Institute for Immunology and Informatics Faculty Publications

The potential for smallpox to be disseminated in a bioterror attack has prompted development of new, safer smallpox vaccination strategies. We designed and evaluated immunogenicity and efficacy of a T-cell epitope vaccine based on conserved and antigenic vaccinia/variola sequences, identified using bioinformatics and immunological methods. Vaccination in HLA transgenic mice using a DNA-prime/peptide-boost strategy elicited significant T cell responses to multiple epitopes. No antibody response pre-challenge was observed, neither against whole vaccinia antigens nor vaccine epitope peptides. Remarkably, 100% of vaccinated mice survived lethal vaccinia challenge, demonstrating that protective immunity to vaccinia does not require B cell priming.

Enhanced Bioavailability Of Fenoterol Transdermal Systems In Rabbits, A. H. Elshafeey, Y. E. Hamza, S. Y. Amin, Fatemeh Akhlaghi, H. Zia

Biomedical and Pharmaceutical Sciences Faculty Publications

The pharmacokinetic and bioavailability of fenoterol, a B2 adrenergic agonist were studied to determine the feasibility of enhanced transdermal delivery. Fenoterol has been widely used to treat asthmatic patients. Two fenoterol formulations were studied; the first was a liquid formulation of fenoterol in Transcutol: Oleic acid in a ratio 1:1(F1), while the second was a matrix system of fenoterol in Duro-tak® 87-2074 adhesive with 10% 1-dodecyl-2-pyrrolidinone as an enhancer (F2). For comparison, control matrix with fenoterol without any enhancer (F3) was also tested. The tested formulations were applied to the shaved back skin of rabbits using HILL TOP CHAMBER® …

Our aim was to identify genes that influence the inverse association of coffee with the risk of developing Parkinson's disease (PD). We used genome-wide genotype data and lifetime caffeinated-coffee-consumption data on 1,458 persons with PD and 931 without PD from the NeuroGenetics Research Consortium (NGRC), and we performed a genome-wide association and interaction study (GWAIS), testing each SNP's main-effect plus its interaction with coffee, adjusting for sex, age, and two principal components. We then stratified subjects as heavy or light coffee-drinkers and performed genome-wide association study (GWAS) in each group. We replicated the most significant SNP. Finally, we imputed the …

Spag16, An Axonemal Central Apparatus Gene, Encodes A Male Germ Cell Nuclear Speckle Protein That Regulates Spag16 Mrna Expression, David R. Nagarkatti-Gude, Ruth Jaimez, Scott C. Henderson, Maria E. Teves, Zhibing Zhang, Jerome F. Strauss Iii

Biochemistry and Molecular Biology Publications

Spag16 is the murine orthologue of Chlamydomonas reinhardtii PF20, a protein known to be essential to the structure and function of the “9+2” axoneme. In Chlamydomonas, the PF20gene encodes a single protein present in the central pair of the axoneme. Loss of PF20 prevents central pair assembly/integrity and results in flagellar paralysis. Here we demonstrate that the murine Spag16 gene encodes two proteins: 71 kDa SPAG16L, which is found in all murine cells with motile cilia or flagella, and 35 kDa SPAG16S, representing the C terminus of SPAG16L, which is expressed only in male germ cells, and is …

Development Of Melanopsin-Based Irradiance Detecting Circuitry, David S. Mcneill, Catherine J. Sheely, Jennifer L. Ecker, Tudor C. Badea, Duncan Morhardt, William Guido, Samer Hattar

Anatomy and Neurobiology Publications

Background

Most retinal ganglion cells (RGCs) convey contrast and motion information to visual brain centers. Approximately 2% of RGCs are intrinsically photosensitive (ipRGCs), express melanopsin and are necessary for light to modulate specific physiological processes in mice. The ipRGCs directly target the suprachiasmatic nucleus (SCN) to photoentrain circadian rhythms, and the olivary pretectal nucleus (OPN) to mediate the pupillary light response. How and when this ipRGC circuitry develops is unknown.

