A Statin-Regulated Microrna Represses C-Myc Expression And Function., Apana Agha L. Takwi 1980-
Electronic Theses and Dissertations
c-Myc dysregulation is one of the most common abnormalities found in human cancer. MicroRNAs (miRNAs) are functionally intertwined with the c-Myc network as multiple miRNAs are regulated by c-Myc, while others directly suppress c-Myc expression. In this work, we identified miR-33b as a primate-specific negative regulator of c-Myc. The human miR-33b gene is located at 17pl1.2, a genomic locus frequently lost in medulloblastomas, which are pediatric brain tumors that display c-Myc overproduction. Through a small-scale screening with drugs approved by the US Food and Drug Administration (FDA), we found that lovastatin up-regulated miR-33b expression, reduced cell proliferation, and impaired c-Myc …

Cost Of A Tax Agenda: The Passage Of Proposition A And Its Effect On Kansas City And St. Louis City, The, Missy Mccoy
Missouri Law Review
This Law Summary begins with a discussion of the history of the earnings tax in Kansas City and St. Louis City starting with both cities' designations as home rule cities.10 Being home rule cities allowed them independence in governing their populace and the eventual implementation of an earnings tax through city charters and state enabling statutes. Next, this Law Summary discusses Proposition A's repeal of the previous enabling statutes, the institution of new statutes, and the prohibition of any other city from passing an earnings tax. This Law Summary takes the position that Proposition A, funded largely by St. Louis-area …

Picking Fights In Missouri: Baldwin's Non-Rule Embraces The Minority Approach To Internet Libel Jurisdiction, Allison Marie Isaak
Missouri Law Review
The competing standards of Internet libel jurisdiction reflect the tensions between the forum state's interest in providing convenient recovery for its injured residents and the defendant's constitutional right to foresee where he might be subject to jurisdiction. In an effort to pursue these two goals as well as integrate modem Internet-related concerns, lower courts have derived numerous divergent tests for Internet libel jurisdiction, leaving the issue in a state of disorder and ambiguity. To analyze this problem, this Note will first survey the historical background of traditional personal jurisdiction principles, with particular emphasis on the U.S. Supreme Court's Calder "effects" …

Western Pacific amyotrophic lateral sclerosis (ALS) and parkinsonism-dementia complex (PDC), a prototypical neurodegenerative disease (tauopathy) affecting distinct genetic groups with common exposure to neurotoxic chemicals in cycad seed, has many features of Parkinson's and Alzheimer's diseases (AD), including early olfactory dysfunction. Guam ALS-PDC incidence correlates with cycad flour content of cycasin and its aglycone methylazoxymethanol (MAM), which produces persistent DNA damage (O6-methylguanine) in the brains of mice lacking O6-methylguanine methyltransferase (Mgmt-/-). We described in Mgmt-/-mice up to 7 days post-MAM treatment that brain DNA damage was linked to brain gene …

Molecular Characterization Of The Host Defense Activity Of The Barrier To Autointegration Factor Against Vaccinia Virus, Nouhou Ibrahim, April Wicklund, Matthew S. Wiebe
Nebraska Center for Virology: Faculty Publications
The barrier to autointegration factor (BAF) is an essential cellular protein with functions in mitotic nuclear reassembly, retroviral preintegration complex stability, and transcriptional regulation. Molecular properties of BAF include the ability to bind double-stranded DNA in a sequence-independent manner, homodimerize, and bind proteins containing a LEM domain. These capabilities allow BAF to compact DNA and assemble higher-order nucleoprotein complexes, the nature of which is poorly understood. Recently, it was revealed that BAF also acts as a potent host defense against poxviral DNA replication in the cytoplasm. Here, we extend these observations by examining the molecular mechanism through which BAF acts …

Species D Adenoviruses As Oncolytics Against B-Cell Cancers, Christopher Y. Chen, Julien S. Senac, Eric A. Weaver, Shannon M. May, Diane F. Jelinek, Philip Greipp, Thomas Witzig, Michael A. Barry
Nebraska Center for Virology: Faculty Publications
Purpose: Oncolytic viruses are self-amplifying anticancer agents that make use of the natural ability of viruses to kill cells. Adenovirus serotype 5 (Ad5) has been extensively tested against solid cancers, but less so against B-cell cancers because these cells do not generally express the coxsackie and adenoviral receptor (CAR). To determine whether other adenoviruses might have better potency, we "mined" the adenovirus virome of 55 serotypes for viruses that could kill B-cell cancers.
Experimental Design: Fifteen adenoviruses selected to represent Ad species B, C, D, E, and F were tested in vitro against cell lines and primary patient B-cell cancers …

Family Affairs Newsletter 2011-11-01, Zack Paakkonen
Family Affairs newsletter (2004-2016)
FAMILY AFFAIRS was a free, twice-a-month, social activities newsletter for the GLBTQI (gay/lesbian/bisexual/trans/queer/intersex) community, sent out around the 1st and 15th of each month. It covered the State of Maine only. The list was begun and maintained for many years by Jean Vermette in Bangor, and later operated by Zack Paakkonen of Portland. Over the years it evolved from a social activities newsletter into a business directory, classified ad service, and community bulletin board.

