interfaces. Although dozens of amino acid residues at the α/β interface have been implicated in ligand binding, the structural elements that mediate ligand binding and receptor activation are not yet fully described. In this study, double-mutant cycle analysis was employed to test for possible interactions between several arginines (α1R67, α1R120, α1R132, and β2R207) and two aromatic residues (β2Y97 and β2F200) …

Peatlands are areas of land with a naturally accumulated layer ofpeat. These are formed under waterlogged conditions from carbonrich, dead and decaying plant material. In the UK mosses, mainlySphagnum species, are the main formers of peat.Peatlands are found in at least 175 countries – from the tropics to the poles– and cover around 4 million km2or 3% of the world’s land area. In Europe, peatlands extend to ca. 515,000 km2. The UK is amongst the top ten nationsof the world in terms of its total peatland area. The UK has between 9-15%of Europe’s peatland area (46,000-77,000 km2) and about 13% of …

A Sense Of Place: Examining Music-Based Tourism And Its Implications In Destination Venue Placement, Richard Bret Campbell
UNLV Theses, Dissertations, Professional Papers, and Capstones
Music, and the ability to make it, understand it and interpret it, is one of the characteristics that make us uniquely human. However, when it comes to seeing the performance of live music, there are other variables that come into play when determining where to see a concert. The potential connection between people, music, and a “sense of place” are explored in this examination of music-based tourism. Based on information from journals and related news media, and using parallels to the sports tourism industry, the paper found a distinct connection between humans and music in the context of human development, …

Identification And Characterization Of A Novel Thermohalophilic Esterase From The Red Sea: Atlantis Ii Brine Pool, Yasmine Mustafa Mohamed
Identification And Characterization Of A Novel Thermohalophilic Esterase From The Red Sea: Atlantis Ii Brine Pool, Yasmine Mustafa Mohamed
Theses and Dissertations
Industrial (white) biotechnology poses an increasing demand for novel biocatalysts that are robust under a wide range of conditions. Traditionally, biocatalysts were isolated from cultured isolates, however, less than 1% of microorganisms are culturable. Therefore, it became evident that the unculturable majority holds a great potential for the discovery of novel biocatalysts. Metagenomics is an invaluable tool for accessing the genomes of the uncultured majority and has led to the isolation of a large number of biocatalysts from various environments. Extreme environments such as hydrothermal vents, brine pools and glaciers, are an attractive source for biocatalysts. Biocatalysts from these environments …
Fighting A War You've Already Lost: Zombies And Zombis In Firefly/Serenity And Dollhouse, Gerry Canavan
Fighting A War You've Already Lost: Zombies And Zombis In Firefly/Serenity And Dollhouse, Gerry Canavan
English Faculty Research and Publications
This article explores the use of zombie imagery in two sf narratives created by Joss Whedon: Firefly (US 2002–3), Serenity (US 2005) and Dollhouse (US 2009–10). The translation of the zombie from its traditional horror-movie context to the far-future space opera of Firefly/Serenity and the near-future cyberpunk of Dollhouse reveals the zombie's allegorisation of the consequences of biopolitical governmentality and neoliberal capitalism. In both series zombies function as a figure for both the dehumanisation caused by state and market forces and the possibility of Utopian resistance to these forces.

Commercial High Technology Innovations Face Uncertain Future Amid Emerging "Brics" Compulsory Licensing And It Interoperability Frameworks, Lawrence A. Kogan
San Diego International Law Journal
The pathways that lead to the success of cutting-edge technologies are often fraught with risk, difficulty, and uncertainty. These issues are particularly prevalent under a regime involving lengthy time horizons for competent research, development, and commercialization, which may require regulatory approvals. These challenges are known to be endemic to capital-intensive technology development which requires significant follow-on funding, particularly in highly regulated industries such as life sciences (e.g., pharmaceuticals/biotechnology and electronic medical devices ) and clean technology (which may be subdivided into clean or renewable energy generation and clean or renewable energy efficiency technologies and services, the former having more direct …

Marketplace Of Memory: What The Brain Fitness Technology Industry Says About Us And How We Can Do Better, Peter J. Whitehouse
Marketplace Of Memory: What The Brain Fitness Technology Industry Says About Us And How We Can Do Better, Peter J. Whitehouse
Faculty Scholarship
In the therapeutic void created by over 20 failed Alzheimer's disease drugs during the past decade, a new marketplace of "brain fitness" technology products has emerged. Ranging from video games and computer software to mobile phone apps and hand-held devices, these commercial products promise to maintain and enhance the memory, concentration, visual and spatial skills, verbal recall, and executive functions of individual users. It is instructive to view these products as sociocultural objects deeply imbued with the values and ideologies of our age; consequently, this article offers a critique of the brain fitness technology marketplace while identifying limitations in the …

There is increasing concern about the toxicities and potential risks, both still poorly understood, of silver nanoparticles for the aquatic environment after their eventual release via wastewater discharges. In this study, the toxicities of sediment associated nano (<100 >nm)-, micron (2–3.5 μm)- and ionic (AgNO3)-Ag on the sediment-dwelling polychaete, Nereis diversicolor, were compared after 10 days of sediment exposure, using survival, DNA damage (comet assay) and bioaccumulation as endpoints. The nominal concentrations used in all exposure scenarios were 0, 1, 5, 10, 25, and 50 μg Ag/g dry weight (dw) sediment. Our results showed that Ag was able to cause …100>