Results

Here, we show that some ipRGCs follow a delayed developmental time course relative to other image-forming RGCs. Specifically, ipRGC neurogenesis extends beyond that of other RGCs, and ipRGCs begin …

Gore's Science The Kairos Of An Inconvenient Truth And The Implications For Science Writing, Carolyn M. Glasshoff Jan 2011

Gore's Science The Kairos Of An Inconvenient Truth And The Implications For Science Writing, Carolyn M. Glasshoff

Electronic Theses and Dissertations

Modern Americans are exposed to scientific and technical information on a daily basis that urges them to react as well as learn about new ideas. The popular science writing that circulates this information must be portrayed in a way that makes it easy for lay people to understand complicated ideas while at the same time remaining complex enough to convince readers that the information is reliable, accurate, and worth learning. In making decisions about how to accomplish this balancing act, science writers make decisions that influence the audience's opinion about new scientific ideas, how easily the audience will accept or …

Five Secondary Teachers: Creating And Presenting A Teaching Persona, Janine S. Davis Jan 2011

Five Secondary Teachers: Creating And Presenting A Teaching Persona, Janine S. Davis

Education Faculty Articles

This qualitative study investigates the ways that five secondary teachers developed and presented personae. The researcher collected and analyzed data using a theoretical frame based in social psychology, including Goffman’s Presentation of Self in Everyday Life (1959), and Miles and Huberman’s (1994) three-step approach to qualitative data analysis. Findings indicate that teachers drew on three major realms—the physical, psychological, and social—when constructing classroom personae. Implications include increased opportunities for teacher reflection on persona and its effects.

Designing And Building A Knowledge Warehouse Oriented, Public Decision Support Site Using Free Google Tools: An Analysis And Case Application To Deer Management, G. Kent Webb

Faculty Publications, Information Systems & Technology

As a case example for the design and implementation of a public decision support site, a variety of free Google tools are applied to supporting deer management decisions. A large number of communities throughout the country are engaged in what is often described as one of the most contentious decisions facing local governments. These decision makers routinely look to the internet for decision support, but information can be contradictory and difficult to find. To determine issues and requirements, data was collected using Google Search, Google keyword data, and a Google Alert was set up to retrieve news and information on …

The Associates In Teaching Program: Graduate Student Development, Faculty Renewal, And Curricular Innovation, Bill Rando

Professional and Organizational Development Network in Higher Education: Archives

Conflicting goals define university life. Luckily, conflicting goals sometimes inspire creative innovation. The Associates in Teaching Program is one such example.

At research universities, administrators must balance the needs of graduate students and those of undergraduate students – graduate students need practice teaching to prepare them for faculty life; undergraduates (and their parents) would prefer instruction by more experienced faculty members. Nearly all universities meet the needs of graduate students through teaching assistant positions in labs and sections. Some go further by allowing graduate students to teach independent courses, which benefits most graduate students, but frustrates others who feel that …

Anaerobic Respiration Of Escherichia Coli In The Mouse Intestine, Shari A. Jones, Terri Gibson, Rosalie C. Maltby, Fatema Z. Chowdhury, Valley Stewart, Paul S. Cohen, Tyrrell Conway Jan 2011

Anaerobic Respiration Of Escherichia Coli In The Mouse Intestine, Shari A. Jones, Terri Gibson, Rosalie C. Maltby, Fatema Z. Chowdhury, Valley Stewart, Paul S. Cohen, Tyrrell Conway

Cell and Molecular Biology Faculty Publications

The intestine is inhabited by a large microbial community consisting primarily of anaerobes and, to a lesser extent, facultative anaerobes, such as Escherichia coli, which we have shown requires aerobic respiration to compete successfully in the mouse intestine (S. A. Jones et al., Infect. Immun. 75:4891-4899, 2007). If facultative anaerobes efficiently lower oxygen availability in the intestine, then their sustained growth must also depend on anaerobic metabolism. In support of this idea, mutants lacking nitrate reductase or fumarate reductase have extreme colonization defects. Here, we further explore the role of anaerobic respiration in colonization using the streptomycin-treated mouse model. We …

Genotype And Phenotypes Of An Intestine-Adapted Escherichia Coli K-12 Mutant Selected By Animal Passage For Superior Colonization, Andrew J. Fabich, Mary P. Leatham, Joe E. Grissom, Graham Wiley, Hongshing Lai, Fares Najar, Bruce A. Roe, Paul S. Cohen, Tyrrell Conway

Cell and Molecular Biology Faculty Publications

We previously isolated a spontaneous mutant of Escherichia coli K-12, strain MG1655, following passage through the streptomycin-treated mouse intestine, that has colonization traits superior to the wild-type parent strain (M. P. Leatham et al., Infect. Immun. 73:8039–8049, 2005). This intestine-adapted strain (E. coli MG1655*) grew faster on several different carbon sources than the wild type and was nonmotile due to deletion of the flhD gene. We now report the results of several high-throughput genomic analysis approaches to further characterize E. coli MG1655*. Whole-genome pyrosequencing did not reveal any changes on its genome, aside from the deletion at the flhDC …

