Rodent models play an essential role in the development of new chemotherapeutics and dosing regimes. It is often difficult to carryout a clinical study for pediatric cancers due to the small patient population. Retinoblastoma, a pediatric cancer of the eye, is one example of a pediatric cancer that can benefit from preclinical studies. Over the years various retinoblastoma rodent models have been developed used to test various combination of broad-spectrum systemic chemotherapy. Bone grafts are commonly used for the treatment of large bone defects. Biodegradable, synthetic bone grafts combined with cells and growth factors as a tissue engineered construct can provide benefits over the common autografts and allografts currently used. Calcium phosphate (CaP) materials offer high biocompatibility and chemical similarity to natural bone tissue. Biodegradable beta-tricalcium phosphate (β-TCP) scaffolds with an interconnected porous network have been created to promote tissue ingrowth and regeneration in vitro and in vivo. Quinic acid (QA) esters found in hot water extracts of Uncaria tomentosa (a.k.a. Cat’s claw) exert anti-inflammatory activity through mechanisms involving inhibition of the pro-inflammatory transcription factor nuclear factor kappa B (NF-κB). Herein, we described the synthesis and biological testing of novel QA derivatives. Inhibition of NF-κB was assessed using A549 (Type II alveolar epithelial-like) cells that stably express a secreted alkaline phosphatase (SEAP) reporter driven by an NF-κB response element. A549- NF-κB cells were stimulated with TNF-α (10 ng/mL) in the presence or absence of QA derivative for 18 hours followed by measurement of SEAP activity. Amide substitution at …

The freshwater planarian has a great capacity to regenerate and is an ideal animal model in the study of stem cell and regeneration biology. In this study planarian regenerating new tails were exposed to nonlethal doses of Betadine®, Polyvinylpyrrolidone (PVP), and Iodine. Betadine® is a topical antiseptic commonly used in the healthcare setting and may have a detrimental effect on wound healing. PVP is linked to iodine to create povidone-iodine, the active ingredient in Betadine®. Thermochemical gasification is one of the most promising technologies for converting biomass into power, fuels and chemicals. The objectives of this study were to maximize the net energy efficiency for biomass gasification, and to estimate the cost of producing industrial gas and combined heat and power (CHP) at a feedrate of 2,000 kg/h. Aspen Plusbased model for gasification was combined with a CHP generation model, and optimized using corn stover and dried distillers grains with solubles (DDGS) as the biomass feedstocks. Genetic analysis has implicated the cell surface glycoprotein gp130 in cell interactions of the social amoeba Dictyostelium, and information about the utilization of the 18 N-glycosylation sequons present in gp130 is needed to identify critical molecular determinants of its activity. Various glycomics strategies, including mass spectrometry of native and derivatized glycans, monosaccharide analysis, exoglycosidase digestion, and antibody binding, were applied to characterize a nonanchored version secreted from Dictyostelium. s-gp130 is modified by a predominant Man8GlcNAc4 species containing bisecting and intersecting GlcNAc residues and additional high-mannose N-glycans substituted with sulfate, methyl-phosphate, and/or core R3-fucose. The α- and β-globin gene families of jawed vertebrates have diversified with respect to both gene function and the developmental timing of gene expression. Phylogenetic reconstructions of globin gene family evolution have provided suggestive evidence that the developmental regulation of hemoglobin synthesis has evolved independently in multiple vertebrate lineages. For example, the embryonic β-like globin genes of birds and placental mammals are not 1:1 orthologs. Despite the similarity in developmental expression profiles, the genes are independently derived from lineage-specific duplications of a β-globin pro-ortholog. OBJECTIVE: To evaluate seminal plasma coenzyme Q10 (CoQ10) levels and oxidative stress in patients with different types of male infertility. METHODS: A case-control study was carried out in the Department of Chemistry and Biochemistry, College of Medicine, Al-Nahrain University, Baghdad, Iraq from the period of November 2004 to July 2006. Sixty patients with male infertility were recruited from Al-Kadhimia Teaching Hospital, Baghdad and included in this study. OBJECTIVE: To identify new tumor specific proteins of renal cell carcinoma (RCC) using proteomic analysis. METHODS: Nine renal cell carcinomas were resected and these surgical operations were carried out from June 2007 to September 2008 in the Urology Department of the Second Affiliated Hospital of Soochow University, Suzhou, Jiangsu Province, China. The cancer tissues and para-cancer normal tissues were preserved in liquid nitrogen. We analyzed the RCC tissues and para-cancer normal tissues by 2-dimensional polyacrylamide gel electrophoresis (2D-PAGE). The 16 differentially expressed protein spots (p<0.05 by Student t-test) were identified by matrix assisted laser desorption ionization/time of flight mass spectrometry and tandem mass spectrometry. RESULTS: Six proteins were down regulated and 10 proteins were up regulated in clear cell RCC compared with corresponding normal kidney tissue. The down regulated proteins were calbindin, ester hydrolase C11orf54, alcohol dehydrogenase, ammecr1-like protein, adenosine diphosphate (ADP)/adenosine-59-triphosphate (ATP) translocase 3 and leucine-tyrosine-arginine (LYR) motif-containing protein 5. The up regulated proteins were hypoxia inducible domain family member 1A, glutathione S-transferase P, thioredoxin-dependent peroxide reductase, peroxiredoxin-6, CD2-associated protein, annexin A5, gamma-enolase, retinal dehydrogenase 1, vimentin, and protein-glutamine gamma-glutamyltransferase 2. CONCLUSION: The data provide potential tumor markers for diagnosis of RCC.

