Lignocellulosic ethanol offers a promising alternative to conventional fossil fuels. One among the major limitations in the lignocellulosic biomass hydrolysis is unavailability of efficient and environmentally biomass degrading technologies. Plant-based production of these enzymes on large scale offers a cost-effective solution. Cellulases, hemicellulases including mannanases and other accessory enzymes are required for conversion of lignocellulosic biomass into fermentable sugars. beta-mannanase catalyzes endo-hydrolysis of the mannan backbone, a major constituent of woody biomass. In this study, the man1 gene encoding beta-mannanase was isolated from Trichoderma reesei and expressed via the chloroplast genome. The CDKN2A/ARF locus encompasses overlapping tumor suppressor genes p16(INK4A) and p14(ARF), which are frequently co-deleted in human malignant mesothelioma (MM). The importance of p16(INK4A) loss in human cancer is well established, but the relative significance of p14(ARF) loss has been debated. The tumor predisposition of mice singly deficient for either Ink4a or Arf, due to targeting of exons 1 alpha or 1 beta, respectively, supports the idea that both play significant and nonredundant roles in suppressing spontaneous tumors. Reliable estimates of the impacts and costs of biological invasions are critical to developing credible management, trade and regulatory policies. Worldwide, forests and urban trees provide important ecosystem services as well as economic and social benefits, but are threatened by non-native insects. More than 450 non-native forest insects are established in the United States but estimates of broad-scale economic impacts associated with these species are largely unavailable. We developed a novel modeling approach that maximizes the use of available data, accounts for multiple sources of uncertainty, and provides cost estimates for three major feeding guilds of non-native forest insects. Accurately interpreting the imaging findings in patients with prior musculoskeletal tumors can be difficult. Because most patients have anatomical changes related to surgery in addition to postradiation and postchemotherapy changes, the radiologist must systematically and critically evaluate all available radiographs, sonograms, computed tomography, magnetic resonance imaging, and positron emission tomography/computed tomography scans to best differentiate normal posttreatment changes from residual or recurrent musculoskeletal tumor. Comparison with presurgical and postsurgical imaging is very important to detect subtle nodular tumor recurrence. Current treatment for type I diabetes includes delivery of insulin via injection or pump, which is highly invasive and expensive. The production of chloroplast-derived proinsulin should reduce cost and facilitate oral delivery. Therefore, tobacco and lettuce chloroplasts were transformed with the cholera toxin B subunit fused with human proinsulin (A, B, C peptides) containing three furin cleavage sites (CTB-PFx3). Transplastomic lines were confirmed for site-specific integration of transgene and homoplasmy. Old tobacco leaves accumulated proinsulin up to 47% of total leaf protein (TLP). Old lettuce leaves accumulated proinsulin up to 53% TLP. Accumulation was so stable that up to similar …

Gold nanoparticles are introduced to a fibrous nanocomposite material prepared from electrospinning a polyelectrolyte solution of poly(acrylic acid) (PAA) and poly(allylamine hydrochloride) (PAH). The functional groups in the fiber allow convenient thermal crosslinking of the fibers and the binding of tetrachloroaurate ions. Gold nanoparticle-modified nanofibers can be further treated by silver enhancement that increased the electrical conductivity of the nanofibers to 10(-2) S/cm. The FT-IR analysis of the nanocomposite fibers shows that the deposition of gold nanoparticles significantly enhances the IR absorption intensity of the polymer fibers, offering a potential sensing capability through enhanced FT-IR absorption of molecules. Evidence suggests that the C-terminal truncation of alpha-synuclein is equally important as aggregation of alpha-synuclein in Parkinson disease (PD). Our previous results showed that an endopeptidase, matrix metalloproteinase-3 (MMP3), was induced and activated in dopaminergic (DA) cells upon stress conditions. Here, we report that MMP3 cleaved alpha-synuclein in vitro and in vivo and that alpha-synuclein and MMP3 were co-localized in Lewy bodies (LB) in the postmortem brains of PD patients. Incubation of alpha-synuclein with the catalytic domain of MMP3 (cMMP3) resulted in generation of several peptides, and the peptide profiles of WT alpha-synuclein (WTsyn) and A53T mutant (A53Tsyn) were different. …

Human population growth and development reduce the area and quality of natural communities and lead to a decline of associated wildlife populations. Sciurus niger avicennia (Big Cypress Fox Squirrel), a state-listed threatened subspecies endemic to south Florida, appears sensitive to habitat fragmentation and fire regime. This research assessed the distribution and habitat use of the Big Cypress Fox Squirrel from interviews with biologists, private land owners, and golf course managers and by transect sampling in natural areas on public and private lands in southwest Florida. This paper describes the design and operation of a simple airtight container devised to facilitate the transfer of atmosphere-sensitive samples from a glovebox to the vacuum chamber of an analytical instrument such as a scanning electron microscope. The use of this device for characterizing the microstructure of highly hygroscopic strontium iodide ceramics by scanning electron microscopy is illustrated as an application example.

