Desain penelitian adalah before and after, dengan metoda pengumpulan data …
Informing Social Work Practice Through The Enhancement Of The Biological Perspective: A Course Intervention Model For Human Service Professionals Working With Youth And Problems Of Conduct., Allison Sampson
Theses and Dissertations
The purpose of this study is to evaluate the effectiveness of an intervention model designed to enhance practitioners’ biological lens when using a biopsychosocial-spiritual model of holistic assessment and planning. The specific intervention utilized is a course curriculum developed to broaden human service professionals’ (including clinical social work professionals) understanding of attachment theory, neuroscience and trauma informed methods of practice. The course teaches professionals how to apply this knowledge to clinical assessment and intervention planning with youth who have experienced significant trauma in their lives and exhibit problems of conduct. Using an experimental design, participants from a large private mental …

Urban Adolescents’ Cognitive Responses To Peer Victimization: Does Psychosocial Adjustment Play A Role?, Amie Bettencourt
Urban Adolescents’ Cognitive Responses To Peer Victimization: Does Psychosocial Adjustment Play A Role?, Amie Bettencourt
Theses and Dissertations
Peer victimization is characterized by acts of physical, relational, and verbal aggression that can contribute to maladjustment. Youths’ responses to peer victimization are guided by social information-processing (SIP) skills that impact their adjustment. Maladjustment can contribute to biases in SIP. Biased processing occurs when youth rely on existing schemas without attending to cues from the immediate social context. These processing deficits contribute to the enactment of problematic responses that may lead to further maladjustment. However, not all youth exhibit SIP deficits. A recent study identified four adjustment clusters based on differences in aggression, anxiety, depression, social acceptance, and victimization within …

Novel Polymer Coupling Chemistry Based Upon Latent Cysteine-Like Residues And Thiazolidine Chemistry, Jospeh S. Carlson, Megan R. Hill, Taiga Young, Philip Costanzo
Chemistry and Biochemistry
Chain end functional polymers were prepared via reversible addition–fragmentation transfer (RAFT) polymerization techniques that were further chain extended with acrylonitrile. Under reducing conditions, latent cysteine-like residues were exposed at the chain ends. A variety of reduction conditions were explored and base polymers were then tethered together via thiazolidine chemistry.

Pharmacological Characterization Of Gabaa Receptors In Taurine-Fed Mice, William J. L’Amoreaux, Alexandra Marsillo, Abdeslem El Idrissi
Pharmacological Characterization Of Gabaa Receptors In Taurine-Fed Mice, William J. L’Amoreaux, Alexandra Marsillo, Abdeslem El Idrissi
Publications and Research
Background Taurine is one of the most abundant free amino acids especially in excitable tissues, with wide physiological actions. Chronic supplementation of taurine in drinking water to mice increases brain excitability mainly through alterations in the inhibitory GABAergic system. These changes include elevated expression level of glutamic acid decarboxylase (GAD) and increased levels of GABA. Additionally we reported that GABAA receptors were down regulated with chronic administration of taurine. Here, we investigated pharmacologically the functional significance of decreased / or change in subunit composition of the GABAA receptors by determining the threshold for picrotoxin-induced seizures. Picrotoxin, an antagonist of GABAA …

Taurine Regulation Of Short Term Synaptic Plasticity In Fragile X Mice, Abdeslem El Idrissi, Lorenz S. Neuwirth, William L’Amoreaux
Publications and Research
Background Fragile X Syndrome is the most common known genetic cause of autism. The Fmr1-KO mouse, lacks the fragile X mental retardation protein (FMRP), and is used as a model of the syndrome. The core behavioral deficits of autism may be conceptualized either as excessive adherence to patterns as seen in repetitive actions and aberrant language, or as insensitivity to subtle but socially important changes in patterns. The hippocampus receives information from the entorhinal cortex and plays a crucial role in the processing of patterned information. To gain more insight into the physiological function of FMRP and the neuronal mechanisms …