OBJECTIVE: To evaluate the therapeutic outcomes of acute myeloid leukemia (AML) in elderly patients. METHODS: This study was conducted at the Aga Khan University Hospital, Karachi, Pakistan over 11 years from January 1997 to August 2008. This was a descriptive case series study. We investigated the impact of disease biology and various treatment protocols on the outcome in this population. RESULTS: A total of 55 evaluable patients (>60 years of age) were diagnosed with AML including 34 (61.8%) males and 21 (38.2%) females. A cross-sectional representative sample of the Saudi population of healthy children below 5 years of age was used to calculate the prevalence of malnutrition. With the increasing use of marijuana, efficient methods to extract and detect Δ-9-THC in urine samples and relate the concentrations found to time is a necessity so that impairment can be determined. Δ-9-THC is conjugated to glucuronic acid in urine and must be freed before the compound can be detected by chromatographic procedures. In past research, either a strong alkaline solution or β-glucuronidase has been used to cleave the bond between the Δ-9-THC and glucuronic acid before an alkaline extraction. The study of cetaceans in captivity provides information on behavior, acoustics, reproduction and physiology that is often difficult to obtain with free-ranging populations. The present study examined the influence of the estrous cycle of a captive female killer whale on the rate, duration, initiation and reception of social behavior she performed with her only pool mate, a male Atlantic bottlenose dolphin. In this study, we examine approaches to the problem of assembling large, contiguous sections of genetic code from short reads generated from laboratory techniques. We explore the Eulerian Path approach in detail, utilizing a de Bruijn Graph, and demonstrate current software technologies and algorithms using a sample genome. We investigate the input parameters of Velvet and discuss choice implications in the context of the E. coli putA/b1014 gene.

Calcium is an important second messenger for cellular communication. Theoretical models help scientists understand its signalling mechanism. A comprehensive model was developed in order to minimize any limitations in the models currently presented in the literature. Experimental results support the model and therefore the theoretical model provides a plausible explanation of the dynamics of the calcium-signaling mechanism. In the future, additional verification will be performed using various experimental configurations on PC12 cells. Further, the model will be used to predict the response of cells to environmental factors such as pesticides and heavy metals.

Purpose—Previous studies have suggested that higher levels of physical activity may lower lung cancer risk; however, few prospective studies have evaluated lung cancer mortality in relation to cardiorespiratory fitness (CRF), an objective marker of recent physical activity habits. Methods—Thirty-eight thousand men, aged 20 to 84 years without history of cancer, received a preventive medical examination at the Cooper Clinic in Dallas, TX, between 1974 and 2002. CRF was quantified as maximal treadmill exercise test duration and was grouped for analysis as low (lowest 20% of exercise duration), moderate (middle 40%), and high (upper 40%). Melanoma is known to be highly resistant to chemotherapy. Treatment with high dose IL-2 has shown significant clinical benefit in a minority of metastatic melanoma patients and has lead to long term survival in a few cases. However, this treatment is associated with excessive multiorgan toxicities, which severely limits its use. We hypothesize that one mechanism of effective IL-2 therapy is through the direct upregulation of IL-24 production in melanoma tumors and subsequent IL-24 mediated tumor growth suppression.

Although much research has been conducted on workplace stressors and their consequences, a small amount of literature exists on how women and Hispanics cope with workplace stressors or how people cope with the stressors in religious ways. To investigate the relationships among these factors, data was collected via an online survey completed by 268 university employees at a southwestern university. The purpose of this study was to investigate how adolescents in a small urban setting in the upper Midwest use alternative modes of written communication (i.e., text messaging, email, instant messaging, and social networking) for discourse purposes in order to identify ways in which these modes could be utilized by non speaking individuals. This study explored the factors related to choice of mode, including identity of the communication partner, number of communication partners, purpose of the communication, and personal preferences of the individual.

This study investigated the efficacy of written and oral peer feedback used as an instructional strategy to support student learning in speech outline development. One hundred seven students from a California Community College participated in a mixed-methods evaluative quasi-experimental study conducted in a two-fold process with both quantitative and qualitative measures. Existing literature on peer-centered learning has various gaps. These include four areas of uncertainty: implementation interventions, trust, self-perceived benefits, and contexts. Stormwater runoff is a major environmental concern, particularly in urban environments. Trends in managing stormwater have evolved (and continue to evolve) from a quantity only approach into a sustainable approach, which integrates quantity, quality, the environment, and aesthetics.
