BACKGROUND: Filarial nematodes, including Brugia malayi, the causative agent of lymphatic filariasis, undergo molting in both arthropod and mammalian hosts to complete their life cycles. An understanding of how these parasites cross developmental checkpoints may reveal potential targets for intervention. Pharmacological evidence suggests that ecdysteroids play a role in parasitic nematode molting and fertility although their specific function remains unknown. In insects, ecdysone triggers molting through the activation of the ecdysone receptor: a heterodimer of EcR (ecdysone receptor) and USP (Ultraspiracle).

METHODS AND FINDINGS: We report the cloning and characterization of a B. malayi EcR homologue (Bma-EcR). Bma-EcR dimerizes with …

Investigating An Intervention Program Linking Writing And Vocabulary Development For Homeless Children, Richard Sinatra, Robert Eschenauer

Journal of Youth Development

The presented study investigated the effects of a four-week academic and activity – enriched summer program on vocabulary development and writing achievement of homeless children residing in traditional shelter facilities. When compared to controls, the experimental students did not reveal gains in vocabulary and spelling as measured by two norm referenced tests. They did however demonstrate highly significant gains in writing ability based on the New York State standards criteria, reflecting five qualities of writing. On two project-developed instruments designed to measure improvement in book vocabulary and tennis skills, they showed significant increases based on analyses of their pre- and …

Historic Fire Regimes On Eastern Great Basin (Usa) Mountains Reconstructed From Tree Rings, Stanley G. Kitchen Mar 2010

Historic Fire Regimes On Eastern Great Basin (Usa) Mountains Reconstructed From Tree Rings, Stanley G. Kitchen

Theses and Dissertations

Management of natural landscapes requires knowledge of key disturbance processes and their effects. Fire and forest histories provide valuable insight into how fire and vegetation varied and interacted in the past. I constructed multi-century fire chronologies for 10 sites on six mountain ranges representative of the eastern Great Basin (USA), a region in which historic fire information was lacking. I also constructed tree recruitment chronologies for two sites. I use these chronologies to address three research foci. First, using fire-scar data from four heterogeneous sites, I assert that mean fire interval (MFI) values calculated from composite chronologies provide suitable estimates …

A Phylogeny And Timescale For The Evolution Of Pseudocheiridae (Marsupialia Diprotodontia) In Australia And New Guinea, Robert Meredith, Miguel A. Mendoza, Karen K. Roberts, Michael Westerman, Mark S. Springer

Department of Biology Faculty Scholarship and Creative Works

Pseudocheiridae (Marsupialia: Diprotodontia) is a family of endemic Australasian arboreal folivores, more commonly known as ringtail possums. Seventeen extant species are grouped into six genera (Pseudocheirus, Pseudochirulus, Hemibelideus, Petauroides, Pseudochirops, Petropseudes). Pseudochirops and Pseudochirulus are the only genera with representatives on New Guinea and surrounding western islands. Here, we examine phylogenetic relationships among 13 of the 17 extant pseudocheirid species based on protein-coding portions of the ApoB, BRCA1, ENAM, IRBP, Rag1, and vWF genes. Maximum parsimony, maximum likelihood, and Bayesian methods were used to estimate phylogenetic relationships. Two different relaxed molecular clock methods were used to estimate divergence times. Bayesian …

Distinct Evolutionary Histories Of The Dna-A And Dna-B Components Of Bipartite Begomoviruses, Rob W. Briddon, Basavaprabhu L. Patil, Basavaraj Bagewadi, Muhammad Shah Nawaz-Ul-Rehman, Claude M. Fauquet Mar 2010

Distinct Evolutionary Histories Of The Dna-A And Dna-B Components Of Bipartite Begomoviruses, Rob W. Briddon, Basavaprabhu L. Patil, Basavaraj Bagewadi, Muhammad Shah Nawaz-Ul-Rehman, Claude M. Fauquet

Plant Pathology Faculty Publications

BACKGROUND: Viruses of the genus Begomovirus (family Geminiviridae) have genomes consisting of either one or two genomic components. The component of bipartite begomoviruses known as DNA-A is homologous to the genomes of all geminiviruses and encodes proteins required for replication, control of gene expression, overcoming host defenses, encapsidation and insect transmission. The second component, referred to as DNA-B, encodes two proteins with functions in intra- and intercellular movement in host plants. The origin of the DNA-B component remains unclear. The study described here was initiated to investigate the relationship between the DNA-A and DNA-B components of bipartite begomoviruses with a …

MicroRNAs (miRNAs) regulate gene expression. It has been suggested that obtaining miRNA expression profiles can improve classification, diagnostic, and prognostic information in oncology. Here, we sought to comprehensively identify the miRNAs that are overexpressed in lung cancer by conducting miRNA microarray expression profiling on normal lung versus adjacent lung cancers from transgenic mice. We found that miR-136, miR-376a, and miR-31 were each prominently overexpressed in murine lung cancers. Real-time RT-PCR and in situ hybridization (ISH) assays confirmed these miRNA expression profiles in paired normal-malignant lung tissues from mice and humans. Engineered knockdown of miR-31, but not other highlighted miRNAs, substantially …

Dartmouth Scholarship

Arterial morphogenesis is an important and poorly understood process. In particular, the signaling events controlling arterial formation have not been established. We evaluated whether alterations in the balance between ERK1/2 and PI3K signaling pathways could stimulate arterial formation in the setting of defective arterial morphogenesis in mice and zebrafish. Increased ERK1/2 activity in mouse ECs with reduced VEGF responsiveness was achieved in vitro and in vivo by downregulating PI3K activity, suppressing Akt1 but not Akt2 expression, or introducing a constitutively active ERK1/2 construct. Such restoration of ERK1/2 activation was sufficient to restore impaired arterial development and branching morphogenesis in synectin-deficient …

Magnesium Excretion In C. Elegans Requires The Activity Of The Gtl-2 Trpm Channel, Takayuki Teramoto, Laura A. Sternick, Eriko Kage-Nakadai, Shirine Sajjadi, Jakub Siembida, Shohei Mitani, Kouichi Iwasaki, Eric J. Lambie

Dartmouth Scholarship

Systemic magnesium homeostasis in mammals is primarily governed by the activities of the TRPM6 and TRPM7 cation channels, which mediate both uptake by the intestinal epithelial cells and reabsorption by the distal convoluted tubule cells in the kidney. In the nematode, C. elegans, intestinal magnesium uptake is dependent on the activities of the TRPM channel proteins, GON-2 and GTL-1. In this paper we provide evidence that another member of the TRPM protein family, GTL-2, acts within the C. elegans excretory cell to mediate the excretion of excess magnesium. Thus, the activity of GTL-2 balances the activities of the paralogous …

Bayesian And Frequentist Approaches For The Analysis Of Multiple Endpoints Data Resulting From Exposure To Multiple Health Stressors., Epiphanie Nyirabahizi

Theses and Dissertations

In risk analysis, Benchmark dose (BMD)methodology is used to quantify the risk associated with exposure to stressors such as environmental chemicals. It consists of fitting a mathematical model to the exposure data and the BMD is the dose expected to result in a pre-specified response or benchmark response (BMR). Most available exposure data are from single chemical exposure, but living objects are exposed to multiple sources of hazards. Furthermore, in some studies, researchers may observe multiple endpoints on one subject. Statistical approaches to address multiple endpoints problem can be partitioned into a dimension reduction group and a dimension preservative group. …