The effect of a pre-workout energy supplement on acute multi-joint resistance exercise was examined in eight resistance-trained college-age men. Subjects were randomly provided either a placebo (P) or a supplement (S: containing caffeine, taurine, glucuronolactone, creatine, beta-alanine, and the amino acids; leucine, isoleucine, valine, glutamine and arginine) 10 minutes prior to resistance exercise. Subjects performed 4 sets of no more than 10 repetitions of either barbell squat or bench press at 80% of their pre-determined 1 repetition-maximum (1RM) with 90 seconds of rest between sets. Dietary intake 24 hours prior to each of the two training trials was kept constant. …

Long-distance dispersal (LDD) events, although rare for most plant species, can strongly influence population and community dynamics. Animals function as a key biotic vector of seeds and thus, a mechanistic and quantitative understanding of how individual animal behaviors scale to dispersal patterns at different spatial scales is a question of critical importance from both basic and applied perspectives. Using a diffusion-theory based analytical approach for a wide range of animal movement and seed transportation patterns, we show that the scale (a measure of local dispersal) of the seed dispersal kernel increases with the organisms' rate of movement and mean seed …

Many phenomena, including life itself and its biochemical foundations are fundamentally rooted in chirality. Combinatorial methodologies for catalyst discovery and optimization remain an invaluable tool for gaining access to enantiomerically pure compounds in the development of pharmaceuticals, agrochemicals, and flavors. Some exotic metamaterials exhibiting negative refractive index at optical frequencies are based on chiral structures. Chiroptical activity is commonly quantified in terms of circular dichroism (CD) and optical rotatory dispersion (ORD). We employ helium-atom scattering (HAS) and density functional theory (DFT) based on the ultrasoft pseudopotential scheme and the plane-wave basis set to investigate the strain and stress balance in nanopatterned N/Cu(001) surfaces. HAS shows that, with increasing N coverage (and decreasing stripe widths), the stress-relief-driven lateral expansion of the averaged lattice parameter within finite-sized N-containing patches reduces from 3.5% to 1.8% and that, beyond a critical exposure, the lateral expansion of the patches increases again slightly, to 2.4%. The latter implies that in this higher coverage range the compressive stress is partially relieved via another mechanism, which turns out to …

We consider a bistable differential-difference equation with inhomogeneous diffusion. Employing a piecewise linear nonlinearity, often referred to as McKean's caricature of the cubic, we construct front solutions which correspond, in the case of homogeneous diffusion, to monotone traveling front solutions or, in the case of propagation failure, to stationary front solutions. A general form for these fronts is given for essentially arbitrary inhomogeneous discrete diffusion, and conditions are given for the existence of solutions to the original discrete Nagumo equation. Background: Gold nanoparticles (AuNPs) scatter light intensely at or near their surface plasmon wavelength region. Using AuNPs coupled with dynamic light scattering (DLS) detection, we developed a facile nanoparticle immunoassay for serum protein biomarker detection and analysis. A serum sample was first mixed with a citrate-protected AuNP solution. Proteins from the serum were adsorbed to the AuNPs to form a protein corona on the nanoparticle surface. An antibody solution was then added to the assay solution to analyze the target proteins of interest that are present in the protein corona. We demonstrate the fabrication of reduced graphene oxide (RGO) Schottky diodes via dielectrophoretic (DEP) assembly of RGO between two dissimilar metal contacts. Titanium (Ti) was used to make a Schottky contact, while palladium (Pd) was used to make an Ohmic contact. From the current-voltage characteristics, we obtain rectifying behavior with a rectification ratio of up to 600. The ideality factor was high (4.9), possibly due to the presence of a large number of defects in the RGO sheets. The forward biased turn-on voltage was 1V, whereas the reverse biased breakdown voltage was -3.1 V, which improved further upon mild annealing …

Evidence is presented for the nuclear presence of a functional heteromeric complex of epidermal growth factor (EGFR), Src and the Signal Transducer and Activator of Transcription (Stat) 3 proteins in pancreatic cancer cells. Stat3 remains nuclear and associated with Src or EGFR, respectively, upon the siRNA knockdown of EGFR or Src, demonstrating the resistance of the complex to the modulation of EGFR or Src alone. Significantly, chromatin immunoprecipitation (ChIP) analyses reveal the nuclear EGFR, Src and Stat3 complex is bound to the c-Myc promoter. The optimal dose of medetomidine-ketamine-buprenorphine was determined in 25 Cape ground squirrels (Xerus inauris) undergoing surgical implantation of a temperature logger into the abdominal cavity. At the end of anaesthesia, the squirrels were given atipamezole intramuscularly to reverse the effects of medetomidine. The mean dose of medetomidine was 67.6 +/- 9.4 mu g/kg, keta mine 13.6 +/- 1.9 mg/kg and buprenorphine 0.5 +/- 0.06 mu g/kg. Induction time was 3.1 +/- 1.4 min. This produced surgical anaesthesia for 21 +/- 4.2 min. Atipamezole 232 +/- 92 mu g/kg produced a rapid recovery. Squirrels were sternally recumbent in 3.5 +/- 2.2 …

