The Differential Effect Of Toxoplasma Gondii Infection On The Stability Of Bcl2-Family Members Involves Multiple Activities, John Cherrington Carmen, Anthony Peter Sinai

Microbiology, Immunology, and Molecular Genetics Faculty Publications

The regulation of mitochondrial permeability, a key event in the initiation of apoptosis is governed by the opposing actions of the pro- and anti-apoptotic members of the BCL2-family of proteins. The BCL2-family can be classified further based on the number of BCL-homology (BH) domains they encode. Pathogen mediated modulation of BCL2-family members play a significant role in their ability to affect the apoptotic pathways in the infected host cell. The protozoan parasite Toxoplasma gondii establishes a profound blockade of apoptosis noted by a requirement for host NFκB activity and correlating with the selective degradation of pro-apoptotic BCL2-family members. In this …

Muscle Atrophy Induced By Sod1(G93a) Expression Does Not Involve The Activation Of Caspase In The Absence Of Denervation, Gabriella Dobrowolny, Michela Aucello, Antonio Musarò

Research outputs 2011

Background: The most remarkable feature of skeletal muscle is the capacity to adapt its morphological, biochemical and molecular properties in response to several factors. Nonetheless, under pathological conditions, skeletal muscle loses its adaptability, leading to atrophy or wasting. Several signals might function as physiopathological triggers of muscle atrophy. However, the specific mechanisms underlying the atrophic phenotype under different pathological conditions remain to be fully elucidated. In this paper, we address the involvement of caspases in the induction of muscle atrophy in experimental models of amyotrophic lateral sclerosis (ALS) expressing the mutant SOD1G93A transgene either locally or ubiquitously.

Unsupplemented Artemia Diet Results In Reduced Growth And Jaw Dysmorphogenesis In Zebrafish, Michael P. Craig, Mitul B. Desai, Kate E. Olukalns, Scott E. Afton, Joseph A. Caruso, Jay R. Hove

Biochemistry and Molecular Biology Faculty Publications

The number of laboratories using zebrafish as an experimental animal model has risen tremendously over the past two decades (Craig et al., 2006). As a result, the number of zebrafish facilities around the world has dramatically increased to meet the elevated demand for proper animal care and maintenance. In order to meet this demand, aquaculture facilities must employ husbandry protocols designed to produce a constant supply of healthy, viable eggs. Surprisingly, many husbandry strategies, particularly feeding protocols, are frequently passed down from members of one lab to another in a colloquial fashion without rigorous experimental validation. An ideal diet should …

Temperature 1 Self-Assembly: Deterministic Assembly In 3d And Probabilistic Assembly In 2d, Matthew Cook, Yunhui Fu, Robert Schweller

Computer Science Faculty Publications

We investigate the power of the Wang tile self-assembly model at temperature 1, a threshold value that permits attachment between any two tiles that share even a single bond. When restricted to deterministic assembly in the plane, no temperature 1 assembly system has been shown to build a shape with a tile complexity smaller than the diameter of the shape. In contrast, we show that temperature 1 self-assembly in 3 dimensions, even when growth is restricted to at most 1 step into the third dimension, is capable of simulating a large class of temperature 2 systems, in turn permitting the …

Ocean Process Analysis Laboratory

Ocean surface layer carbon dioxide (CO2) data collected in the Gulf of Maine from 2004 to 2008 are presented. Monthly shipboard observations are combined with additional higher‐resolution CO2 observations to characterize CO2 fugacity ( fCO2) and CO2 flux over hourly to interannual time scales. Observed fCO2 andCO2 flux dynamics are dominated by a seasonal cycle, with a large spring influx of CO2 and a fall‐to‐winter efflux back to the atmosphere. The temporal results at inner, middle, and outer shelf locations are highly correlated, and observed spatial variability is generally small relative to the monthly to seasonal temporal changes. The averaged …

Inhibition Of Proprotein Convertase Ski-1 Blocks Transcription Of Key Extracellular Matrix Genes Regulating Osteoblastic Mineralization, Jeff P. Gorski, Nichole T. Huffman, Sridar Chittur, Ronald J. Midura, Claudine Black, Julia Oxford, Nabil G. Seidah