Within the classroom, literacy learning plays a central role in what children are asked to adopt to be full functioning members of the culture. Children are asked to negotiate the signs of texts, as well as those of the classroom and larger society. The process of learning to read and write, needless to say, is a complex one. Research in reading has shown that to teach children how to participate in this culture successfully, teachers must build upon what children do well in a meaningful context (Calkins, 1980; Wray, 1997) as opposed to the teaching of skills and items in …

Microsurgical repair of transected peripheral nerves is compromised by the formation of scar tissue and the development of a neuroma, thereby limiting the success of regeneration. The aim of this study was to quantify histomorphometrically the structural changes in neural tissue that result from repair, and determine the effect of mannose-6-phosphate (M6P), a scar-reducing agent previously shown to enhance regeneration. In anaesthetised C57-black-6 mice, the left sciatic nerve was sectioned and repaired using four epineurial sutures. Either 100 mu L of 600 mM M6P (five animals) or 100 mu L of phosphate-buffered saline (placebo controls, five animals) was injected into …

Boron is an essential micronutrient for plants, and it is beneficial for animals. However, at high concentrations boron is toxic to cells although the mechanism of this toxicity is not known. Atr1 has recently been identified as a boron efflux pump whose expression is upregulated in response to boron treatment. Here, we found that the expression of ATR1 is associated with expression of genes involved in amino acid biosynthesis. These mechanisms are strictly controlled by the transcription factor Gcn4 in response to boron treatment. Further analyses have shown that boron impaired protein synthesis by promoting phosphorylation of eIF2α in a …

We identified a previously unrecognized component of airborne particulate matter (PM) formed in combustion and thermal processes, namely, environmentally persistent free radicals (EPFRs). The pulmonary health effects of EPFRs are currently unknown. In the present study, we used a model EPFR-containing pollutant-particle system referred to as MCP230. We evaluated the effects of MCP230 on the phenotype and function of bone marrow - derived dendritic cells (BMDCs) in vitro and lung dendritic cells (DCs) in vivo, and the subsequent T-cell response. We also investigated the adjuvant role of MCP230 on airway inflammation in a mouse model of asthma. MCP230 decreased intracellular …

Unhinged, Gayane Bagdasaryan
Theses
"Unhinged" is an animated graduate thesis film 7 minutes, 30 seconds long, which was originally conceived as a story about decision making, and it turned into a story about loneliness and an acceptance of it. It tells a story of a troll, Lancelot, who carries on a lonely life in a neglected cabin in the middle of a desert. He entertains himself by playing masquerade with his friends, mannequins-hat-holders. One day he hears a knock on the door and struggles with fear to open the door. This film is a hand-drawn animation that was mostly produced in TVPaint Animation Software. …

Clinical evidence that the blockade of IL-1β in type-2 diabetic patients improves glycemia is indicative of an autoinflammatory mechanism that may trigger adiposity-driven pancreatic damage. IL-1β is a key contributor to the obesity-induced inflammation and subsequent insulin resistance, pancreatic β-cell dysfunction, and the onset of type 2 diabetes. Our previous studies demonstrated that the ceramides activate the Nod-like receptor family, pyrin domain containing 3 (Nlrp3) inflammasome to cause the generation of mature IL-1β and ablation of the Nlrp3 inflammasome in diet-induced obesity improves insulin signaling. However, it remains unclear whether the post-translational processing of active IL-1β in pancreas is regulated …

Rodents are natural reservoirs for a variety of species of Borrelia that cause relapsing fevers in humans. The murine model of this disease recapitulates many of the clinical manifestations of the human disease and has revealed that T cell-independent antibody responses are required to resolve the bacteremic episodes. However, it is not clear whether such protective humoral responses are mounted in humans.

Cultural Metaphors: Their Use In Management Practice As A Method For Understanding Cultures, Martin J. Gannon
Online Readings in Psychology and Culture
This paper argues that the dimensional perspective on culture, arguably the most important development in cross-cultural understanding in 50 years, needs to be supplemented by in-depth treatments of national cultures. Such a treatment is the goal of a cultural metaphor, which is any activity, phenomenon, or institution with which members of a given culture emotionally and/or cognitively identify. As such, cultural metaphors reflect the underlying values of a culture. Examples of national cultural metaphors include the Japanese garden, the Chinese family altar, and American Football. The paper also argues that the focus on specific national cultures can lead to the …
