This dissertation includes a comprehensive current review of reversible electroporation (EP) and other related physical gene transfection techniques; an overview of results of electrochemogene therapy (ECGT) used to treat naturally occurring spontaneous neoplasms in dogs; and the results of comprehensive, pre-clinical toxicology testing of electrogene therapy (EGT) of a tumor-targeted version of interleukin-12 (IL-12) in mice. Intralesional bleomycin (BLM) and feline interleukin-12 (fIL-12) DNA injection combined with trans-lesional EP resulted in complete cure of two recurrent oral squamous cell carcinomas and an acanthomatous ameloblastoma in a series of six cases of spontaneous neoplasia in pet dogs. Nanoparticle (NP) interactions that may destabilize cell membranes and cause toxicity can also be used to design and control therapeutic assemblies. Fundamentally examining interactions between NPs and cell membranes is crucial to understanding NP toxicity mechanisms and the development of safe and non-toxic NP-based commercial and therapeutic applications. In this dissertation it is demonstrated that self-assembled lipid bilayers can be employed as model membranes to determine NP interact ions. First, protocols were developed for examining hydrophobic and hydrophilic NP-bilayer interactions using lipid bilayer vesicles or liposomes. Prematurity is a national problem with rates climbing to the highest levels in four decades. Social competence is a critically important skill for children to master in order to become socially competent adults. Social competence is not fully studied in children born prematurely. Yet they are at risk for poorer social competence due to their physical, cognitive, motor and developmental difficulties. It is inherently difficult to study the direct interactions between marine microbes due to the size of the organisms and microscale environments in which they exist. Recent evidence suggests that the direct associations between marine phytoplankton and particle-colonizing bacteria can influence marine microbial community succession, microalgal primary production, toxin production by marine phytoplankton, and have potential implications for human health. Tumor initiation and progression are dependent on both aberrant gene expression in tumor cells and the communication between tumor cells and its micro- and systemic microenvironment. Many tumor suppressor genes and oncogenes have been characterized to suppress or promote tumor growth, but fewer genes in tumors are well-characterized as interacting with immune cells in the host to promote or inhibit tumor growth. The interleukin (IL) 27 receptor WSX1 is expressed in immune cells and induces an IL27-dependent immune response. Response blocking and response interruption are common interventions for problem behavior maintained by automatic reinforcement in the treatment literature, but these interventions may be extremely challenging for caregivers to implement with fidelity (i.e., immediately blocking each instance). The differentiation potential of adult stem cells (ASC) has long been thought to be limited to cell lineages present in the organ from which they are derived; however, several studies have challenged this notion by demonstrating that some ASC exhibit a remarkably high degree of plasticity. Unlike terminally differentiated somatic cells, the less differentiated state of ASC can assume the functional phenotypes and expression profiles of cells unique to other tissues. The expansive repertoire of differentiation potential exhibited by ASC suggests these cells possess characteristics similar to pluripotent cells, including epigenomic regulatory pattern. CD4+ memory T cells are depleted in mucosal tissues post human immunodeficiency virus (HIV) and simian immunodeficiency virus (SIV) infection without restoration to pre-infection levels during progressive course of disease. Bone marrow (BM) as a hematopoietic organ has been investigated for hematologic and morphologic changes during HIV infection. However, BM as a primary lymphoid tissue during HIV infection has been poorly characterized. Brucellosis, a zoonotic disease caused by Brucella spp., presents both health and economic difficulties for livestock, wildlife, and humans. While brucellosis is nearly eradicated in the United States, the disease remains detrimental in many countries worldwide. Attempts to produce a safe and effective small ruminant vaccine have been met with limited success. The current vaccine for bovine brucellosis in the United States is B. abortus RB51. Mutualisms are reciprocally exploitative interactions providing net benefits to both partners. These interactions can be exploited, in turn, by individuals that take advantage of benefits offered by one or both partners in a mutualism, while offering no benefits in return. For many mutualist-exploiter interactions the mechanisms allowing exploitation, and the maintenance of mutualisms in the face of exploitation, are still poorly understood. Here I describe manipulative field and laboratory experiments to investigate the mechanisms used by an exploiter to invade an ant-plant mutualism. Dissolution of chitosan in ionic liquids was