Isolation, Production, And Characterization Of An Extracellular Lipase From Trichoderma Harzianum Isolated From Soil, Serdar Ülker, Arzu Özel, Ahmet Çolak, Şengül Alpay Karaoğlu

This is the first report about the characterization of Trichoderma harzianum lipase. A novel strain of Trichoderma harzianum IDM14D was isolated from soil. The isolated strain was cultivated for lipase production in shake flasks at 30 °C for 7 days. For lipase production, it was determined that the best carbon source was glucose and the best nitrogen source was peptone. Maximum biomass was produced at a concentration of 1.25 g/L, in 7 days. The optimum pH and temperature for activity of the enzyme were 8.5 and 40 °C, respectively. The lipase was stable at a pH range of 8.0-10.0 and …

In recent years, the incidence of fungal infections has been rising all over the world. Although the amount of research in the field of pathogenic fungi has also increased, there is still a need for studies on fungal virulence. In this investigation, we focused on testing bloodstream-identified Candida albicans in experimental in vitro virulence assays. The proteinase enzyme activities of 30 C. albicans strains isolated from the bloodstream and sterile body fluids were investigated (15 colonizations, 15 infections). Polyphenol oxidases (PPOs EC 1.14.18.1) were isolated from Mucuna pruriens and Mucuna prurita and confirmed as tyrosinases involved in L-DOPA production. PPOs were extracted by using a 0.05 M phosphate buffer, pH 7.0. The purified enzyme was resolved into a single band by PAGE, the enzyme was confirmed as PPO by activity staining, and SDS-PAGE analysis revealed that the purified PPO of both the species is a tetramer. Substrate specificity experiments were carried out with catechol, L-DOPA, L-tyrosine, and p-cresol. The chemical composition and nutritional value of a wild edible ectomycorrhizal mushroom from southwestern Anatolia, Tricholoma anatolicum, were analyzed. Moisture, crude oil, protein, ash, total carbohydrate content, and mineral composition of the mushrooms studied, including Fe, Na, K, Zn, Cu, Ca, Cd, and Pb, were determined. The energy values of the samples were also calculated. The analyses were conducted during the 3 different growing stages of the mushrooms: mycelium, young fruiting bodies, and mature fruiting bodies. Bioassay Studies Of 2'-O-Ethylmurrangatin Isolated From A Medicinal Plant, Murraya Paniculata, Azizuddin Shaikh, Muhammad Iqbal Choudhary

A secondary metabolite, 2^'-O-ethylmurrangatin (1), was isolated from the leaves of a medicinal plant, Murraya paniculata. The structure of compound 1 was identified with the help of spectroscopic techniques. Compound 1 was screened for its antioxidant, lipoxygenase, prolyl endopeptidase, and respiratory burst inhibitory activities. 2^'-O-Ethylmurrangatin (1) was found to have significant activity against lipoxygenase enzyme and moderate respiratory burst inhibitory activity, but no activity against either DPPH radical scavenging assay or prolyl endopeptidase enzyme.