On The Connection Between Law And Justice, Anthony D'Amato Jan 2011

On The Connection Between Law And Justice, Anthony D'Amato

Faculty Working Papers

What does it mean to assert that judges should decide cases according to justice and not according to the law? Is there something incoherent in the question itself? That question will serve as our springboard in examining what is—or should be—the connection between justice and law. Legal and political theorists since the time of Plato have wrestled with the problem of whether justice is part of law or is simply a moral judgment about law. Nearly every writer on the subject has either concluded that justice is only a judgment about law or has offered no reason to support a …

Phony Originalism And The Establishment Clause, Andrew M. Koppelman Jan 2011

Phony Originalism And The Establishment Clause, Andrew M. Koppelman

Faculty Working Papers

The "originalist" interpretations of the Establishment Clause by Supreme Court Justices William Rehnquist, Antonin Scalia, and Clarence Thomas are remarkably indifferent to the original purposes of that clause. Their arguments are a remarkable congeries of historical error and outright misrepresentation. This is not necessarily a criticism of originalism per se. However, the abuse of originalist scholarship that these judges have practiced raises questions about what originalist scholars are actually accomplishing.

A Moral Contractual Approach To Labor Law Reform: A Template For Using Ethical Principles To Regulate Behavior Where Law Failed To Do So Effectively, Zev J. Eigen, David S. Sherwyn Jan 2011

A Moral Contractual Approach To Labor Law Reform: A Template For Using Ethical Principles To Regulate Behavior Where Law Failed To Do So Effectively, Zev J. Eigen, David S. Sherwyn

Faculty Working Papers

If laws cease to work as they should or as intended, legislators and scholars propose new laws to replace or amend them. This paper posits an alternative—offering regulated parties the opportunity to contractually bind themselves to behave ethically. The perfect test-case for this proposal is labor law, because (1) labor law has not been amended for decades, (2) proposals to amend it have failed for political reasons, and are focused on union election win rates, and less on the election process itself, (3) it is an area of law already statutorily regulating parties' reciprocal contractual obligations, and (4) moral means …

Why Jack Balkin Is Disgusting, Andrew Koppelman Jan 2011

Why Jack Balkin Is Disgusting, Andrew Koppelman

Faculty Working Papers

Yale Law Professor Jack Balkin didn't win friends when he announced that (1) he is now a constitutional originalist and (2) the original meaning of the Fourteenth Amendment protects the right to abortion. His claim to membership in the originalist club brought forth a small army of eager bouncers, who were sure that originalism couldn't possibly defend the paradigmatic departure from the Constitution's original meaning.

Balkin has indeed posed a radical challenge to the vision of law that drives the originalists – more radical than he is willing to admit. His theory is in such deep tension with a commonly …

The Limits Of Constructivism: Can Rawls Condemn Female Genital Mutilation?, Andrew Koppelman Jan 2011

The Limits Of Constructivism: Can Rawls Condemn Female Genital Mutilation?, Andrew Koppelman

Faculty Working Papers

The strategy for coping with value pluralism that Rawls has proposed is to permit political decisions, at least with respect to basic rights, to depend only on those goods that can be inferred from the bare requirements of respectful relations between persons. His account offers such a parsimonious conception of the good that it cannot cognize some atrocities. I focus on one extreme human rights case: the practice of female genital mutilation (FGM), which, it is well established, violates basic human rights. Doubtless Rawls was appalled by the practice. Yet his theory cannot generate a basis for condemning it. A …

The New American Civil Religion: Lesson For Italy, Andrew Koppelman Jan 2011

The New American Civil Religion: Lesson For Italy, Andrew Koppelman

Faculty Working Papers

American civil religion has been changing, responding to increasing religious plurality by becoming more abstract. The problem of increasing plurality is not only an American one. It is also presented in Italy, where civic identity has been centered around a Catholicism that is no longer universal. Perhaps Italy has, in this respect, an American future.

Safety First? The Role Of Emotion In Safety Product Betrayal Aversion, Jonathan Koehler, Andrew D. Gershoff Jan 2011

Safety First? The Role Of Emotion In Safety Product Betrayal Aversion, Jonathan Koehler, Andrew D. Gershoff

Faculty Working Papers

Consumers often face decisions about whether to purchase products that are intended to protect them from possible harm. However, safety products rarely provide perfect protection and sometimes "betray" consumers by causing the very harm they are intended to prevent. Examples include vaccines that may cause disease and air bags that may explode with such force that they cause death. Expanding research on betrayal aversion, this study examines the role of emotions in consumers' tendency to choose safety options that provide less overall protection in order to eliminate a very small probability of harm due to safety product betrayal. In five …