accomplished. It has been shown that 1-butyl-3-methylimidazolium acetate (BMIMAc) is a better solvent than 1-butyl-3-methylimidazolium chloride (BMIMCl). Dissolution of chitosan in BMIMCl required a prior regeneration of chitosan from 1% acetic acid solution. In the case of BMIMAc, both dried chitosan and regenerated chitosan from acetic acid solution have been dissolved in a relatively short amount of time. While concentrations up to 2 wt% of chitosan in BMIMCl could be obtained, concentrations of 10 wt % of chitosan in BMIMAc were realized. To control gene expression in vivo with spatial and temporal precision remains a significant hurdle in laboratory studies of development as well as clinical genetic therapies. Here we demonstrate such control over gene expression by use of photochemistry to reversibly inactivate the hybridization of a nucleic acid analog used for specific protein knockdown. A morpholino oligonucleotide, commonly used for knockdown of protein expression in developmental studies, was “caged” using carbodiimide conjugate chemistry which yielded photocleavable adducts that can be removed with light exposure. Obtaining real-time and accurate estimates of energy intake while people reside in their natural environment is technically and methodologically challenging. The goal of this project is to estimate energy intake accurately in real-time and free-living conditions. In this study, we propose a computer vision based system to estimate energy intake based on food pictures taken and emailed by subjects participating in the experiment. The system introduces a reference card inclusion procedure, which is used for geometric and photometric corrections. Image classification and segmentation methods are also incorporated into the system to have fully-automated decision making.

The purpose of this phenomenological study was to examine the role of race in cross-race advising relationships between White faculty advisors and their Black doctoral student protégés. I examined the racial context of doctoral education experiences and relationships between doctoral students and faculty. Blacks with doctoral degrees comprise only 0.3% of those 18 years and older and only 3.5% of those with doctoral degrees (U.S. Census, 2000). Although there has been an increase in the number of Blacks enrolling in doctoral programs (Cook & Cordova, 2006), Nettles and Millett (2006) found that Blacks and Latin Americans have higher attrition rates …

Eutrophication and coastal wetland loss are the major environmental problems affecting estuaries around the world. In Louisiana, controlled diversions of the Mississippi River water back into coastal wetlands are thought to be an important engineering solution that could reverse coastal land loss. There are concerns, however, that freshwater diversions may increase nutrient inputs and create severe eutrophication problems in estuaries and wetlands adjacent to the diversion sites. Colonial nesting seabirds are threatened by habitat loss and degradation, human disturbance, predation, and climate change. Several species of conservation concern concentrate high percentages of their total U.S. populations in Louisiana breeding colonies. We studied seabirds, including Royal Terns, Sandwich Terns, and Black Skimmers, nesting on Isles Dernieres barrier islands along the Gulf coast of Louisiana. Two of the four islands in this chain host extensive seabird colonies and two do not. A confluence of factors determines the biological diversity we observe today. Here, I conducted three investigations of the historic, geographic and ecological factors that shaped the morphological and genetic diversity of rainforest birds in Southeast Asia. (1) In the Oriental Dwarf Kingfisher (Ceyx erithaca) complex, the birds are highly polymorphic in plumage. Quantitative analysis of this variation indicated that a large proportion of birds in Sumatra, the Malay Peninsula and Borneo have plumages intermediate between the northerly black form from mainland Asia and the southerly rufous form from Java. Recent food recalls and food scares in the United States have increased consumers’ risk perceptions about food borne illness and decreased their confidence in the safety of the U.S. food supply. Results from a continuous tracking of consumer confidence and media coverage of food safety events over a 67 week period between May 2008 and August 2009 are reported in the study. However, several observable trends have recently made investing in the US agricultural sector an area of interest among investors. Stock price indexes are a commonly used tool for summarizing the historical performance of a specific sector. Twelve neonatal Holstein bull calves (38.52 ± 5.87 kg) were fed milk replacer at a fixed or varied meal time to determine the effects on metabolic hormone secretion, average daily intake, growth, and energy metabolism. Body weights were measured every two weeks from birth to 8 weeks. Rumen fluid was collected every two weeks from week 2 through 8. Serial blood collections were conducted every two weeks from week 2 through 8. Blood was collected, beginning at 0530, at 0, 15, 30, 45, 60, 75, 90, 105, 120, 135, and 150 minutes.