Eight different soil tillage systems (TS) for winter wheat after soybean crop production were compared at the chernozem soil type in Croatian Baranya region in a 4-year period (2001/2002, 2002/2003, 2003/2004, 2004/2005). Rains during grain ripening in wheat (Triticum aestivum L.) can cause preharvest sprouting (PHS), which drastically affects the grain yield and the baking quality of flour. In the present study, to screen 7 cultivars and 4 lines of spring bread wheat for PHS, germination tests were conducted with seeds obtained from wet spikes harvested immediately after natural rainfall. The aim of this study was to assess the effects of spermidine (Spd) on ameliorating adverse effects of salinity in Troyer citrange seedlings. For this purpose, 6-month-old, uniform-sized seedlings of Troyer citrange were transplanted to plastic containers containing Hoagland nutrient solution. Addition of 75 mM NaCl into the nutrient solution reduced the plant growth, leaf chlorophyll content, chlorophyll fluorescence yield (Fv/Fm), net photosynthetic rate, respiration rate, and total Spd, N, and Ca^{2+} + Mg^{2+} contents of the leaves. Near-infrared spectroscopy (NIRS) as an alternative technology to conventional methods allows us to obtain results in a few seconds. Most legumes in natural conditions form a symbiosis with arbuscular mycorrhizal (AM) fungi. AM fungi in saline soils have been reported to improve salinity tolerance and growth in plants. In the present study, interaction between mycorrhizal fungus, Glomus mosseae, and salinity stress in relation to plant growth, nitrogen fixation, and nutrient accumulation was evaluated in Cicer arietinum (L.) (chickpea). Two genotypes of chickpea (Pusa-329, salt tolerant, and Pusa-240, salt sensitive) were compared under different levels of salinity with and without mycorrhizal inoculations. Eight primocane fruiting red raspberry cultivars grown at the Experimental station "Radmilovac", a collective raspberry orchard of the Faculty of Agriculture, Belgrade University, were harvested in 2008 and 2009. Using irrigation water with a salinity level of 0.8, 2, and 5 g L^{-1}, the impact of saline water irrigation on the yield and quality of 2 Chinese Cucurbit species, melon (Cucumis melo cv. Huanghe) and watermelon (Citrullus lanatus. convar megulaspemus) were studied in Minqin Oasis, northwestern China. Our results show that melon yields decreased as water salinity increased, just as concentrations of glutamic acid content rose, although the concentration of most amino acids did not differ significantly. The aim of this study was (i) to evaluate the impact of water deficit and rehydration on nitrogen compounds and abscisic acid of Vigna unguiculata leaves, and (ii) to investigate the hypothesis that abscisic acid influences proline. The experimental design was entirely randomized in a factorial design, with 2 water conditions (control and stress) and 7 evaluation periods (0, 1, 2, 3, and 4 days under water deficit, followed by the 5th and 6th day under rehydration). The container type used in nursery culture is among the most important determinants of containerised seedling quality. The objective of the present study was to analyse root and shoot growth, root growth potential, and photochemical efficiency of seedlings of Picea abies, Robinia pseudoacacia, Pinus brutia, and Pinus nigra cultivated for 5 weeks in mini-plugs of 2 different cavity depths (37 and 60 mm). The results showed that precultivation of P. nigra and, to a lesser extent, of R. pseudoacacia in deeper mini-plug containers improved seedling morphological attributes and quality, but shallow containers produced better quality seedlings of P. abies. In order to describe changes in water regime and production parameters caused by red polyethylene (PE) mulch in dehydrated watermelon (Citrullus lanatus (Thunb.) Matsum. & Nakai) plants, as well as to define their transpiration efficiency (TE) and water use efficiency (WUE) under different temperature environments, 2 individual pot experiments (summer external experiment and autumnal greenhouse experiment) were conducted. We found that red mulch reduced leaf relative water content (RWC) fall caused by water restriction but osmotic adjustment (OA) was not responsible for this stress alleviation. Transport today is mostly sustained by liquid fossil fuels, despite the fact that the invention of the combustion engine started with biofuels. However, known environmental impacts and the instability of the fossil-fuel market have revived the status of biofuels. Some concerns have appeared regarding how emerging global climate changes will affect biofuels' feedstock production, since agriculture is strongly dependent on climate variables. The present study was conducted during the growing seasons of 2005, 2006, and 2007 to determine the critical period of weed control (CPWC) in chickpea (cv. Aziziye 94). In order to evaluate the beginning of CPWC, weeds were allowed to compete at weekly intervals for 1 to 8 weeks after emergence (WAE) and, at the end of CPWC, plots were kept weed-free at weekly intervals for 1 to 8 WAE by periodic hand hoeing. Na^+/H^+ antiporters have been suggested to play important roles in salt tolerance in plants. English cordgrass (Spartina anglica) is a gramineous halophyte with very strong salt tolerance. It possesses salt glands in its stems and leaves, through which excess salt can be excreted. In this study, a vacuolar Na^+/H^+ antiporter homologous gene (temporarily named SaNHX1) was isolated from Spartina anglica by RT-PCR and rapid amplification of cDNA ends (RACE).
