Mouse Versus Rat: Profound Differences In Meiotic Regulation At The Level Of The Isolated Oocyte, Stephen M. Downs

Cumulus cell-enclosed oocytes (CEO), denuded oocytes (DO), or dissected follicles were obtained 44–48 hr after priming immature mice (20–23 days old) with 5 IU or immature rats (25–27 days old) with 12.5 IU of equine chorionic gonadotropin, and exposed to a variety of culture conditions. Mouse oocytes were more effectively maintained in meiotic arrest by hypoxanthine, dbcAMP, IBMX, milrinone, and 8-Br-cGMP. Atrial natriuretic peptide, a guanylate cyclase activator, suppressed maturation in CEO from both species, but mycophenolic acid reversed IBMX-maintained meiotic arrest in mouse CEO with little activity in rat CEO. Parenting is a primary site for the socialization of young children, including socialization around issues of race and racism. Giving careful attention to the implications of a socially privileged racial status, this study draws on the personal narratives of three White, middle-class, heterosexual mothers living in Chicago to improve understanding of White, middle-class parenting around issues of race and racism and to critically examine the ways parenting practices relate to larger social Discourses in the United States that perpetuate or disrupt White supremacy. The FDA employs an average-patient standard when reviewing drugs: it approves a drug only if is safe and effective for the average patient in a clinical trial. It is common, however, for patients to respond differently to a drug. Therefore, the average-patient standard can reject a drug that benefits certain patient subgroups (false negative) and even approval a drug that harms other patient subgroups (false positives). These errors increase the cost of drug development – and thus health care – by wasting research on unproductive or unapproved drugs. Under section 404(c) of the Clean Water Act, the Environmental Protection Agency (EPA) has the power to veto permits for the discharge of fill or dredged materials issued by the U.S. Army Corps of Engineers.  This veto power can only be used if the discharge will have unacceptable adverse effects on water supplies, aquatic life, wildlife, or recreational areas.  The EPA’s use of its veto power is rare and is usually invoked before a section 404 permit has been issued.  The recent veto of a section 404 permit for the Spruce No. 1 Mine in Logan County, West Virginia, by …

When social science methods are being employed in a new context — such as the assessment of international investment law — there is value in exploring the underlying assumptions and normative baselines of the enterprise. There is an increasingly sophisticated literature on the role played by museums in reaffirming social norms of examination in the antebellum United States. This literature has largely focused on the way museums presented this knowledge to the public, not how audiences reacted to claims of scientific authority. Diabetes exacerbates ischemic heart disease morbidity and mortality via incompletely understood mechanisms. Although adiponectin (APN) reduces myocardial ischemia/reperfusion (MI/R) injury in nondiabetic animals, whether APN's cardioprotective actions are altered in diabetes, a pathologic condition with endogenously reduced APN, has never been investigated. High-fat diet (HD)-induced diabetic mice and normal diet (ND) controls were subjected to MI via coronary artery ligation, and given vehicle or APN globular domain (gAPN, 2 μg/g) 10 min before reperfusion. Compared to ND mice (where gAPN exerted pronounced cardioprotection), HD mice manifested greater MI/R injury, and a tripled gAPN dose was requisite to achieve cardioprotective extent …