This qualitative study included women who had gone through the menopausal transition and had experienced obesity, and it focused on their weight histories and experiences across the life course. The goal of this research was to add to the body of knowledge concerning weight gain by applying a novel middle range theory (story theory). The purpose of this thesis is to show that stable isotope analysis can be used to predict K’inich Yax K’uk Mo’s stable isotope measures based on Stuart’s (2007) hypothesis that K’inich Yax K’uk Mo’, the dynastic founder of the Copan royal lineage, was a Caracol lord. There is significant and convincing evidence that K’inich Yak K’uk Mo’ had a non-Copanec origin. Stable isotope analysis is a tested and reliable method for detailing diets and migratory paths of ancient humans and this theory is applied as a predictor of the stable isotope measures of K’inich Yax K’uk Mo’, if he did …

John Rawls‟s stated intergenerational justice scheme, known as the just-savings principle, does not include an institutional concern for the environment and is therefore incomplete and incapable of maintaining meaningfully just relations between generations. The theory‟s emphasis on economic theory and capital accumulation demonstrates a misinterpretation of environmental issues and concerns as well as their underlying causes and repercussions. This lapse in Rawls‟s intergenerational scheme exposes flaws in his larger theory of justice by leaving the stability of society in question and placing arbitrary burdens on generations and peoples without institutional recourse. The purpose of this study was to determine the relationship of participation in single-gender classrooms on student performance on the reading and mathematics developmental scale scores (DSS) of third, fourth, and fifth grade students on the Florida Comprehensive Assessment Test (FCAT). The FCAT is a standardized test that is administered to all grade 3 through grade 10 public school students in the state of Florida and has been used to assess students‟ achievement in reading and mathematics. This study investigated the influence of Tyler Perry‟s House of Payne and Meet the Browns on black viewers‟ racial identity, based on a survey of 145 members of four predominantly African American churches in the Central Florida area. Mirroring Allen, Dawson, and Brown‟s (1989) model of an African American racial belief system, this study proposed that both shows would positively influence three dimensions of the black identity including closeness to blacks, black separatism, and the belief in positive stereotypes about blacks, while negatively influencing the dimension that emphasizes negative stereotypes about blacks. This thesis focuses on the use of geographic information systems (GIS) to examine mortuary practices in the Romano-Byzantine period Kellis 2 cemetery located in the Dakhleh Oasis, Egypt. The first research objective examines the relationship between age, sex and grave substructures of 701 burials in Kellis 2 cemetery. The aim of this research objective was to determine if the presence and style of grave substructures were influenced by sex or age. Although not statistically significant, GIS analysis revealed that most of the graves in the Kellis 2 cemetery have no associated substructures, but of those that did have associated substructures, …

Settlement and growth of quagga mussels Dreissena rostriformis bugensis were monitored in Lake Mead, NV, USA, where the first dreissenid occurrence was confirmed in the western United States. To measure the settlement rate of these invasive mussels, seven acrylonitrile butadiene styrene (ABS) pipes were attached to a line in shallow water (7.7 m below the surface) since November 23, 2007; eight ABS pipes were placed on another line in deep water (13.4 m below the surface) since January 3, 2008. Quagga mussels were sampled from these pipes on March 19, May 21, July 9, October 20, and December 19, 2008. …

The 100th Meridian Initiative was developed to help prevent the spread of aquatic nuisance species (ANS) through boater education and research on boater movement patterns and behaviors. Surveys employing these elements were conducted at Lake Mead National Recreation Area (LMNRA) in 2002-2003 before the discovery of the quagga mussel (Dreissena rostiformis bugensis Andrusov 1897) and in 2007-2008 after an established population of quagga mussels was found in the Lake. Following the discovery of quagga mussels in Lake Mead, Nevada-Arizona, USA, a variety of federal, state and regional agencies set up monitoring programs to evaluate and gain information to help minimize the impacts, or potential impacts, of quagga mussels to their facilities and lake ecology. While the agencies have worked closely and shared monitoring data and findings from the beginning of the infestation, there has been no documented comprehensive monitoring program to describe and record the various quagga mussel-related monitoring needs. Ad hoc interagency quagga mussel meeting representatives established an Interagency Monitoring Action Plan (I-MAP), which outlines agency objectives related …
