Anion Activation Site Of Insulin-Degrading Enzyme, Nicholas Noinaj, Eun Suk Song, Sonia Bhasin, Benjamin J. Alper, Walter K. Schmidt, Louis B. Hersh, David W. Rodgers

Chemistry & Physics Faculty Publications

Insulin-degrading enzyme (IDE) (insulysin) is a zinc metallopeptidase that metabolizes several bioactive peptides, including insulin and the amyloid β peptide. IDE is an unusual metallopeptidase in that it is allosterically activated by both small peptides and anions, such as ATP. Here, we report that the ATP-binding site is located on a portion of the substrate binding chamber wall arising largely from domain 4 of the four-domain IDE. Two variants having residues in this site mutated, IDEK898A,K899A,S901A and IDER429S, both show greatly decreased activation by the polyphosphate anions ATP and PPPi. IDEK898A,K899A,S901A is also deficient …

Lift High The Cross?: Contrasting The New European And American Cases On Religious Symbols On Government Property, John Witte Jr., Nina-Louisa Arold

Emory International Law Review

A comparative anthropologist could not have asked for a better script: two high profile cases, one before the European Court of Human Rights, the other before the U.S. Supreme Court, each involving challenges to traditional displays of crosses on government property. The European high court struck down the cross. The American high court upheld the cross. Both cases are procedurally complicated and are factually distinguishable. Together, these two Articles illustrate some of the complexity of the legal issues surrounding the place of religious symbols on government land, and how serious scholars and judges can take opposing views and marshal reasoned …

Casein Α S1 Is Expressed By Human Monocytes And Upregulates The Production Of Gm-Csf Via P38 Mapk, Stefan Vordenbäumen, Achim Braukmann, Klaudia Petermann, Andrea Scharf, Ellen Bleck, Anna Von Mikecz, Joachim Jose, Matthias Schneider

Biological Sciences Faculty Research & Creative Works

Caseins are major constituents of mammalian milks that are thought to be exclusively expressed in mammary glands and to function primarily as a protein source, as well as to ameliorate intestinal calcium uptake. In addition, proinflammatory and immunomodulatory properties have been reported for bovine caseins. Our aim was to investigate whether human casein α s1 (CSN1S1) is expressed outside the mammary gland and possesses immunomodulatory functions in humans as well. For this purpose, CSN1S1 mRNA was detected in primary human monocytes and CD4+ and CD8+ T cells, but not in CD19 + B cells. CSN1S1 protein was traceable in supernatants …

Assessment Of Pubertal Development Of Boars Derived From Ultrasonographic Determination Of Testicular Diameter, J.J. Ford, T.H. Wise Jan 2011

Assessment Of Pubertal Development Of Boars Derived From Ultrasonographic Determination Of Testicular Diameter, J.J. Ford, T.H. Wise

Roman L. Hruska U.S. Meat Animal Research Center: Reports

At the onset of puberty, seminiferous tubules rapidly increase in diameter, thereby occupying a greater proportion of the testis, resulting in a rapid increase in testicular size. The objective of the current studies was to evaluate ultrasonography for assessing testicular diameter, as a basis for ranking boars relative to their extent of pubertal development. In the initial study, prior to castration at 4, 5, 6, or 7 mo of age, testicular length and diameter were assessed by ultrasonography in 160 anesthetized boars. After castration, testes were weighed. Mean diameter of seminiferous tubules and percentage of the testis occupied by tubules …

Determinants Of Bovine Thermal Response To Heat And Solar Radiation Exposures In A Field Environment, Brad Scharf, Michael J. Leonard, Robert L. Weaber, Terry L. Mader, G. Leroy Hahn, Donald E. Spiers

Roman L. Hruska U.S. Meat Animal Research Center: Reports

Continuous exposure of cattle to summer heat in the absence of shade results in significant hyperthermia and impairs growth and general health. Reliable predictors of heat strain are needed to identify this condition. A 12-day study was conducted during a moderate summer heat period using 12 Angus x Simmental (Bos taurus) steers (533± 12 kg average body weight) to identify animal and ambient determinations of core body temperature (Tcore) and respiration rate (RR) responses to heat stress. Steers were provided standard diet and water ad libitum, and implanted intraperitoneally with telemetric transmitters to monitor Tcore …

Effect Of Feeding Wet Distillers Grains With Solubles To Beef Cattle On Air And Manure Quality, M. J. Spiehs, D. N. Miller, B. L. Woodbury, R. A. Eigenberg, V. H. Varel, D. B. Parker Jan 2011