Attention-Deficit/Hyperactivity Disorder (ADHD) is one of the most prevalent mental health disorders of childhood, but despite the availability of several evidence-based interventions, Latino children are more likely than non-minority children to have an unmet need for services related to ADHD. Because parental beliefs about the etiology of ADHD likely influence which services are sought, more research is needed to examine this aspect of help-seeking behavior in order to address the unmet need among Latino families. Human genetic variation occurs more commonly than was recognized after the completion of the Human Genome Sequencing Project in 2003. Submicroscopic human DNA analysis has revealed copy number variation (CNV) as the deletion or duplication of a genomic region potentially affecting gene dosage. Advanced genetic research now includes the study of CNVs in diseased subject groups compared to in house controls or online published datasets of control CNV data. Research labs choose from different bioinformatic algorithms to make the copy number calls. Posttraumatic Stress Disorder (PTSD) is a response to trauma exposure that involves a number of symptoms that can be highly impairing to affected individuals. Only a subset of those exposed to traumatic events will develop the disorder, which is conceptualized as developing via conditional fear. Research into factors predisposing for PTSD is needed. Furthermore, little work has been done to investigate predisposing factors in children more specifically. This research tests the effects of stress exposure on subsequent fear learning, across developmental stages in mice, as a model for PTSD. A year ago while surfing the Internet, I came across a website called Garfield Minus Garfield (GMG), operated by artist/businessman Dan Walsh. Walsh received notoriety for his adaptations of the popular Garfield comic strip in which he removed its title character [1]. This act results in Garfield’s human counterpart, Jon Arbuckle, alone and talking to himself in the now empty space of the strip. Canada geese (Branta canadensis) can cause considerable damage to crops during summer when geese are flightless. We evaluated the effectiveness of a program designed to alleviate crop damage on soybeans by Canada geese in South Dakota, USA. The applications of electric fences, feeding stations, and propane cannons reduced the area of crop damage by 90% in 2006 and 80% in 2007, but the timing was important. Fields where abatement practices were applied early in the growing season had less damage than fields where they were applied later. Differences in resource acquisition between native and exotic plants is one hypothesis to explain invasive plant success. Mechanisms include greater resource acquisition rates and greater plasticity in resource acquisition by invasive exotic species compared to non-invasive natives. We assess the support for these mechanisms by comparing nitrate acquisition and growth of invasive annual and perennial grass seedlings in western North America. Two invasive exotic grasses (Bromus tectorum and Taeniatherum caput-medusae) and three perennial native and exotic grasses (Pseudoroegneria spicata, Elymus elymoides, and Agropyron cristatum) were grown at various temperatures typical of autumn and springtime when resource are abundant and dominance …

Follicular T helper (Tfh) cells provide critical help to B cells for germinal center (GC) formation. Mutations affecting SLAM-associated protein (SAP) prevent GC formation because of defective T cell-B cell interactions, yet effects on Tfh cell differentiation remain unclear. We describe the in vitro differentiation of functionally competent “Tfh-like” cells that expressed interleukin-21, Tfh cell markers, and Bcl6 and rescued GC formation in SAP-deficient hosts better than other T helper (Th) cells. SAP-deficient Tfh-like cells appeared virtually indistinguishable from wild-type, yet failed to support GCs in vivo. Urban stormwater runoff is excess runoff created by increased imperviousness in an urbanized watershed and can have significant impacts on both water quantity and quality. Natural communities and human communities are faced with damages that occur as a result of the change in the type and magnitude of stormwater runoff flows, including but not limited to increased flooding and degradation of natural aquatic systems. Diffusion coefficients (D) can be readily measured by nuclear magnetic resonance (NMR) spectroscopy and magnetic resonance imaging (MRI) instruments. Operators of these instruments often utilize standards with known diffusion coefficients to rapidly and conveniently test the performance of the NMR or MRI system to measure diffusion. A variety of these standards have been proposed in the scientific literature. This thesis describes a diffusion standard based on water constrained by container geometry, specifically water between tightly packed, parallel glass fiber filaments. Suicide is one of the leading causes of male mortality. In nearly every country in the world, more males than females end their life by suicide. Previous research indicates male-specific risk factors include social factors such as being unmarried, low income, and unemployment. An analytic model of male suicide is developed, proposing that the traditional male gender role creates a culturally-conditioned narrowing of perceived options and cognitive rigidity when under stress that increases male suicide risk. Family history of suicide is among the strongest predictors of suicide risk. From the context of gene by environment interactions, this manuscript presents a case study of the M family, which experienced 4 committed suicides within a short time period. Over the course of 5 years, the father and 3 sons committed suicide. Suicidal ideations developed in several other members of the family. The family9s suicide risk appears to have stemmed from both environmental and genetic factors, and likely from an interactive effect between both.