Patently Impossible, Sean B. Seymore
Patently Impossible, Sean B. Seymore
Vanderbilt Law Review
The quest to achieve the impossible fuels creativity, spawns new fields of inquiry, illuminates old ones, and extends the frontiers of knowledge. It is difficult, however, to obtain a patent for an invention which seems impossible, incredible, or conflicts with well- established scientific principles. The principal patentability hurdle is operability, which an inventor cannot overcome if there is reason to doubt that the invention can really achieve the intended result. Despite its laudable gatekeeping role, this Article identifies two problems with the law of operability. First, though objective in theory, the operability analysis rests on subjective credibility assessments. These credibility …

The current study (N = 242) seeks to establish the relationship between traits known collectively as the Dark Triad – narcissism, psychopathy, Machiavellianism – and mating standards and preferences. Using a budget-allocation task, we correlated scores on the Dark Triad traits with mate preferences for a long-term and short-term mate. Men scoring high on the Dark Triad may be more indiscriminate than most when selecting for short-term mates in order to widen their prospects. Furthermore, those high on the Dark Triad – psychopathy in particular – tend to select for mates based on self-interest, assortative mating, or a predilection for …
Functional Characterization Of Putative Non-Specific Phospholipase C (Npc) In Arabidopsis Thaliana, Carlotta Antonetta Peters
Functional Characterization Of Putative Non-Specific Phospholipase C (Npc) In Arabidopsis Thaliana, Carlotta Antonetta Peters
Dissertations
Abstract Phospholipases are enzymes that hydrolyze phospholipids. In terms of the position of bond hydrolysis, phospholipases are classified into four major types: phospholipase C (PLC), phospholipase D (PLD), phospholipase A1 (PLA1) and phospholipase A2 (PLA2). PLC hydrolyzes phospholipids at the first phosphodiester bond, producing diacylglycerol (DAG) and a phosphorylated head group. Based on substrate specificity, PLC is divided into two distinctively different groups: phosphoinositide-specific phospholipase C (PI-PLC) and non-specific phospholipase C (NPC). There are six members of the NPC family in Arabidopsis, designated NPC1 through 6 that bears sequence homology to bacterial phosphatidylcholine hydrolyzing PLC (PC-PLC). However, their function remains …

Chromatin immunoprecipitation coupled with high throughput DNA Sequencing (ChIP-Seq) has emerged as a powerful tool for genome wide profiling of the binding sites of proteins associated with DNA such as histones and transcription factors. However, no peak calling program has gained consensus acceptance by the scientific community as the preferred tool for ChIP-Seq data analysis. Analyzing the large data sets generated by ChIP-Seq studies remains highly challenging for most molecular biology laboratories. Here we profile H3K27me3 enrichment sites in rice young endosperm using the ChIP-Seq approach and analyze the data using four peak calling algorithms (FindPeaks, PeakSeq, USeq, and MACS). The Pcdp1 Complex Coordinates The Activity Of Dynein Isoforms To Produce Wild-Type Ciliary Motility, Christen G. Dipetrillo, Elizabeth F. Smith
Dartmouth Scholarship
Generating the complex waveforms characteristic of beating cilia requires the coordinated activity of multiple dynein isoforms anchored to the axoneme. We previously identified a complex associated with the C1d projection of the central apparatus that includes primary ciliary dyskinesia protein 1 (Pcdp1). Reduced expression of complex members results in severe motility defects, indicating that C1d is essential for wild-type ciliary beating. To define a mechanism for Pcdp1/C1d regulation of motility, we took a functional and structural approach combined with mutants lacking C1d and distinct subsets of dynein arms. Unlike mutants completely lacking the central apparatus, dynein-driven microtubule sliding velocities are …
Age At Diagnosis Of Diabetes In Appalachia, Lawrence Barker, Robert Gerzoff, Richard Crespo, Molly Shrewsberry
Age At Diagnosis Of Diabetes In Appalachia, Lawrence Barker, Robert Gerzoff, Richard Crespo, Molly Shrewsberry
Family and Community Health
Background
Appalachia is a region of the United States noted for the poverty and poor health outcomes of its residents. Residents of the poorest Appalachian counties have a high prevalence of diabetes and risk factors (obesity, low income, low education, etc.) for type 2 diabetes. However, diabetes prevalence exceeds what these risk factors alone explain. Based on this, the history of poor health outcomes in Appalachia, and personally observed high rates of childhood obesity and lack of concern about prediabetes, we speculated that people in Appalachia with diagnosed diabetes might tend to be diagnosed younger than their non-Appalachian counterparts.

SUMMARY Understanding insulin-like growth factor-1 (IGF1) biology is of particular importance because, apart from its role in mediating growth, it plays key roles in cellular transformation, organ regeneration, immune function, development of the musculoskeletal system and aging. IGF1 bioactivity is modulated by its binding to IGF-binding proteins (IGFBPs) and the acid labile subunit (ALS), which are present in serum and tissues. To determine whether IGF1 binding to IGFBPs is necessary to facilitate normal growth and development, we used a gene-targeting approach and generated two novel knock-in mouse models of mutated IGF1, in which the native Igf1 gene was replaced by …

Models for contaminant transport in streams commonly idealize transient storage as a well-mixed but immobile system. These transient storage models capture rapid (near-stream) hyporheic storage and transport, but do not account for large-scale, stage-dependent interaction with the alluvial aquifer. The objective of this research was to document transient storage of phosphorus (P) in coarse gravel alluvium potentially influenced by large-scale, stage-dependent preferential flow pathways (PFPs). Long-term monitoring was performed at floodplain sites adjacent to the Barren Fork Creek and Honey Creek in northeastern Oklahoma. Based on results from subsurface electrical resistivity mapping which was correlated to hydraulic conductivity data, observation …