Effect Of Feeding Wet Distillers Grains With Solubles To Beef Cattle On Air And Manure Quality, M. J. Spiehs, D. N. Miller, B. L. Woodbury, R. A. Eigenberg, V. H. Varel, D. B. Parker

Roman L. Hruska U.S. Meat Animal Research Center: Reports

Air quality is becoming a pressing issue for beef feedlot producers. Feeding practices influence the excretion of starch, fiber, nitrogen (N), and sulfur (S) in manure, thereby affecting nutrient content and the production of ammonia (NH3) and odorous compounds. Wet distillers grains with solubles (WDGS) are a common and economical ingredient in feedlot diets. WDGS are high in protein, fiber, phosphorus (P), and S but low in starch. The objective of this study was to compare NH3 concentration in the air and nutrients and volatile organic compounds (VOC) concentration in manure between two dietary treatments fed to feedlot …

A Solid Phase Assay For Topoisomerase I Interfacial Poisons And Catalytic Inhibitors, Vidusha Cyril Jan 2011

A Solid Phase Assay For Topoisomerase I Interfacial Poisons And Catalytic Inhibitors, Vidusha Cyril

Electronic Theses and Dissertations

We report a mechanism based screening technique to rapidly identify eukaryotic topoisomerase I targeting agents. The method is based on genetic tagging of topoisomerase I to immobilize the enzyme on a solid surface in a microtiter well format. DNA is added to the wells and retained DNA is detected by Picogreen fluorescence. Compounds that result in an increase in Picogreen staining represent potential topoisomerase interfacial poisons while those that reduce fluorescence report catalytic inhibitors; therefore, the solid phase assay represents a „bimodal‟ readout that reveals mechanisms of action. The method has been demonstrated to work with known interfacial poisons and …

MacDonald on BBC Television

George MacDonald received welcome publicity on BBC Television’s popular Sunday afternoon religious programme Songs of Praise on 16th January this year when I was interviewed about his life and work in a programme broadcast from Arundel in West Sussex. Songs of Praise, which celebrated its 50th anniversary this year, is centred around hymns chosen by local people and sung in a cathedral or large church, but also includes interviews with local people and others about matters of interest.

Co-Culture Of Yeast Antagonists Of Fusarium Head Blight And Their Effect On Disease Development In Wheat, D. A. Schisler, P. J. Slininger, Michael J. Boehm, P. A. Paul Jan 2011

Co-Culture Of Yeast Antagonists Of Fusarium Head Blight And Their Effect On Disease Development In Wheat, D. A. Schisler, P. J. Slininger, Michael J. Boehm, P. A. Paul

Department of Plant Pathology: Faculty Publications

Multistrain mixtures of biocontrol agents which can reduce plant disease to a greater extent than the individual strains of the mixture, commonly, are prepared by blending separately produced fermentation products. Co-cultivation of strains to equivalent biomass yields would provide mixture advantages without incurring the cost disadvantages of multiple fermentation and processing protocols. Fusariwn Head Blight (FHB) antagonists Cryptococcus flavescens OR 182.9 (NRRL Y-302l6), C. aureus OH 181.1 (NRRL Y-302l5) and C. aureus OR 181.1 (NRRL Y -30215), were grown in two- and three-strain co-cultures to assess the quality and efficacy of the fermentation end products produced. Final cell counts of …

The Gc-Rich Mitochondrial And Plastid Genomes Of The Green Alga Coccomyxa Give Insight Into The Evolution Of Organelle Dna Nucleotide Landscape, David Roy Smith, Fabien Burki, Takashi Burki, Jane Grimwood, Igor V, Grigoriev, James L. Van Etten, Patrick J. Keeling

Department of Plant Pathology: Faculty Publications

Most of the available mitochondrial and plastid genome sequences are biased towards adenine and thymine (AT) over guanine and cytosine (GC). Examples of GC-rich organelle DNAs are limited to a small but eclectic list of species, including certain green algae. Here, to gain insight in the evolution of organelle nucleotide landscape, we present the GC-rich mitochondrial and plastid DNAs from the trebouxiophyte green alga Coccomyxa sp. C-169. We compare these sequences with other GC-rich organelle DNAs and argue that the forces biasing them towards G and C are nonadaptive and linked to the metabolic and/or life history features of this …
