In recent years, the incidence of fungal infections has been rising all over the world. Although the amount of research in the field of pathogenic fungi has also increased, there is still a need for studies on fungal virulence. In this investigation, we focused on testing bloodstream-identified Candida albicans in experimental in vitro virulence assays. The proteinase enzyme activities of 30 C. albicans strains isolated from the bloodstream and sterile body fluids were investigated (15 colonizations, 15 infections). Polyphenol oxidases (PPOs EC 1.14.18.1) were isolated from Mucuna pruriens and Mucuna prurita and confirmed as tyrosinases involved in L-DOPA production. PPOs were extracted by using a 0.05 M phosphate buffer, pH 7.0. The purified enzyme was resolved into a single band by PAGE, the enzyme was confirmed as PPO by activity staining, and SDS-PAGE analysis revealed that the purified PPO of both the species is a tetramer. Substrate specificity experiments were carried out with catechol, L-DOPA, L-tyrosine, and p-cresol. A rapid protocol using shoot tip explants for micropropagation of Diyarbakır watermelon [Citrullus lanatus (Thunb.) Matsum. & Nakai] types Sürme, Beyazkış and Karakış was achieved. Shoot tips from 5-day-old in vitro germinated seedlings were cultured on shoot regeneration medium for 3 weeks. The effects of the different concentrations of benzyl adenine (BA) and carbohydrate types on shoot proliferation were examined. The results confirmed that a range of 0.5-1.0 mg/L of BA was almost equally effective in promoting the shoot length of cultures in the 3 genotypes. The fruits of Carica papaya L. (Caricaceae) are valuable as food and are also used in traditional medicine. The present study was designed to assess the antioxidant potential of the juices of 3 papaya cultivars (PCJ): Sunrise Solo, Red Lady, and Tainung. The antioxidant capacity of PCJ obtained from fully ripened fruit was determined by the following methods: scavenging of the free radical 1,1-diphenyl-2-picrylhydrazyl (DPPH), reducing power assay, scavenging of superoxide radicals, 2-deoxyribose oxidation assay, and thiocyanate assay. The chemical composition and nutritional value of a wild edible ectomycorrhizal mushroom from southwestern Anatolia, Tricholoma anatolicum, were analyzed. Moisture, crude oil, protein, ash, total carbohydrate content, and mineral composition of the mushrooms studied, including Fe, Na, K, Zn, Cu, Ca, Cd, and Pb, were determined. The energy values of the samples were also calculated. The analyses were conducted during the 3 different growing stages of the mushrooms: mycelium, young fruiting bodies, and mature fruiting bodies. The highest values for moisture and crude oil contents were found to be in the mycelium, ash, and carbohydrate content of young fruiting bodies. …

In recent years, emphasis has been placed on the use of natural materials in the control and treatment of various infections, as some chemically synthesized drugs have undesirable side effects. In this study, the ethanol extract of Iranian walnut leaves was examined for antibacterial activities on Streptococcus mutans, Streptococcus salivarius, Streptococcus sanguinis, and Actinomyces viscosus using the microdilution method. Total phenols, flavonoids, and flavonols were also determined colorimetrically. The minimum inhibitory concentrations (MIC) for ethanolic extract ranged between 15.6 and 187.5 mg/mL and minimum bactericidal concentrations (MBC) ranged between 31.25 and 250 mg/mL. The volatile compounds of 13 Turkish pine (Pinus brutia Ten.) honey samples were characterized by solid phase microextraction (SPME) analysis, followed by gas chromatography-mass spectrometry (GC-MS) analysis. A total of 42 volatile compounds were identified, but volatile compounds such as nonana1, benzene, 4-hexen-3-ol, alpha-pinene, and 2-heptanone were recognized to be specific floral origin markers of the pine honey. The SPME extraction method was proposed as an alternative way to carry out pollen analysis for floral source detection, especially for products such as pine honey, by characterization of honeydew elements.

In the present study, the 2n = 54 chromosomal race of blind mole rats, Nannospalax xanthodon superspecies, from Kırıkkale Province in Turkey was investigated. Conventional chromosome staining, Ag-NOR (Nucleolus Organizer Region) staining, and C-banding analysis were carried out on specimens of mole rats. The karyotype including 3 metacentric pairs (nos. 1-3), 3 submetacentric pairs (nos. 4-6), 3 subtelocentric pairs (nos. 7-9), and 17 acrocentric pairs (nos. 10-26) of autosomes (NFa = 70). Mitotic abnormalities, chromosomal behavior, and protein content in wheat (Triticum durum) due to imazethapyr (IM) treatment were determined. Wheat seeds (variety HI-8663) were exposed to 5 IM concentrations (0.5, 0.89, 1.58, 2.81, and 5 ppm) according to the between paper method, in laboratory conditions. The protein content in the root-shoot axis and endosperm decreased significantly with the dosage increase. Cytological observations showed that the mitotic frequency in the root meristematic cells decreased parallel to the increase in concentrations. The most common types of the observed anomalies were sticky chromosomes, lagging chromosomes, scattered chromosomes, chromatin bridges, and micronuclei. The results revealed …