Biomedical Research Institute Publications and Presentations

Mineralization, a characteristic phenotypic property of osteoblastic lineage cells, was blocked by AEBSF and dec-RRLL-cmk, inhibitors of SKI-1 (site 1; subtilisin kexin like-1) protease. Since SKI-1 is required for activation of SREBP and CREB/ATF family transcription factors, we tested the effect of these inhibitors on gene expression. AEBSF decreased expression of 140 genes by 1.5- to 3.0-fold including Phex, Dmp1, COL1A1, COL11A1 and fibronectin. Direct comparison of AEBSF and dec-RRLL-cmk, a more specific SKI-1 inhibitor, demonstrated that expression of Phex, Dmp1, COL11A1 and fibronectin was reduced by both while COL1A2 and HMGCS1 were reduced …

Tests Of Integrin Transmembrane Domain Homo-Oligomerization During Integrin Ligand Binding And Signaling, Wei Wang, Jieqing Zhu, Timothy A. Springer, Bing Hao Luo Jan 2011

Tests Of Integrin Transmembrane Domain Homo-Oligomerization During Integrin Ligand Binding And Signaling, Wei Wang, Jieqing Zhu, Timothy A. Springer, Bing Hao Luo

Faculty Publications

Integrin transmembrane (TM) and/or cytoplasmic domains play a critical role in integrin bidirectional signaling. Although it has been shown that TM and/or cytoplasmic α and β domains associate in the resting state and separation of these domains is required for both inside-out and outside-in signaling, the role of TM homomeric association remains elusive. Formation of TM homo-oligomers was observed in micelles and bacterial membranes previously, and it has been proposed that homomeric association is important for integrin activation and clustering. This study addresses whether integrin TM domains form homo-oligomers in mammalian cell membranes using cysteine scanning mutagenesis. Our results show …

Operation Of A Dna-Based Autocatalytic Network In Serum, Elton Graugnard, Amber Cox, Jeunghoon Lee, Cheryl Jorcyk, Bernard Yurke, William L. Hughes

Materials Science and Engineering Faculty Publications and Presentations

The potential for inferring the presence of cancer by the detection of miRNA in human blood has motivated research into the design and operation of DNA-based chemical amplifiers that can operate in bodily fluids. As a first step toward this goal, we have tested the operation of a DNA-based autocatalytic network in human serum and mouse serum. With the addition of sodium dodecyl sulfate to prevent degradation by nuclease activity, the network was found to operate successfully with both DNA and RNA catalysts.

Specific Thiazolidinediones Inhibit Ovarian Cancer Cell Line Proliferation And Cause Cell Cycle Arrest In A Pparγ Independent Manner, Linah Al-Alem, R. Chase Southard, Michael W. Kilgore, Thomas E. Curry

Pharmacology and Nutritional Sciences Faculty Publications

BACKGROUND: Peroxisome Proliferator Activated Receptor gamma (PPARγ) agonists, such as the thiazolinediones (TZDs), have been studied for their potential use as cancer therapeutic agents. We investigated the effect of four TZDs--Rosiglitazone (Rosi), Ciglitazone (CGZ), Troglitazone (TGZ), and Pioglitazone (Pio)--on ovarian cancer cell proliferation, PPARγ expression and PPAR luciferase reporter activity. We explored whether TZDs act in a PPARγ dependent or independent manner by utilizing molecular approaches to inhibit or overexpress PPARγ activity.

PRINCIPAL FINDINGS: Treatment with CGZ or TGZ for 24 hours decreased proliferation in three ovarian cancer cell lines, Ovcar3, CaOv3, and Skov3, whereas Rosi and Pio had no …

Despite its importance in cell biology and evolution, the centromere has remained the final frontier in genome assembly and annotation due to its complex repeat structure. However, isolation and characterization of the centromeric repeats from newly sequenced species are necessary for a complete understanding of genome evolution and function. In recent years, various genomes have been sequenced, but the characterization of the corresponding centromeric DNA has lagged behind. Here, we present a computational method (RepeatNet) to systematically identify higher-order repeat structures from unassembled whole-genome shotgun sequence and test whether these sequence elements correspond to functional centromeric sequences. We analyzed genome …