Neuro-Endocrine Basis For Altered Plasma Glucose Homeostasis In The Fragile X Mouse, Abdeslem El Idrissi, Xin Yan, Francoise Sidime, William L’Amoreaux
Publications and Research
Background The fragile X mouse model shows an increase in seizure susceptibility, indicating an involvement of the GABAergic system via an alteration in cellular excitability. In the brain, we have previously described a reduction in GABAA receptor expression as a likely basis for this susceptibility. In the brains of fragile X mice, this reduction in receptor expression culminates with a concomitant increase in the expression of glutamic acid decarboxylase (GAD), the enzyme responsible for GABA synthesis. Further, voltage-sensitive calcium channel expression is reduced in the pancreas of the fragile X mouse. Since there are considerable similarities in the GABAergic system …
Taurine Regulates Insulin Release From Pancreatic Beta Cell Lines, William J. L’Amoreaux, Christina Cuttitta, Allison Santora, Jonathan F. Blaize, Janto Tachjadi, Abdeslem El Idrissi
Taurine Regulates Insulin Release From Pancreatic Beta Cell Lines, William J. L’Amoreaux, Christina Cuttitta, Allison Santora, Jonathan F. Blaize, Janto Tachjadi, Abdeslem El Idrissi
Publications and Research
Background Pancreatic β-cells release insulin via an electrogenic response triggered by an increase in plasma glucose concentrations. The critical plasma glucose concentration has been determined to be ~3 mM, at which time both insulin and GABA are released from pancreatic β-cells. Taurine, a β-sulfonic acid, may be transported into cells to balance osmotic pressure. The taurine transporter (TauT) has been described in pancreatic tissue, but the function of taurine in insulin release has not been established. Uptake of taurine by pancreatic β-cells may alter membrane potential and have an effect on ion currents. If taurine uptake does alter β-cell current, …

Virus Silicification Under Simulated Hot Spring Conditions, James R. Laidler, Kenneth M. Stedman
Virus Silicification Under Simulated Hot Spring Conditions, James R. Laidler, Kenneth M. Stedman
Biology Faculty Publications and Presentations
Silicification of organisms in silica-depositing environments can impact both their ecology and their presence in the fossil record. Although microbes have been silicified under laboratory and environmental conditions, viruses have not. Bacteriophage T4 was successfully silicified under laboratory conditions that closely simulated those found in silica-depositing hot springs. Virus morphology was maintained, and a clear elemental signature of phosphorus was detected by energy-dispersive X-ray spectrophotometry (EDS).
Microarray Analysis Of Gene Expression Profiles Of Tetrahymena H2b Ubiquitylation Mutant Strains, Zhe Wang
Microarray Analysis Of Gene Expression Profiles Of Tetrahymena H2b Ubiquitylation Mutant Strains, Zhe Wang
Theses
Microarray analyses were performed to compare the gene expression profiles of wild-type and mutant strains of Tetrahymena thermophila. These studies suggest that, in Tetrahymena, ubiquitylation of histone H2B is not required for H3K4 methylation. Even though the E2 ubiquitin-conjugating enzyme Ubc2 and the E3 ubiquitin ligase Bre1 are required for H2B ubiquitylation, these two enzymes affect the expression of a much broader spectrum of genes than H2B ubiquitylation, suggesting that Ubc2 and Bre1 are involved in pathways other than H2B ubiquitylation in Tetrahymena.

Plasticity Of Lipid-Protein Interactions In The Function And Topogenesis Of The Membrane Protein Lactose Permease From Escherichia Coli, Mikhail Bogdanov, Philip Heacock, Ziqiang Guan, William Dowhan
Faculty, Staff and Student Publications
Phosphatidylcholine (PC) has been widely used in place of naturally occurring phosphatidylethanolamine (PE) in reconstitution of bacterial membrane proteins. However, PC does not support native structure or function for several reconstituted transport proteins. Lactose permease (LacY) of Escherichia coli, when reconstituted in E. coli phospholipids, exhibits energy-dependent uphill and energy-independent downhill transport function and proper conformation of periplasmic domain P7, which is tightly linked to uphill transport function. LacY expressed in cells lacking PE and containing only anionic phospholipids exhibits only downhill transport and lacks native P7 conformation. Reconstitution of LacY in the presence of E. coli-derived PE, but not …

Alternative Processing Of Scrambled Genes Generates Protein Diversity In The Ciliate Chilodonella Uncinata, Laura A. Katz, Alexandra M. Kovner
Alternative Processing Of Scrambled Genes Generates Protein Diversity In The Ciliate Chilodonella Uncinata, Laura A. Katz, Alexandra M. Kovner
Biological Sciences: Faculty Publications
In ciliates, chromosomal rearrangements occur during the development of the somatic macronuclear genome from the germline micronuclear genome. These rearrangements are extensive in three ciliate classes-Armophorea, Spirotrichea, and Phyllopharyngea-generating a macronucleus with up to 20,000,000 gene-sized chromosomes. Earlier, we have shown that these three classes also share elevated rates of protein evolution relative to other ciliates. To assess the evolution of germline-limited sequences in the class Phyllopharyngea, we used a combination of traditional and walking PCR to analyze micronuclear copies of multiple genes from two lines of the morphospecies Chilodonella uncinata for which we had previously characterized macronuclear sequences. Analyses …
