A cell culture assembly and a method for culturing cells that provide mechanical stimulation to cells. A novel binding site for angiotensins II and III that is unmasked by parachloromercuribenzoate has been reported in rat, mouse and human brains. Initial studies of this binding site indicate that it is not expressed in the adrenal, liver or kidney of the rat and mouse. To determine if this binding site occurs in other mouse tissues, 8 tissues were assayed for expression of this binding site by radioligand binding assay and compared with the expression of this binding site in the forebrain. High-Activity Mutants Of Butyrylcholinesterase For Cocaine Hydrolysis And Method Of Generating The Same, Chang-Guo Zhan, Hoon Cho, Hsin-Hsiung Tai

A novel computational method and generation of mutant butyrylcholinesterase for cocaine hydrolysis is provided. The method includes molecular modeling a possible BChE mutant and conducting molecular dynamics simulations and hybrid quantum mechanical/molecular mechanical calculations thereby providing a screening method of possible BChE mutants by predicting which mutant will lead to a more stable transition state for a rate determining step. Site-directed mutagenesis, protein expression, and protein activity is conducted for mutants determined computationally as being good candidates for possible BChE mutants, i.e., ones predicted to have higher catalytic efficiency as compared with wild-type BChE. Mycelial morphology is a critically important process property in fermentations of lamentous micro-organisms, as particular phenotypes are associated with maximum productivity. The design of systems capable of rapidly and accurately characterising morphology within a given process represents a signi cant challenge to biotechnologists, as the complex phenotypes that are manifested are often not easily quanti ed. A system has been developed for high-resolution characterisation of lamentous fungal growth, using membrane immobilization and fully-automatic imageprocessing software. Phylogenetic analysis is becoming an increasingly important tool for biological research. Applications include epidemiological studies, drug development, and evolutionary analysis. Phylogenetic search is a known NP-Hard problem. The size of the data sets which can be analyzed is limited by the exponential growth in the number of trees that must be considered as the problem size increases. A better understanding of the problem space could lead to better methods, which in turn could lead to the feasible analysis of more data sets. Oral Histories

Roger Dean Ray came to Rollins College in 1969 to teach psychology. Over his tenure, he has authored numerous publications and engaged in many experimental projects. Eventually, Ray became the head of the Department of Psychology and the founding director of the Quantitative Learning and Teaching Center at Rollins.

Born in New Castle, Indiana, in 1941, Ray earned his B.A. in mathematics and psychology from Rollins College. He then studied at the University of Wisconsin for two years before completing his Ph.D. in general experimental psychology at the University of Tennessee in 1969.

Tobacco Mosaic Virus Based Thin Film Sensor For Detection Of Volatile Organic Compounds, Michael A. Bruckman, Jie Liu, Goutam Koley, Yu Li, Brian C. Benicewicz, Zhongwei Niu, Qian Wang

A thin film sensor for the detection of volatile organic compounds (VOC) was fabricated by deposition of oligo-aniline grafted tobacco mosaic virus (TMV) onto a glass substrate. The oligo-aniline motifs were conjugated onto the TMV surface by a traditional diazonium coupling reaction to tyrosine residues followed by Cu(I) catalyzed alkyne-azide cycloaddition (CuAAC) reaction. The modified TMV was easily fabricated into a thin film by directly drop coating onto a glass substrate. Upon integration of the glass substrate into a prototypical device, the virus-based thin film exhibited good sensitivity and selectivity toward ethanol and methanol vapour.

Transcriptomic Responses In Mouse Brain Exposed To Chronic Excess Of The Neurotransmitter Glutamate, Xinkun Wang, Xiaodong Bao, Ranu Pal, Abdulbaki Agbas, Elias K. Michaelis

Background: Increases during aging in extracellular levels of glutamate (Glu), the major excitatory neurotransmitter in the brain, may be linked to chronic neurodegenerative diseases. Little is known about the molecular responses of neurons to chronic, moderate increases in Glu levels. Genome-wide gene expression in brain hippocampus was examined in a unique transgenic (Tg) mouse model that exhibits moderate Glu hyperactivity throughout the lifespan, the neuronal Glutamate dehydrogenase (Glud1) mouse, and littermate 9 month-old wild type mice.

Results: Integrated bioinformatic analyses on transcriptomic data were used to identify bio-functions, pathways and gene networks underlying neuronal responses to increased Glu synaptic release. …

Patients seeking help for hypersexual behavior often exhibit features of impulsivity, cognitive rigidity, poor judgment, deficits in emotion regulation, and excessive preoccupation with sex. Some of these characteristics are also common among patients presenting with neurological pathology associated with executive dysfunction. These observations led to the current investigation in which differences across scores on objective neuropsychological tests of executive functioning were explored in a group of hypersexual male patients (n = 30) compared with a non-hypersexual community sample (n = 30) of men. Using multivariate statistics, differences between the groups were examined yielding significant differences on measures of hypersexuality. However, …

Project I Celogentin C, isolated by Kobayashi from the seeds of Celosia argentea in 2001 exhibits strong inhibitory behavior towards polymerization of tubulin – globular protein, which plays crucial role during the cell division cycle. We have attempted synthesis of the left-hand ring of celogentin C via the intramolecular alkylation and the Knoevenagel condensation strategies. Utilizing synthetic methodologies in the field of nitroacetyl chemistry, developed by Kornblum and Rajappa we have successfully prepared the intermolecular Knoevenagel condensation product – the late-stage precursor to the left-hand ring of celogentin C. A clade 1 sequence of H5 haemaglutinin from an Asian Avian H5N1 isolate was used as a the template to chemically synthetize a codon optimized version for expression in insect cells. A single clone was chosen for expression optimization and changes were introduced in order to maximize the amount of protein to be produced and to resemble another sequence demonstrated to be present in an isolate causing disease in Humans.

Here we report the utilization of Drosophila melanogaster Schneider 2 (S2) cells and the Semliki Forest Virus (SFV) expression system to produce the rabies virus glycoprotein (RVGP). Although they represent different expression systems, they offer similar advantages in generating high-level expression of functional membrane proteins. They are both relatively easy and safe to handle and are scalable. RVGP synthesized by S2 cells and SFV carrying an mRNA coding for RVGP (SFV-RVGP) were assayed in mouse protection studies. Centrifugation is a unit operation used within a wide range of bioprocesses, including the production of a whole cell cancer vaccine to treat Hormone Refractory Prostate Cancer (HRPC) which has been tested in Phase II clinical trials. To quantify the effect of centrifugation-related stresses upon a whole cell vaccine population, a design of experiments (DOE) based investigation at micro-scale was implemented. Qualification of the cells may be by membrane integrity and by surface marker density as well as cytokines released and retained during bioprocessing.