Legionella pneumophila (L. pneumophila), known to cause legionnaires' disease, has been found in various aquatic environments and man-made water systems. Free living amoebas (FLAs) live in the same environments and can cause infections such as encephalitis, keratitis. Moreover, FLAs play a major role in L. pneumophila transmission. In the present study, the presence of L. pneumophila and FLAs was examined in water and swab samples obtained from shower heads (hot water) in 61 houses in İstanbul. The effects of various carbon and nitrogen sources on production of \alpha-amylase by Bacillus subtilis and its mutant derivates were investigated. The maximum production of \alpha-amylase by all strains was obtained in the presence of mesoinositol as the carbon source. The mitochondrial cytochrome oxidase I (COI) gene region of Cephus pygmeus (L.) (Hymenoptera: Cephidae) was characterized at the nucleotide and amino acid levels. The partial sequence (687 bp) of the gene was determined and compared with other cephids and insect species at both the nucleic acid and amino acid sequence levels. The examined fragment was placed at an equal distance to both ends of the gene and consisted of 13 structural regions. The suprachiasmatic nucleus (SCN) generates and controls the circadian rhythms in mammals including the rhythm of locomotor activity. Leptin is a hormone secreted by adipose tissue that informs the brain about the fat stores. SCN neurons express leptin receptors. Here we investigated the effects of 3 different leptin administrations on the locomotor activity of the Syrian hamsters maintained in constant darkness. The objectives of the present study were to determine the effects of essential oil from Turkish oregano (Origanum onites L.) and its 2 major phenolic components, carvacrol and thymol, on cell viability, and to investigate their cytoprotective (antioxidant) effects against hydrogen peroxide-induced cytotoxicity and membrane damage in hepatoma G2 (Hep G2) cells. This study describes, for the first time, an in vitro protocol for the development of somatic embryos and shoots from a callus derived from cotyledonary leaf segments excised from 3-week-old in vitro-germinated seedlings of Digitalis lamarckii Ivan. (dwarf foxglove), an endemic medicinal species of Turkey. The embryogenic callus was induced on Murashige and Skoog (MS) medium containing 0.54 \muM \mu-naphthalene acetic acid (NAA) and 2.22 \muM 6-benzylaminopurine (BAP). A secondary metabolite, 2^'-O-ethylmurrangatin (1), was isolated from the leaves of a medicinal plant, Murraya paniculata. The structure of compound 1 was identified with the help of spectroscopic techniques. Compound 1 was screened for its antioxidant, lipoxygenase, prolyl endopeptidase, and respiratory burst inhibitory activities. 2^'-O-Ethylmurrangatin (1) was found to have significant activity against lipoxygenase enzyme and moderate respiratory burst inhibitory activity, but no activity against either DPPH radical scavenging assay or prolyl endopeptidase enzyme.

Ethanolic (EEAA) and sesquiterpene lactone (SEAA) extracts of the aerial parts of Anthemis aciphylla var. aciphylla Boiss. (Asteraceae) were evaluated for anti-inflammatory activity on carrageenin-induced paw edema (acute model) and cotton pellet-induced granuloma (chronic model) in rats. In carrageenin-induced paw edema, EEAA and SEAA at intraperitoneal doses of 50, 100, and 200 mg/kg dose-dependently inhibited the paw edema. In cotton pellet-induced granuloma, the oral administration of EEAA and SEAA at 50, 100, and 200 mg/kg dosages was also found to significantly inhibit granuloma tissue formation.

Rains during grain ripening in wheat (Triticum aestivum L.) can cause preharvest sprouting (PHS), which drastically affects the grain yield and the baking quality of flour. In the present study, to screen 7 cultivars and 4 lines of spring bread wheat for PHS, germination tests were conducted with seeds obtained from wet spikes harvested immediately after natural rainfall. Effect Of Exogenously Applied Spermidine On Growth And Physiology Of Citrus Rootstock Troyer Citrange Under Saline Conditions, Muhammad Akbar Anjum

The aim of this study was to assess the effects of spermidine (Spd) on ameliorating adverse effects of salinity in Troyer citrange seedlings. For this purpose, 6-month-old, uniform-sized seedlings of Troyer citrange were transplanted to plastic containers containing Hoagland nutrient solution. Addition of 75 mM NaCl into the nutrient solution reduced the plant growth, leaf chlorophyll content, chlorophyll fluorescence yield (Fv/Fm), net photosynthetic rate, respiration rate, and total Spd, N, and Ca^{2+} + Mg^{2+} contents of the leaves. Hacıhaliloğlu is one of the most widely cultivated apricot (Prunus armeniaca L.) cultivars in Turkey. It produces fruit of excellent quality and is consumed both fresh and dried. In this study several factors affecting in vitro culture, rapid proliferation, rooting, and acclimatization of the apricot cultivar Hacıhaliloğlu were examined. During this study significant differences in harvesting dates were observed, with the month of May being noted as the best time to examine the in vitro culture of Hacıhaliloğlu. The objective of this paper was to develop an artificial neural network (ANN) model in order to predict monthly mean soil temperature for the present month by using various previous monthly mean meteorological variables. For this purpose, the measured soil temperature and other meteorological data between the years of 2000 and 2007 at Adana meteorological station were used. In this study, 42 bread wheat pure lines obtained from 340 landraces, collected from 35 provinces of 7 regions in Turkey, and 7 modern bread wheat cultivars were evaluated under rainfed conditions in Konya, in 2005-2006 and 2006-2007 growing seasons. Experiments were conducted in a 7 × 7 lattice design in 3 replicates. Both genotype-traits (GT) biplot analysis and path analysis were used to investigate the relationships between grain yield and 12 traits. The aim of this study is to investigate in-vitro antimicrobial effects of the essential oils from oregano (Origanum acutidens and Origanum rotundifolium) and thyme (Thymus sipyleus subsp. sipyleus var. rosulans). The chemical composition and antimicrobial attributes of the essential oils obtained from the aerial parts of the plants, of which there were 3 Lamiaceae species, have been studied.
