Department of Biochemistry: Faculty Publications

Hydrogen peroxide is thought to regulate cellular processes by direct oxidation of numerous cellular proteins, whereas antioxidants, most notably thiol peroxidases, are thought to reduce peroxides and inhibit H2O2 response. However, thiol peroxidases have also been implicated in activation of transcription factors and signaling. It remains unclear if these enzymes stimulate or inhibit redox regulation and whether this regulation is widespread or limited to a few cellular components. Herein, we found that Saccharomyces cerevisiae cells lacking all eight thiol peroxidases were viable and withstood redox stresses. They transcriptionally responded to various redox treatments, but were unable to …

Laser-Induced Breakdown Spectroscopy (Libs): An Innovative Tool For Studying Bacteria, Qassem I. Mohaidat Jan 2011

Laser-Induced Breakdown Spectroscopy (Libs): An Innovative Tool For Studying Bacteria, Qassem I. Mohaidat

Wayne State University Dissertations

Laser-induced breakdown spectroscopy (LIBS) has gained a reputation as a flexible and convenient technique for rapidly determining the elemental composition of samples with minimal or no sample preparation. In this dissertation, I will describe the benefits of using LIBS for the rapid discrimination and identification of bacteria (both pathogenic and non-pathogenic) based on the relative concentration of trace inorganic elements such as Mg, P, Ca, and Na. The speed, portability, and robustness of the technique suggest that LIBS may be applicable as a rapid point-of-care medical diagnostic technology.

LIBS spectra of multiple genera of bacteria such as Escherichia, Streptococcus, Mycobacterium, …

A Multi-Species Analysis Of Biomechanical Responses Of The Head To A Shock Wave, Richard Bolander Jan 2011

A Multi-Species Analysis Of Biomechanical Responses Of The Head To A Shock Wave, Richard Bolander

Wayne State University Dissertations

Shock wave induced brain injury remains a field of research that has great consequences for the rehabilitation of soldiers and civilians that are exposed to an explosion. As such, for the research to be successful in developing strategies to mitigate the effects of these injuries, appropriate research methods need to be developed. Animal models are currently employed to understand the brain's response to a shock wave exposure. Unfortunately no criteria have been established that indicates in what way the mechanical inputs that the cells in an animal's brain are subjected to are similar to a human. The purpose of this …

The Relationship Between Study Abroad And Motivation, Attitude And Anxiety In University Students Learning A Foreign Language, Stefania Gabriella Morreale Jan 2011

The Relationship Between Study Abroad And Motivation, Attitude And Anxiety In University Students Learning A Foreign Language, Stefania Gabriella Morreale

Wayne State University Dissertations

The objective of this dissertation was to discover the program effects of long- and short-term study abroad for university students at a midwestern, public university. Affective outcomes, such as motivation, attitude and anxiety, were examined using Gardner's Attitudes and Motivational Test Battery (AMTB) survey and a student demographic questionnaire. Quantitative data from the student demographic survey reported that the typical foreign language student surveyed is female, between the ages of 18-22, a junior, a liberal arts major with non-native fluency in the target language. The typical study abroad program attended by this sample is: short-term (less than eight weeks), most …

Analysis Of Gaba And Glutamate In The Mouse Dorsal Striatum, Stella Wisidagamage Jan 2011

Analysis Of Gaba And Glutamate In The Mouse Dorsal Striatum, Stella Wisidagamage

Wayne State University Theses

ABSTRACT

ANALYSIS OF GABA AND GLUTAMATE IN MICE BRAIN

by

STELLA WISIDAGAMA

May 2011

Advisor: Dr. Tiffany Mathews

Major: Chemistry (Analytical)

Degree: Master of Science

GABA and glutamate are the predominant inhibitory and excitatory amino acid neurotransmitters in the central nervous system (CNS), respectively. However, their altered levels cause several neurological diseases including Parkinson's and Huntington's disease, schizophrenia, and epilepsy. It is important to measure their levels in the extra- and intracellular environments to better understand and develop improved therapeutics that will treat these neurological disorders. The principle aim of this study is to study the impacts of different endogenous …

Supercritical Carbon Dioxide-Processed Resorbable Polymer Nanocomposites For Bone Graft Substitute Applications, Kevin Baker Jan 2011

Supercritical Carbon Dioxide-Processed Resorbable Polymer Nanocomposites For Bone Graft Substitute Applications, Kevin Baker

Wayne State University Dissertations

Numerous clinical situations necessitate the use of bone graft materials to enhance bone formation. While autologous and allogenic materials are considered the gold standards in the setting of fracture healing and spine fusion, their disadvantages, which include donor site morbidity and finite supply have stimulated research and development of novel bone graft substitute materials. Among the most promising candidate materials are resorbable polymers, composed of lactic and/or glycolic acid. While the characteristics of these materials, such as predictable degradation kinetics and biocompatibility, make them an excellent choice for bone graft substitute applications, they lack mechanical strength when synthesized with the …

Computational Simulation Of Skull Fracture Patterns In Pediatric Subjects Using A Porcine Model, Christina Devito Wagner Jan 2011

Computational Simulation Of Skull Fracture Patterns In Pediatric Subjects Using A Porcine Model, Christina Devito Wagner

Wayne State University Dissertations

In cases of suspected child abuse with skeletal trauma, it is often the role of the injury biomechanist, forensic pathologist, clinical radiologist, and forensic anthropologist to determine the mechanism of injury when the child victims cannot speak for themselves. This is a challenging task, especially for the head, as comprehensive biomechanical data on skull fracture in infants and children do not currently exist, and frequently the determination regarding cause of injury is based on anecdotal evidence from the medical literature and unsubstantiated eyewitness accounts. The current process may result in unreliable autopsy interpretation and miscarriages of justice due to a …

A New Hepato-Protective Flavone Glycoside From The Flowers Of Onopordum Alexandrinum Growing In Egypt, Maha Salama, Shahira Mohammed Ezzat Jan 2011

A New Hepato-Protective Flavone Glycoside From The Flowers Of Onopordum Alexandrinum Growing In Egypt, Maha Salama, Shahira Mohammed Ezzat

Pharmacy

A bioactivity-guided fractionation of the ethyl acetate fraction of the flowers of Onopordum alexandrinum L. (Asteraceae) yielded a new flavonoidal glycoside designated as acacetin-7-O-galacturonide (9), alongside with nine known flavonoids; 6-methoxy-apigenin (hispidulin) (1), acacetin (2), apigenin (3), luteolin (4), kaempferol (5), eriodictyol (6), apigenin-7-O-glucoside (7), luteolin-7-O-glucoside (8), and kaempferol-3-O-rutinoside (10). The compounds were assayed for their hepatoprotective activity against CCl4-induced hepatic cell damage in rats and free radical scavenging activity using 2,2-diphenyl-1-picrylhydrazyl (DPPH). Compounds 4, 6, 9, and 10 have not been previously reported from flowers of O. alexandrinum L., and this is the first report of acacetin-7-O-galacturonide (9) in …
